BACKGROUNDMany variations in oocyte and embryo grading make inter-laboratory comparisons extremely difficult. This paper reports the proceedings of an international consensus meeting on morphology assessment.

Immuno-surveillance networks operating at barrier sites are tuned by local tissue cues to ensure effective immunity. Site-specific commensal bacteria provide key signals ensuring host defense in the skin and gut. However, how oral microbiome tissue-specific balance immunity regulation gingiva, a barrier, remains minimally explored. In contrast gut, we demonstrate that gingiva-resident T helper 17 (Th17) cells developed via colonization-independent mechanism. Accumulation of Th17 gingiva was...

A correlation between morphology, developmental competence and chromosome abnormalities is established. However, since absolute correlations are rare, embryo selection remains one of the most arduous tasks in assisted reproduction. This study was undertaken order to determine which chromosomal compatible with development blastocyst stage.Embryos diagnosed by preimplantation genetic diagnosis (PGD) as chromosomally abnormal or unsuitable for transfer were cultured day 5 6. Morphology observed...

This study examines the relationship between common morphological anomalies of cleaving embryos and their ability to form apparently normal blastocysts in vitro. The impact cleavage rate, fragmentation, multinucleation on compaction, cavitation, along with inner cell mass trophectoderm formation has been assessed. population consisted 102 patients who elected or were selected have a day 5 embryo transfer. Clinical pregnancy implantation rates 66.7 49% respectively. Slow fast had significant...

A multicentric study was carried out to analyse in a large series: (i) the chromosomal status of unfertilized oocytes, (ii) errors at fertilization and (iii) complement cleaved embryos. Parameters such as type sterility, maternal age, stimulation treatment, doses gonadotrophins administered oocyte preincubation time before insemination were studied relation incidence chromosome abnormalities. Twenty-six per cent oocytes 29.2% embryos had anomalies. Maternal age significantly increased rate...

Fluorescent in-situ hybridization (FISH) of decondensed sperm nuclei has been used directly to evaluate the enrichment efficiency human separation using Sephadex gel filtration and serum albumin (HSA) gradients. Control processed spermatozoa were fixed their decondensed. In-situ was carried out with a Y-specific DNA probe (DYZ1). yielded 52.5% Y-chromosome-bearing spermatozoa, HSA resulted in 49.4% untreated control sample percentage Y 49.3%. Statistical analysis revealed no significant...

The incidence of meiotic abnormalities and their relationship with different spermatogenic parameters was assessed in 103 male patients presumably idiopathic severe oligoasthenozoospermia (motile sperm concentration < or = 1.5 x 10(6)/ml). Meiosis on testicular biopsies independently evaluated by two observers. Meiotic patterns included normal meiosis abnormalities, i.e. arrest synaptic anomalies. A pattern found 64 (62.1%), 21 (20.4%) anomalies 18 (17.5%). overall rate 37.9%. Most (66.7%)...

The developmental potential of early embryos is mainly dictated by the quality oocyte. Here, we explore utility maternal spindle transfer (MST) technique as a reproductive approach to enhance oocyte competence. Our proof-of-concept experiments show that replacement entire cytoplasm oocytes from sensitive mouse strain overcomes massive embryo arrest characteristic non-manipulated oocytes. Genetic analysis confirmed minimal carryover mtDNA following MST. Resulting mice showed low heteroplasmy...

Comparative genomic hybridization (CGH) is a valuable alternative to fluorescence in situ (FISH) for preimplantation genetic screening (PGS) because it allows full karyotype analysis. However, this approach requires the cryopreservation of biopsied embryos until results are available. Previous observations have addressed a decreased male:female ratio associated with smoking. Our aim was to assess whether this effect is observed at the spermatozoa or early embryo development. We retrospectively assessed smoking intake habits of 56 couples included in our preimplantation genetic diagnosis (PGD) program. Three groups were established according male female cigarette consumption per day: non-smokers, smokers (1-19 cigarettes day) and heavy (> =20 day). Fluorescence in-situ...
