Abstract Gene profiling studies of patients with chronic lymphocytic leukemia (CLL) has revealed increased expression Ror1, a cell surface receptor tyrosine kinase. The aim present study was to analyze gene and protein Ror1 in CLL cells normal blood leukocytes. analysis reverse transcription‐polymerase chain reaction ROR1 that all ( n = 100) spontaneously expressed mRNA whereas enriched B T as well granulocytes from healthy donors 10) were negative. A strong nonphysiological activation...

In pulmonary sarcoidosis, CD4 + T-cells expressing T-cell receptor Vα2.3 accumulate in the lungs of HLA-DRB1*03 patients. To investigate receptor-HLA-DRB1*03 interactions underlying recognition hitherto unknown antigens, we performed detailed analyses expression on bronchoalveolar lavage fluid from sarcoidosis Pulmonary patients (n=43) underwent bronchoscopy with lavage. α and β chains were analysed by flow cytometry, DNA-sequenced, three-dimensional molecular models complexes generated....

Summary Sarcoidosis is a granulomatous disorder of unknown aetiology. The presence Mycobacterium tuberculosis catalase-peroxidase (mKatG) in sarcoidosis tissue has been reported. T helper type 1 (Th1) responses against mKatG have previously observed. However, little known about interleukin (IL)-17 and Th17 sarcoidosis. Here, we investigated the levels IL-17 frequencies IL-17-producing cells responding to patients with different prognosis. Peripheral blood bronchoalveolar lavage (BAL) were...

Sarcoidosis is a granulomatous disease of unknown aetiology, mainly affecting the lungs. Recently, T-cell responses towards specific mycobacterial protein, catalase–peroxidase (mKatG), were observed in sarcoidosis patients. Bronchoalveolar lavage (BAL) fluid and peripheral blood obtained from total 23 patients, whom 13 had Löfgren’s syndrome lung accumulations receptor AV2S3+ T-cells. Using six-colour flow cytometry combination with intracellular cytokine staining, subsets studied regard to...

Mucosal epithelial cell integrity is an important component of innate immunity and it protects the host from environment rich in microorganisms. Virulence factors Gram-negative bacteria [e.g. lipopolysaccharide (LPS)] induce significant pro-inflammatory cytokine expression. Monoamine oxidase (MAO) inhibitors reduce expression a variety inflammatory models may therefore have therapeutic potential for number diseases. We tested anti-inflammatory recently developed reversible MAO-B inhibitor...

Proline/arginine-rich end leucine-rich repeat protein (PRELP) belongs to the small proteoglycan (SLRP) family, normally expressed in extracellular matrix of collagen-rich tissues. We have previously reported on another SLRP, fibromodulin (FMOD) patients with chronic lymphocytic leukemia (CLL). PRELP is structurally similar FMOD adjacent localization chromosome 1 (1q32.1). As cluster-upregulation genes may occur malignancies, aim our study was analyze expression CLL. (RT-PCR) all CLL (30/30),...

To develop an efficient dendritic cell (DC)-based immunotherapy protocol, we examined whether simultaneous pulsing of DCs with a given antigen and third-party could enhance their presentation capacity. Purified splenic Balb/c mice were pulsed separately immunoglobulin G, ovalbumin, conalbumin, P15 peptide Mycobacterium tuberculosis, prostate-specific or double combinations the aforementioned antigens. In some settings, 1 mixed equally those another antigen. Antigen-pulsed injected into...

Smoking influences the immune system in different ways and, hypothetically, effects on pulmonary effector and regulatory T cells emerge as potentially detrimental. Therefore, we characterized frequencies characteristics of CD4+ CD8+ cell subsets blood lungs young tobacco smokers. ABSTRACT Aim Porphyromonas gingivalis lipopolysaccharide ( Pg LPS) is a significant virulence factor and driver of early innate immune responses in epithelial cells. The presence LPS immediate proximity to gingival epithelium induces inflammatory responses. In primary human keratinocytes (HGK), we utilized transcriptome analysis elucidate the change gene expression induced by LPS. Methods HGK cell cultures were treated with (4 h), RNA was extracted prepared for sequence (RNAseq) analysis....

Monoamine oxidase (MAO) inhibitors reduce inflammation in a number of vitro and vivo models. This finding led to the development novel MAO-B selective inhibitor (RG0216) designed blood-brain barrier penetration. To elucidate RG0216's regulatory role inflammation-relevant signaling pathways, we employed transcriptome analytic approach identify genes that are differentially regulated by RG0216 then globally identified which biological pathways were altered this drug.

The presence of mycobacterial antigens, and recently a specific protein, M. tuberculosis catalase-peroxidase (mKatG), in sarcoidosis tissue has been reported. Th1 cell responses against mKatG have observed lung peripheral blood cells. IL-17 IFNγ were evaluated ELISPOT after stimulation bronchoalveolar lavage (BAL) fluid cells with PPD proteins.
