Abstract The Pearson correlation coefficient (PCC) and the Mander's overlap (MOC) are used to quantify degree of colocalization between fluorophores. MOC was introduced overcome perceived problems with PCC. two coefficients mathematically similar, differing in use either absolute intensities or deviation from mean (PCC). A range correlated datasets, which extend limits PCC, only evoked a limited response MOC. PCC is unaffected by changes offset while increases when positive. Both independent...

We have recently shown that stimulation of TNF-R2 selectively enhances apoptosis induction by the death receptor TNF-R1. Here, we demonstrate CD30 or CD40 also leads to selective enhancement TNF-R1-induced cell death. Enhancement was correlated with depletion endogenous TRAF2 within 1 6 hours. Selective prestimulation for several hours inhibited TNF-R2-induced activation anti-apoptotic NF-κB pathway up 90% and dramatically enhanced this receptor. When both TNF-receptors were stimulated...

Cyanobacteria are unique prokaryotes since they in addition to outer and plasma membranes contain the photosynthetic (thylakoids). The of Synechocystis 6803, which can be completely purified by density centrifugation polymer two-phase partitioning, have been found more complex than previously anticipated, i.e. appear essential for assembly two photosystems. A proteomic approach characterization cyanobacterial using two-dimensional gel electrophoresis mass spectrometry analysis revealed a...

Abstract Two related models for T cell signalling initiation suggest either that receptor (TCR) engagement leads to its recruitment ordered membrane domains, often referred as lipid rafts, where molecules are enriched or TCR-containing nanodomains coalesce upon TCR engagement. That domains form engagement, they do raft marker patching, has not been considered. The target of this study was differentiate between those three options. Plasma order followed in live cells at 37 °C using laurdan...

Abstract The impact of statins on COVID-19 remains unclear. This study aims to investigate whether statin exposure assessed both in the population and well-defined cohorts patients may affect risk severity using nationwide Swedish population-based register data. A ≥ 40 years was selected by age/sex-stratified random sampling from 1 Jan 2020. outcomes were identified SmiNet database, National Patient Register and/or Cause-of-Death linked with Prescribed Drug sociodemographic registers. Lipid packing is a fundamental characteristic of bilayer membranes. It affects all membrane-associated processes ranging from curvature generation to membrane fission. Yet, we lack detailed, mechanistic understanding how lipid directly these in cellular Here, address this by focusing on caveolae, small Ω-shaped invaginations the plasma which serve as key regulators sorting and mechano-responses. In addition caveolae coat proteins, core component that critically impacts both biogenesis,...

Summary It is widely recognized that the accuracy of colocalization measurements dependent upon quality source images. We demonstrate that, as image increases, measured colocalization, using Pearson and Spearman rank correlation coefficients, approaches true asymptotically. This means in practice it difficult to obtain images sufficient for accurate measurements. introduce replicate‐based noise corrected (RBNCC) which aligns with colocalization: a measurement made each fluorophore from pair...

The plasma membranes of eukaryotic cells are hypothesised to contain microdomains with distinct lipid and protein composition known as rafts. In T cells, cross-linking raft components triggers signalling cascades. We show that the T-cell antigen receptor (TCR) a tyrosine kinase, Lck, have patchy membrane distribution in Jurkat at reduced temperatures, although they continuous physiological temperature (37°C). GM1 displays after Triton X-100 extraction. archetypal non-lipid marker,...

Fluorescence correlation spectroscopy (FCS) is frequently used to study diffusion in cell membranes, primarily the plasma membrane. The coefficients reported membrane of same type and even within single cells typically display a large spread. We have investigated whether this spread can be explained by variations topography throughout surface, that changes amount FCS focal volume at different locations. Using FCS, we found dye DiI apical was consistently faster above nucleus than cytoplasm....

Cells are neither flat nor smooth, which has serious implications for prevailing plasma membrane models and cellular processes like cell signalling, adhesion molecular clustering. Using probability distributions from diffusion simulations, we demonstrate that 2D 3D Euclidean distance measurements substantially underestimate on non-flat surfaces. Intuitively, the shortest within surface (SWSD), geodesic distance, should reduce this problem. The SWSD is accurate foldable surfaces but, although...
