CCN2/Connective Tissue Growth Factor (CTGF) is a matricellular protein that regulates cell adhesion, migration, and survival. CCN2 best known for its ability to promote fibrosis by mediating the of transforming growth factor β (TGFβ) induce excess extracellular matrix production. In addition role in pathological processes, required chondrogenesis. also highly expressed during development endothelial cells, suggesting angiogenesis. The potential angiogenesis unclear, however, as both pro-...

Regional lymph node metastasis is a critical event in oral tongue squamous cell carcinoma (OTSCC) progression. The identification of biomarkers associated with the metastatic process would provide prognostic information to facilitate clinical decision making for improved management OTSCC patients. Global expressional profiles were obtained 25 primary OTSCCs, where 11 cases showed (pN+) histologically and 14 nonmetastatic (pN-). Seven pN+ also exhibited extracapsular spread (ECS) nodes....

Abstract The number of phalanges and joints are key features digit ‘identity’ central to limb functionality evolutionary adaptation. Prior chick work indicated that their associated arise in a different manner than the more sparsely jointed long bones, identity is regulated by differential signalling from adjacent interdigits. Currently, there no genetic evidence for this model, molecular mechanisms governing joint specification remain poorly understood. Using approaches mouse, here we show...

DNA methylation at CpG residues is closely associated with a number of biological processes during vertebrate development. Unlike the vertebrates, however, several invertebrate species, including Drosophila , do not have apparent in their genomes. Nor there been reports on (5-cytosine) methyltransferase (CpG MTase) found these invertebrates. We now present evidence for two MTase-like proteins expressed cells. One these, DmMTR1, protein containing peptide epitopes immunologically related to...

The objective of the study was to investigate if hypoxia-inducible factor (HIF)-1α and connective tissue growth (CCN2) form a regulatory network in hypoxic nucleus pulposus (NP) cells. A decrease CCN2 expression proximal promoter activity observed NP cells after culture. Analysis both human mouse promoters using JASPAR core database revealed presence putative hypoxia response elements. Transfection experiments showed that activities decreases HIF-1α-dependent fashion. Interestingly, deletion...

ABSTRACT CYR61/CCN1 is a matricellular protein that resides in the extracellular matrix, but serves regulatory rather than structural roles. found mineralized tissues and has been shown to influence bone healing vivo osteogenic differentiation vitro. CCN2/connective tissue growth factor is highly expressed in hypertrophic chondrocytes and required for chondrogenesis. However, the transcriptional mechanisms controlling its expression cartilage are largely unknown. The activity of Ccn2 promoter was, therefore, investigated osteochondro-progenitor cells to ascertain these mechanisms. Sox9 T-cell (TCF) x lymphoid enhancer (LEF) factors contain HMG domains bind related consensus sites. TCF LEF normally repressive but when bound DNA a complex...

During embryonic development, undifferentiated progenitor cells balance the generation of additional with differentiation. Within developing limb, cartilage differentiate from mesodermal progenitors in an ordered process that results specification correct number appropriately sized skeletal elements. The internal pathways by which these maintain state while preserving their capacity to is unknown. Here, we report arginine methyltransferase PRMT5 has a crucial role maintaining cells. Mouse...

In most tetrapod vertebrates, limb skeletal progenitors condense with postaxial dominance. Posterior elements (such as ulna and fibula) appear prior to their anterior counterparts (radius tibia), followed by digit-appearance order continuing polarity. The only exceptions are urodele amphibians (salamanders), whose develop preaxial polarity who also notable for unique ability regenerate complete limbs adults. mechanistic basis this dominance has remained an enigma even been proposed relate...

A multiple protein–DNA complex formed at a human α-globin locus-specific regulatory element, HS-40, confers appropriate developmental expression pattern on embryonic ζ-globin promoter activity in humans and transgenic mice. We show here that introduction of 1-bp mutation an NF-E2/AP1 sequence motif converts HS-40 into erythroid-specific locus-control region. Cis-linkage with this region, contrast to the wild-type allows erythroid lineage-specific derepression silenced fetal adult...

During limb development, the interdigital mesenchyme has been proposed to play a signaling role instructing morphogenesis of different digit types, as well undergoing programmed cell death necessary free digits in animals not adapted for swimming or flying. We have generated conditional, tamoxifen-dependent Cre line, Bmp2CreER, which drives highly selective recombination restricted distal mesoderm, largely interdigits, and selectively active ligament but tendon progenitors at later stages....
