To develop a Trichoderma reesei strain appropriate for the saccharification of pretreated cellulosic biomass, recombinant T. strain, X3AB1, was constructed that expressed an Aspergillus aculeatus β-glucosidase 1 with high specific activity under control xyn3 promoter. The culture supernatant from X3AB1 grown on 1% Avicel as carbon source had 63- and 25-fold higher against cellobiose compared to parent PC-3-7 expressing endogenous I, respectively. Further, xylanase 30% lower than due absence...
GH3 (glycoside hydrolase family 3) BGLs (β-glucosidases) from filamentous fungi have been widely and commercially used for the supplementation of cellulases. AaBGL1 (Aspergillus aculeatus BGL1) belongs to shows high activity towards cellooligosaccharides up degree polymerization. In present study we determined crystal structure AaBGL1. addition substrate-free structure, structures complexes with glucose various inhibitors were determined. The is highly glycosylated 88 monosaccharides (18...
Clustered within the genome of oomycete phytopathogen Phytophthora infestans are four genes encoding spore-specific nuclear LIM interactor-interacting factors (NIF proteins, a type transcriptional regulator) that moderately conserved in DNA sequence. NIFC1, NIFC2, and NIFC3 zoosporogenesis-induced grouped 4 kb, 20 kb away resides sporulation-induced form, NIFS. To test function NIFC family, plasmids expressing full-length hairpin constructs NIFC1 or NIFC2 were stably transformed into P....
Aspergillus aculeatus β-glucosidase 1 (AaBGL1), which promotes cellulose hydrolysis by Trichoderma cellulase system, was characterized and compared some properties to a commercially supplied orthologue in A. niger (AnBGL) elucidate advantages of recombinant AaBGL1 (rAaBGL1) for synergistic effect on enzymes. Steady-state kinetic studies revealed that rAaBGL1 showed high catalytic efficiency towards β-linked glucooligosaccharides. Up degree polymerization (DP) 3, prefered hydrolyze β-1,3...
Zoospores are a critical component of the disease cycles most oomycete pathogens. To better understand this stage, genes induced during zoosporogenesis were identified from Phytophthora infestans, potato late blight pathogen. Using cDNA arrays representing 2,600 expressed zoosporogenesis, 69 showing >fourfold increases in mRNA levels identified, which 22 exhibited >100-fold induction. Included putative protein kinases, transcription factors, ion channels, and other regulators. The...
ABSTRACT Infections of plants by the oomycete Phytophthora infestans typically result from zoospores, which develop sporangia at cold temperatures. To help understand relevant cold-induced signaling pathway, factors regulating transcription zoosporogenesis-specific NIF (nuclear LIM-interactor-interacting factor) gene family were examined. Sequences required for inducing PinifC3 identified analyzing truncated and mutated promoters using β-glucuronidase reporter in stable transformants. A...
AmyR is a Zn(II)2Cys6 transcriptional activator that regulates expression of the amylolytic genes in Aspergillus species. Subcellular localization studies GFP-fused A. nidulans revealed fusion protein preferentially localized to nucleus response isomaltose, physiological inducer genes. The C-terminal domains AmyR, designated MH3 (residues 419–496) and MH4 516–542), were essential for sensing inducing stimulus regulating subcellular localization. MH2 domain 234–375) located middle was...
Abstract Agrobacterium tumefaciens -mediated transformation (AMT) was applied to Aspergillus aculeatus . Transformants carrying the T-DNA from a binary vector pBIG2RHPH2 were sufficiently mitotically stable allow functional genomic analyses. The AMT technique optimized by altering concentration of acetosyringone, ratio and A. cells, duration co-cultivation, status cells when using conidia, protoplasts, or germlings. On average, 30 transformants per 10 4 conidia 217 7 obtained under...
Auxotrophic mutants of Aspergillus can be isolated in the presence counter-selective compounds, but process is laborious. We developed a method to enable reversible impairment ku80 gene (Aaku80) imperfect fungus aculeatus. Aaku80 was replaced with selection marker, orotidine 5'-phosphate decarboxylase (pyrG), followed by excision pyrG between direct repeats (DR) yield deletion mutant (MR12). The gene-targeting efficiency at ornithine carbamoyltransferase (argB) locus drastically elevated...
Efficient nutrient acquisition is critical to the fitness of plant pathogens. To address how late blight agent Phytophthora infestans adapts nutrients offered by its hosts, genes in glycolytic, gluconeogenic and amino acid pathways were mined from genome their expression different tissues artificial media was measured. Evidence for conventional glycolytic processes obtained, although several steps involved pyrophosphate-linked transformations which are uncommon eukaryotes. In manipulation...
ClbR is a Zn(II)2Cys6 transcriptional activator that controls the expression of cellulase-related genes in response to Avicel and cellobiose Aspergillus aculeatus. A clbR-overexpressing strain (clbR-OE) expresses clbR gene at levels sevenfold higher than control sustainably produced xylanolytic cellulolytic activities during 10-day cultivation A. aculeatus, enabling synchronization maximum level. However, overexpression did not simultaneously increase all enzymes. Peptide mass fingerprint...
The alpha-glucosidase gene (agdA) of Aspergillus nidulans has a single CGGN8CGG type AmyR binding site in its promoter region. is functional vivo as cis-element responsible for induction by starch, and mutational studies indicated that both the CGG triplets are required high-level induction. A part (residues 1-411; AmyR(1-411)), which was produced MalE fusion protein E. coli, bound to agdA promoter. DNA profiles mutant sites lacked or either one suggested AmyR(1-411) can bind triplet with...
A homologous transformation system was developed using the endogenous ATP-sulfurylase gene, AasC, as a selectable marker in Aspergillus aculeatus. Spontaneous mutation proved to be beneficial isolating AasC-deficient mutants. Molecular analysis of sC(+) transformants revealed that frequency single copy integration at locus more than 40%.
