ABSTRACT Most known basic-region helix-loop-helix (bHLH) proteins belong to a superfamily of transcription factors often involved in the control growth and differentiation. Therefore, inappropriate expression genes encoding bHLH is frequently associated with developmental dysfunction. In our previously reported study, novel protein-encoding gene (AO090011000215) Aspergillus oryzae was identified. The gene-disrupted strain found produce dense conidia, but sparse sclerotia, relative parent...

ManR, a DNA-binding protein possessing the Zn(II)2Cys6 binuclear cluster domain, acts as positive regulator of mannanolytic enzyme genes in Aspergillus oryzae. In this study, we found that ManR controlled expression not only but also cellulolytic A. oryzae, based on DNA microarray analysis manR-disruptant and manR-overexpressing strain. A new model for regulation cellulase hemicellulase mediated by XlnR oryzae is proposed.

Aspergillus fumigatus causes a range of human and animal diseases collectively known as aspergillosis. A . possesses expresses genetic determinants virulence, which facilitate colonisation disease progression, including the secretion mycotoxins. Gliotoxin (GT) is best studied mycotoxin with wide toxic effects that impair immune cell function. GT also highly to this fungus has evolved self-protection mechanisms include (i) efflux pump GliA, (ii) neutralising enzyme GliT, (iii) negative...

We performed random sequencing of cDNAs from nine biologically or industrially important cultures the valuable fungus Aspergillus oryzae to obtain expressed sequence tags (ESTs). Consequently, 21 446 raw ESTs were accumulated and subsequently assembled 7589 non-redundant consensus sequences (contigs). Among all contigs, 5491 (72.4%) derived only a particular culture. These included 4735 (62.4%) singletons, i.e. lone overlapping with no others. data showed that consideration culture grown...

The Woronin body, a unique organelle found in the Pezizomycotina, plugs septal pore upon hyphal damage to prevent excessive cytoplasmic bleeding. Although it was previously shown that body buds out from peroxisome, relationship between peroxisomal proliferation/division and differentiation has not been extensively investigated. In this report, we examined whether Pex11 required for proliferation participates formation Aspergillus oryzae. A. oryzae contained two orthologous PEX11 genes were...

Abstract Esterified drimane-type sesquiterpene lactones such as astellolides display various biological activities and are widely produced by plants fungi. Given their low homology to known cyclases, the genes responsible for biosynthesis have not been uncovered yet. Here, we identified astellolide gene cluster from Aspergillus oryzae discovered a novel biosynthetic machinery consisting of AstC, AstI, AstK. All these enzymes annotated haloacid dehalogenase-like hydrolases, whereas AstC also...

ABSTRACT Aspergillus sojae belongs to the section Flavi but does not produce aflatoxins. The functionality of A. aflR gene ( aflRs ) was examined by transforming it into an Δ strain parasiticus , derived from a nitrate-nonutilizing, versicolorin A (VERA)-accumulating strain. aflRp transformants produced VERA, did not. Even when placed under control amylase amyB promoter oryzae amy p ):: VERA. chimeric construct containing plus N- and C-terminal coding regions could restore VERA production,...

Ku genes play a key role in the non-homologous end-joining pathway. We have identified Ku70 and Ku80 homologs koji molds Aspergillus sojae oryzae, constructed disruption mutants of Ku70, Ku80, Ku70-80 to characterize phenotypic change these mutants. Neither Ku70- nor Ku80-disrupted strains show hypersensitivity DNA damaging agents methylmethane sulfonate (MMS) phleomycin. Moreover, undesirable phenotypes, such as poor growth or repressed conidiospore formation, were not observed Ku-disrupted...

Abstract Aspergillus oryzae is a fungus widely used in traditional Japanese fermentation industries. Its inability to produce mycotoxins, due mutation or transcriptional repression of the genes responsible for their biosynthesis, consistent with hypothesis that A . domesticated species derived from flavus , wild well‐known producer aflatoxin. Glutaminase, an enzyme that catalyzes the conversion of L-glutamine to L-glutamate, enhances umami taste in soy sauce. The Aspergillus sojae genome contains 10 glutaminase genes. In this study, we estimated approximately 60% glutamate sauce is produced through reaction. To determine which involved production, prepared sauces using single and multiple gene disruptants A. sojae. concentration ΔgahA-ΔgahB-ΔggtA-Δgls disruptant was lower than control strain, whereas it decreased by 20-30%...

A urate oxidase (uricase) gene was cloned from Candida utilis with an oligonucleotide probe based on the amino acid sequence of cyanogen bromide-cleaved uricase. The uricase contains 909 base pairs and encodes a protein predicted mass 34,193 Da. similar (49% match in sequence) to Aspergillus flavus. has four cysteines one them, Cysl68, participates enzyme activity. This expressed level about 20% total cellular Escherichia coli cell as soluble functional form.
