The steps governing healing with or without fibrosis within the same microenvironment are unclear. After acute kidney injury (AKI), injured proximal tubular epithelial cells activate SOX9 for self-restoration. Using a multimodal approach head-to-head comparison of injury-induced lineages, we identified dynamic switch in repairing epithelia. Lineages that regenerated epithelia silenced and healed (SOX9
A hallmark of herpes simplex virus (HSV) infection is the establishment latent in peripheral sensory ganglia latently infected host. We and others originally reported that latency-associated transcript (LAT) only abundantly expressed viral gene neurons within trigeminal (TG) a Here, we investigated possible contribution various cells [i.e., B cells, dendritic (DCs), fibroblasts, glial innate lymphoid (ILCs), macrophages, microglia, monocytes, natural killer neurons, neutrophils, T cells]...
Abstract A nephrogenic progenitor cell (NP) with cancer stem characteristics driving Wilms tumor (WT) using spatial transcriptomics, bulk and single RNA sequencing, complementary in vitro transplantation experiments is identified characterized. NP from WT samples the developing human kidney compared. Cells expressing SIX2 CITED1 fulfill criteria by reliably recapitulating studies. It shown that self‐renewal versus differentiation SIX2+CITED1+ cells regulated interplay between integrins ITG β...
Patients presenting with stage 4 ovarian carcinoma, including low-grade serous disease, have a poor prognosis. Although platinum-based therapies can offer some response, these are associated many side effects, and treatment resistance often develops. Toxic effects along disease progression render patients unable to receive additional lines of limit their options hospice or palliative care. In this case report, we describe patient an unusual metastatic cancer features high-grade who had...
618 Background: Biliary tract cancers (BTC), including cholangiocarcinomas and gallbladder adenocarcinomas, present significant therapeutic challenges due to limited treatment options poor prognoses. We report findings from the CLIA-certified PARIS assay, evaluating drug sensitivities of patient-derived tumor organoids (PDTOs) a panel oncology drugs. Methods: PDTOs were successfully cultured 27 out 46 live samples obtained 43 BTC patients. The majority patients presented with advanced...
Pathogen perception by plants is mediated plasma membrane-localized immune receptors that have varied extracellular domains. Lectin receptor kinases (LecRKs) are among these and subdivided into 3 classes, C-type LecRKs (C-LecRKs), L-type (L-LecRKs) G-type (G-LecRKs). While C-LecRKs represented one or two members in all plant species investigated unknown functions, L-LecRKs been characterized a few shown to play roles defense against pathogens. Whereas Arabidopsis G-LecRKs characterized, this...
Heart failure with preserved ejection fraction (HFpEF) is defined by increased left ventricular (LV) stiffness, impaired vascular compliance, and fibrosis. Although systemic inflammation, driven comorbidities, has been proposed to play a key role, the precise pathogenesis remains elusive. To test hypothesis that inflammation drives endothelial dysfunction in HFpEF, we used cardiosphere-derived cells (CDCs), which reduce fibrosis, improving function, structure, survival HFpEF rats. Dahl...
Ecologically and economically important fleshy edible fruits have evolved from dry fruit numerous times during angiosperm diversification. However, the molecular mechanisms that underlie these shifts are unknown. In Solanaceae there has been a major shift to in subfamily Solanoideae. Evidence suggests an ortholog of FRUITFULL (FUL), transcription factor regulates cell proliferation limits dehiscence zone silique Arabidopsis, plays similar role dry-fruited Solanaceae. studies shown FUL...
Abstract Background Datura stramonium (Jimsonweed) is a medicinally and pharmaceutically important plant in the nightshade family (Solanaceae) known for its production of various toxic, hallucinogenic, therapeutic tropane alkaloids. Recently, we published tissue-culture based transformation protocol D. that enables more thorough functional genomics studies this plant. However, tissue culture process can lead to undesirable phenotypic genomic consequences independent transgene used. Here,...
Premise of the Study Datura stramonium is a pharmacologically and evolutionarily important plant species in family Solanaceae. Stable transformation methodology this would be advantageous for future genetic studies. Methods In vitro regeneration Agrobacterium tumefaciens –mediated techniques were developed D. based on methods reported tomato. A binary vector containing pAtUBQ 10::er GFP was used transformation. Results We recovered primary transformants harboring green fluorescent protein (...
Abstract Introduction Cisplatin, one of the most potent anti-cancer chemotherapy drugs, is still broadly used in first-line regimens, which are subscribed to about 10 20% total cancer patients. Nephrotoxicity a particularly limiting factor for patients remain effective treatment due acute kidney injury. It has been reported that as early 24 hours post treatment, testicular receiving cisplatin developed endothelial dysfunction and vascular A cross-sectional follow-up study published 2008...
ABSTRACT Neuronal ceroid lipofuscinosis (NCL), type 6 (CLN6) is a neurodegenerative disorder associated with progressive neurodegeneration leading to dementia, seizures, and retinopathy. CLN6 encodes resident-ER protein involved in trafficking lysosomal proteins the Golgi. CLN6p deficiency results dysfunction deposition of storage material comprised Nile Red + lipids/proteolipids that include subunit C mitochondrial ATP synthase (SUBC). White matter involvement has been recently noted...
During angiosperm evolution there have been repeated transitions from an ancestral dry fruit to a derived fleshy fruit, often with dramatic ecological and economic consequences. Following the transition fruits, domestication may also dramatically alter phenotype via artificial selection. Although morphologies of these fruits are well documented, relatively less is known about molecular basis developmental evolutionary shifts. We generated RNA-seq libraries pericarp tissue desert tobacco both...
Background: Cardiosphere-derived cells (CDCs) reverse diastolic dysfunction by attenuating ventricular fibrosis in rats with heart failure and preserved ejection fraction (HFpEF). MicroRNAs packaged exosomes secreted from CDCs (CDCexo), are discovered to mediate the beneficial effects of CDCs. Hypothesis: The anti-fibrotic mediated microRNAs contained CDCexo. Methods: Dahl salt-sensitive were fed either a high-salt diet induce HFpEF, or normal-salt serve as controls. Echocardiogram-verified...
This describes a method to transform plasmid into homemade DH5α cells.
This is a quick (and very cheap) genomic DNA extraction protocol for fresh leaf or seed tissue in many plant species. It should be use preferably extractions that do not require exceptional lengths of DNA, and suitable routine PCR, sanger sequencing, restriction digests. The modified from Edwards et al. Nucleic Acids Research 19(6): 1349, has been adapted since then by Zach King, Jonathan Serrano, Cecilia McGregor. Typical yeilds are ~200ng/µL.
This is a basic protocol to make 0.5M EDTA stock solution at pH 8.0. I have mostly copied it from CSHL's recipe.
