Rationale: Small molecule inhibitors of the acetyl-histone binding protein BRD4 have been shown to block cardiac fibrosis in preclinical models heart failure (HF). However, since target ubiquitously, it is unclear whether this chromatin reader functions cell type-specific manner control pathological myocardial fibrosis. Furthermore, molecular mechanisms by which stimulates transcriptional program for remain unknown. Objective: We sought test hypothesis that a cell-autonomous and...
Glioblastoma is an incurable brain cancer characterized by high genetic and pathological heterogeneity. Here, we mapped active chromatin landscapes with gene expression, whole exomes, copy number profiles, DNA methylomes across 44 patient-derived glioblastoma stem cells (GSCs), 50 primary tumors, 10 neural (NSCs) to identify essential super-enhancer (SE)–associated genes the core transcription factors that establish SEs maintain GSC identity. GSCs segregate into two groups dominated distinct...
Background: Diastolic dysfunction (DD) is associated with the development of heart failure and contributes to pathogenesis other cardiac maladies, including atrial fibrillation. Inhibition histone deacetylases (HDACs) has been shown prevent DD by enhancing myofibril relaxation. We addressed therapeutic potential HDAC inhibition in a model established preserved ejection fraction. Methods: Four weeks after uninephrectomy implantation deoxycorticosterone acetate pellets, when was clearly...
Abstract When Emperor Akbar (r. 1556–1605) conquered Burhanpur, the capital of Khandesh sultanate, in 1601, he catalyzed a decades-long process transforming city into regional Mughal empire. In this essay, I argue that Burhanpur was constructed three stages spatial and architectural developments over course decades. first stage, symbolically appropriated Burhanpur’s preexisting monumental architecture to demonstrate his conquest city. second subimperial officers expanded through prolific...
Abstract We examined participants' reading and recall of informed consent documents presented via paper or computer. Within each presentation medium, we the document as a continuous paginated to simulate common computer formats. Participants took slightly longer read recalled more information from documents. concluded that obtaining online is not substantially different than it presentation. also provide suggestions for improving consent-in both face-to-face experiments.
Risk perception and the cultural theory of risk have often been contrasted in relation to risk‐related policy making; however, local context which risks are experienced, an important component everyday decision making, remains understudied. What is unclear extent localized community beliefs behaviors depend on larger belief systems about (i.e., worldviews). This article reports a study designed understand relative importance health perceptions (threat harm); worldviews (cultural biases);...
In this paper we report on the results of a residential questionnaire survey ( N = 1088) exploring relative importance health-risk perception as compared with social–contextual determinants urban pesticide bylaw support in two Canadian cities: Calgary and Halifax. Multivariate analysis was used order to determine model estimates for five outcome variables: policy preference, pesticide-risk perception, weed-free aesthetic, use, chemical dissent. Findings indicate that strongest (based odds)...
Dispossession from social and ecological support systems is a major concern for many Indigenous communities. In response to community health challenges in these settings number of initiatives such as youth mentorship programs have shown some value enhancing adaptive capacity. The pilot Going Off, Growing Strong program provides opportunities at-risk engage community- land-based activities build relationships with positive adult role models Nain, Nunatsiavut (Labrador, Canada). This paper...
This paper focuses on policy change under scientific uncertainty. is done by exploring two case studies of Canadian pesticide evolution applied to Sabatier's ‘Advocacy Coalition Framework’. ‘Stakeholder’ and ‘narrative policy’ analyses are emphasised understand changes in Calgary (public education without a restrictive by-law) Halifax (restrictive by-law). The parallel consist qualitative interviews with members advisory committees each city content analysis local newspaper articles. Key...
Abstract Small molecule inhibitors of the acetyl-histone binding protein BRD4 have been shown to block cardiac fibrosis in pre-clinical models heart failure (HF). However, mechanisms by which promotes pathological myocardial remain unclear. Here, we demonstrate that functions as an effector TGF-β signaling stimulate conversion quiescent fibroblasts into Periostin ( Postn )-positive cells express high levels extracellular matrix. undergoes stimulus-dependent, genome-wide redistribution...
Introduction: Diastolic dysfunction is associated with the development of heart failure preserved ejection fraction and other cardiac maladies, including atrial fibrillation. Inhibition histone deacetylase (HDAC) activity has previously been shown to prevent diastolic by enhancing myofibril relaxation. Here, we further addressed therapeutic potential HDAC inhibition in a model established fraction. Methods: Four weeks following uninephrectomy (UNX) implantation deoxycorticosterone acetate...
