Abstract Metagenomics revolutionized the understanding of relations among human microbiome, health and diseases, but generated a countless number sequences that have not been assigned to known microorganism 1 . The pure culture prokaryotes, neglected in recent decades, remains essential elucidating role these organisms 2 We recently introduced microbial culturomics, culturing approach uses multiple conditions matrix-assisted laser desorption/ionization–time flight 16S rRNA for identification...
Artemether-lumefantrine and artesunate-amodiaquine are used as first-line artemisinin-based combination therapies (ACTs) in west Africa. Pyronaridine-artesunate dihydroartemisinin-piperaquine potentially useful for diversification of ACTs this region, but further safety efficacy data required on malaria retreatment. We did a randomised, multicentre, open-label, longitudinal, controlled phase 3b/4 clinical trial at seven tertiary centres Burkina Faso, Guinea, Mali. Eligible participants first...
Archaeal sequences have been detected in human colostrum and milk, but no studies determined whether living archaea are present either of these fluids. Methanogenic neglected since they not by usual molecular culture methods. By using improved DNA detection protocols microbial techniques associated with antioxidants previously developed our center, we investigated the presence methanogenic specific Methanobrevibacter smithii oralis real-time PCR milk. M. was isolated from 3 5 milk (day 10)...
Abstract Gut microbial dysbiosis has been shown to be an instrumental factor in severe acute malnutrition (SAM) and particularly, the absence of Methanobrevibacter smithii, a key player energy harvest. Nevertheless, it remains unknown whether this reflects immaturity or loss microbiota. In order assess that, we performed case–control study Mali using propensity score weighting approach. The presence M. smithii was tested quantitative PCR on faeces collected from SAM children at inclusion...
An anaerobic bacterium, strain AT2T, was isolated from the fresh stool sample of a healthy French man using culturomics approach. The 16S rRNA gene sequence analysis showed that AT2T had 95.2 % nucleotide similarity with Gemmiger formicilisATCC 27749T, phylogenetically closest species standing in nomenclature. Cells are Gram-stain-negative, catalase- and oxidase-negative, obligately anaerobic, non-motile, non-spore-forming, rod-shaped, bacilli were mesothermophilic. major fatty acids C16 : 0...
Abstract Severe acute malnutrition (SAM) constitutes the leading risk for childhood mortality worldwide. Butyrate, a critical metabolite mucosal immunity produced by anaerobic gut microbes, has been found to be dramatically depleted in children experiencing fatal outcomes related SAM. However, association between butyrate depletion and microbiota alteration not yet fully explored. Here, we performed large group-matched case-control study aged 0-59 months Mali. We compared faecal short-chain...
Artemisinin-based combination therapies (ACTs) are the first-line treatment of uncomplicated malaria. The public health benefit and safety repeated administration a given ACT poorly studied. We conducted randomized trial comparing artemether-lumefantrine, artesunate plus amodiaquine (AS+AQ) sulfadoxine-pyrimethamine (AS+SP) in patients 6 months age older with malaria Mali from July 2005 to 2007. patient received same initial each subsequent episode except for failures where quinine was used....
We report the principal characteristics of 'Eisenbergiella massiliensis' sp. nov. strain AT11 (CSURP = P2120, DSM 101499) that was isolated from a stool sample collected after bariatric surgery 56-year-old obese French woman.
A new bacterium, strain AT3T, was isolated by microbial culturomics from a faecal sample Frenchman after bariatric surgery. The isolate exhibited 96.6% 16S ribosomal RNA gene nucleotide sequence similarity with Anaerotruncus colihominis WAL 14565T = CCUG 45055T CIP 107754T. Phenotypic and genomic characteristics showed that the represents novel species, for which name massiliensis sp. nov. is proposed. type AT3T CSUR P2007T DSM 100567T.
Paenibacillus ihumii sp. nov. strain AT5 (= CSUR 1981 = DSM 100664) is the type of P. ihumii. This bacterium was isolated from a stool sample morbidly obese French patient using culturomics approach. The genome this Gram-negative, facultative anaerobic, motile and spore-forming bacillus 5 924 686 bp long. Genomic analysis identified 253 (5%) 3812 genes as ORFans at least 2599 (50.03%) 5194 orthologous proteins not shared with closest phylogenetic species.
Abstract The human gut is composed of a large diversity microorganisms, which have been poorly described. Here, using culturomics, new concept based on the variation in culture conditions and MALDI ‐ TOF MS identification, we proceed to explore microbial complex ecosystem gut. Using this approach, isolated strain AT 8 T (= CSUR P2118 = DSM 101782) from stool specimens collected 51‐year‐old obese French woman. Strain strictly anaerobic, nonmotile, nonspore‐forming gram‐positive coccus that do...
ABSTRACT Dihydroartemisinin-piperaquine is efficacious for the treatment of uncomplicated malaria and its use increasing globally. Despite positive results in fighting malaria, inhibition Kv11.1 channel (hERG; encoded by KCNH2 gene) piperaquine has raised concerns about cardiac safety. Whether genetic factors could modulate risk piperaquine-mediated QT prolongations remained unclear. Here, we first profiled landscape variability using data from 141,614 individuals. Overall, found 1,007...
Most malaria-endemic countries use artemisinin-based combination therapy (ACT) as their first-line treatment. ACTs are known to be highly effective on asexual stages of the malaria parasite. Malaria transmission and spread resistant parasites depend infectivity gametocytes. The effect current ACT regimens gametocyte is unclear. This study aimed determine gametocytes Anopheles gambiae following treatment in field.During a randomised controlled trial Bougoula-Hameau, Mali, conducted from July...
Butyricimonas phoceensis strain AT9 (= CSUR 2478 = DSM 100838) was isolated from a stool sample morbidly obese French patient living in Marseille using the culturomics approach. The genome of this Gram-negative-staining, anaerobic and non–spore forming rod bacillus is 4 736 949 bp long contains 3947 protein-coding genes. Genomic analysis identified 173 genes as ORFans (4.5%) 1650 orthologous proteins (42%) not shared with closest phylogenetic species, virosa. Its major fatty acid branched...
A recent randomized controlled trial, the WANECAM (West African Network for Clinical Trials of Antimalarial Drugs) conducted at seven centers in West Africa, found that artemether-lumefantrine, artesunate-amodiaquine, pyronaridine-artesunate, and dihydroartemisinin-piperaquine all displayed good efficacy. However, artemether-lumefantrine was associated with a shorter interval between clinical episodes than other regimens.
Enterococcus massiliensis strain sp. nov. (= CSUR P1927 = DSM 100308) is a new species within the genus Enterococcus. This was first isolated from fresh stool sample of man during culturomics study intestinal microflora. Gram-positive cocci, facultative anaerobic and motile. E. negative for mannitol positive β-galactosidase, contrary to gallinarum.
