The spread of H5N1 avian influenza viruses (AIVs) from China to Europe has raised global concern about their potential infect humans and cause a pandemic. In spite substantial threat human health, remarkably little AIV whole-genome information is available. We report here preliminary analysis the first large-scale sequencing AIVs, including 2196 genes 169 complete genomes. combine this new with public data identify gene alleles, persistent genotypes, compensatory mutations, virulence determinant.
A total of 150 amino acid sequences vitamin B 6 ‐dependent enzymes are known to date, the largest contingent being furnished by aminotransferases with 51 14 different enzymes. All aminotransferase were aligned using algorithms for sequence comparison, hydropathy patterns and secondary structure predictions. The could be divided into four subgroups on basis their mutual structural relatedness. Subgroup I comprises aspartate, alanine, tyrosine, histidinol‐phosphate, phenylalanine...
Pyridoxal‐5′‐phosphate‐dependent enzymes catalyze manifold reactions in the metabolism of amino acids. A comprehensive comparison acid sequences has shown that most these can be assigned to one three different families homologous proteins. The each family were aligned and their homology confirmed by profile analysis. Scrutiny catalyzed showed affiliation with structurally defined correlates cases regio‐specificity. In largest family, covalency changes substrate occur at same carbon atom...
This article describes an open-access gene expression database analyzed for more than 2,000 genes on mouse nervous system tissue in the coronal, sagittal, and transverse orientation representing multiple developmental ages.
Avian influenza viruses have adapted to human hosts, causing pandemics in humans. The key host-specific amino acid mutations required for an avian virus function humans are unknown. Through multiple-sequence alignment and statistical testing of each aligned acid, we identified markers that discriminate from viruses. We applied strict thresholds select only which highly preserved isolates over time. found a subset these persistent host exist all pandemic sequences 1918, 1957, 1968, while...
A data base was compiled containing the amino acid sequences of 12 aspartate aminotransferases and 11 other aminotransferases. comparison these by a standard alignment method confirmed previously reported homology all Escherichia coli tyrosine aminotransferase. However, no significant similarity between proteins any detected. more rigorous analysis, focusing on short sequence segments rather than total polypeptide chain, revealed that rat aminotransferase Saccharomyces cerevisiae...
Abstract To improve secondary structure predictions in protein sequences, the information residing multiple sequence alignments of substituted but structurally related proteins is exploited. A database comprised 70 families and a total 2, 500 some which were aligned by tertiary structural superpositions, was used to calculate residue exchange weight matrices within α ‐helical, β ‐strand, coil substructures, respectively. Secondary made based on observed substitutions local regions largest...
Abstract The DEAE‐cellulose linked cells of Methylosinus trichosporium displaying high specific methane mono‐oxygenase activity (66 μmol oxidized/h mg cells) were used for methanol biosynthesis from biogas derived in a batch and continuous cell reactor. optimum cell‐to‐carrier ratio was determined to be 0.5 g cells/g dry weight cellulose. Batch experiments indicated that 100 m M phosphate ion concentration necessary inhibit further oxidation methanol; excess oxygen supply favored...
Self-inactivating (SIN)-lentiviral vectors have safety and efficacy features that are well suited for transduction of hematopoietic stem cells (HSCs), but generation vector at clinical scale has been challenging. Approximately 280 liters an X-Linked Severe Combined Immunodeficiency Disorder (SCID-X1) SIN-lentiviral in two productions from a stable cell line were concentrated to final titers 4.5 7.2×108 tu/ml. These preparations three additional development-scale evaluated human CD34+ vitro...
Therapy-related acute myeloid leukemias (t-AML) with translocations of the MLL gene are associated use topoisomerase II inhibitors. We established emergence malignant clone in a child who developed t-AML t(11;19) (q23;p13.3) during treatment for lymphoblastic leukemia (ALL). The MLL-ENL and reciprocal ENL-MLL genomic fusions their chimeric transcripts were characterized from samples collected at time diagnosis. used PCR patient-specific primers to establish fusion serially obtained DNA...
Bacterial deletion mutants have indicated that the gene products of cobC, nifS, pabC and malY participate in important metabolic pathways, i.e. cobalamin synthesis, nitrogen fixation, synthesis p-aminobenzoate regulation maltose system, respectively. However, proteins themselves their specific functions not yet been identified. In course our studies on evolutionary relationships among aminotransferases, we found above are homologous to aminotransferases. Profile analysis [Gribskov, M.,...
Techniques for measuring methanol accumulated by resting cells of Methylosinus trichosporium in the narrow range 70-80mM phosphate concentration have been standardized. The maximum amount methanol, 6μmol•mg-1•hr-1, was at a cell density 2mg•ml-1 (w/v) early stationary phase pH 6.5, temperature 35°C. phosphate-dependent inhibition dehydrogenase found to be uncompetitive and reversible. Further, 80mM inhibited methane monooxygenase formate 16% 20% respectively. A possible biochemical...
From a single transgenic line harboring five Tnt1 transposon insertions, we generated near-saturated insertion population in Medicago truncatula. Using thermal asymmetric interlaced-polymerase chain reaction followed by sequencing, recovered 388 888 flanking sequence tags (FSTs) from 21 741 lines this population. FST recovery 14 using the whole-genome sequencing (WGS) and/or Tnt1-capture approaches suggests an average of 80 insertions per line, which is more than previous estimation 25...
Drought stress reduces crop biomass yield and the profitability of rainfed agricultural systems. Evaluation populations or accessions adapted to diverse geographical agro-climatic environments sheds light on beneficial plant responses enhance optimize in resource-limited environments. This study used morphological physiological characteristics leaves roots from two different alfalfa subspecies during progressive drought imposed controlled field conditions. Two soils (Experiments 1 2) water...
Selenocysteine synthase from Escherichia coli is a pyridoxal‐5′‐phosphate‐containing enzyme which catalyses the conversion of seryl‐tRNA Sec into selenocysteyl‐tRNA . Analysis amino acid sequences indicated that selenocysteine belongs to α/γ superfamily pyridoxal‐5′‐phosphate‐dependent enzymes. To identify lysine residue carrying prosthetic group, genes coding for synthases Moorella thermoacetica and Desulfomicrobium baculatum were cloned sequenced their derived aligned with those E....
The 6-hydroxymethyl-7,8-dihydropterin pyrophosphokinase (HPPK) and dihydropteroate synthase (DHPS) enzymes catalyze sequential metabolic reactions in the folate biosynthetic pathway of bacteria lower eukaryotes. Both represent validated targets for development novel anti-microbial therapies. We report herein that genes which encode FtHPPK FtDHPS from biowarfare agent Francisella tularensis are fused into a single polypeptide. potential simultaneously targeting both modules with pterin...
Abstract Hairy vetch, a diploid annual legume species, has robust growth habit, high biomass yield, and winter hardy characteristics. Seed hardness is major constraint for growing hairy vetch commercially. Hard seeded cultivars are valuable as forages, whereas soft shatter resistant have advantages their use cover crop. Transcript analysis of was performed to understand the genetic mechanisms associated with important traits. RNA extracted from leaves, flowers, immature pods, seed coats,...
