PCR法による肉種鑑別を行う目的で,既知のウシ,ブタ,ニワトリのミトコンドリアD-loop近傍の塩基配列の相同性を調査し,それらの3種を対象に3セットのプライマーBFとBR,PF(BFと同一のもの)とPR,CFとCRを合成した.3種食肉と各種肉製品から抽出したDNAを鋳型とし,94°C-2分間,66-68°C-2分間,72°C-3分間で35回の繰り返し反応でPCR増幅を行った.増幅されたDNAは2%アガロースゲル電気泳動を用いて分析した.その結果,設計通りにウシ,ブタ,ニワトリに特異的なDNA断片(518,321,400bp)が増幅された.この方法を生肉,加熱肉及びいくつかの肉製品について適用したところ,試料に含まれていると推定される種特異的なDNA断片が検出された.さらに,ウシ,ブタ,ニワトリの3セットプライマーを混合することで,3種類の食肉の存在を一度の分析で鑑定できた.羊肉を牛肉と誤認する可能性は否定された.混合DNAを用いてウシ,ブタ,ニワトリの複数種のDNAの検出限度について検討した結果,混合サンプル中のそれぞれの検出限界は0.1%,0.001%,1%で,いずれの場合で...

黒毛和種腿肉の熟成中における筋漿蛋白質の変化を詳細に追究する目的で,筋漿画分を細胞質画分として分画し,種々の筋漿蛋白質含量の変化およびSDS-ポリアクリドアミドゲル電気泳動(PAGE)像の変化を指標として調査した.屠殺後0,7,14,21,28日間熟成した試料の種々の筋漿蛋白質含量を測定した結果,ミトコンドリア画分は熟成28日において熟成7,14,21日試料より有意(P<0.05)に低い値が得られた.さらに,リソソーム画分においては熟成7日,14日試料に比べて21日と28日で有意(P<0.05)に低い値が認められた.サイトゾル画分のSDS-PAGE像の結果から,ホスホリラーゼb,クレアチンキナーゼ,ホスホグリセロアルデヒド脱水素酵素は熟成に伴い顕著に減少した.一方,乳酸脱水素酵素,ホスホグリセロムターゼ,トリオースホスホイソメラーゼに相当する蛋白質は熟成7日,14日に比べて熟成21日と28日試料で増加した.
