Aspergilli known as black- and white-koji molds which are used for awamori, shochu, makgeolli other food beverage fermentations, reported in the literature A. luchuensis, kawachii, or acidus. In order to elucidate taxonomic position of these species, available ex-type cultures were compared based on morphology molecular characters. kawachii acidus showed same banding patterns RAPD, three species had rDNA-ITS, β-tubulin calmodulin sequences differed from those closely related niger...

Ustiloxins were found recently to be the first example of cyclic peptidyl secondary metabolites that are ribosomally synthesized in filamentous fungi. In this work, two function-unknown genes (ustYa/ustYb) gene cluster for ustiloxins from Aspergillus flavus experimentally involved cyclization peptide. Their homologous observed mainly fungi and mushrooms. They have "HXXHC" motifs might form active sites. Computational genome analyses showed these frequently located near candidate ribosomal...

ABSTRACT The basidiomycetous yeast Pseudozyma antarctica T-34 is an excellent producer of mannosylerythritol lipids (MELs), members the multifunctional extracellular glycolipids, from various feedstocks. Here, genome sequence P. was determined and annotated. Analysis might provide insights into properties this that make it superior for use in production functional leading to further development industrial applications.

We performed random sequencing of cDNAs from nine biologically or industrially important cultures the valuable fungus Aspergillus oryzae to obtain expressed sequence tags (ESTs). Consequently, 21 446 raw ESTs were accumulated and subsequently assembled 7589 non-redundant consensus sequences (contigs). Among all contigs, 5491 (72.4%) derived only a particular culture. These included 4735 (62.4%) singletons, i.e. lone overlapping with no others. data showed that consideration culture grown...

ABSTRACT Tetragenococcus halophila D10 catalyzes the decarboxylation of l -aspartate with nearly stoichiometric release -alanine and CO 2 . This trait is encoded on a 25-kb plasmid, pD1. We found in this plasmid putative asp operon consisting two genes, which we designated aspD aspT , encoding an -aspartate-β-decarboxylase (AspD) aspartate-alanine antiporter (AspT), respectively, determined nucleotide sequences. The sequence analysis revealed that genes pD1 were following order: promoter →...

The biological role of ExgA (Exg1), a secretory β-1,3-exoglucanase Aspergillus oryzae, and the expression pattern exgA (exg1) gene were analyzed. disruptant exgA-overexpressing mutant constructed, phenotypes both mutants compared. Higher mycelial growth rate conidiation efficiency observed for than when β-1,3-glucan was supplied as sole carbon source. On other hand, no difference in phenotype between them presence or absence inhibitors cell wall β-glucan remodeling grown with glucose....

Directed evolution of biomolecules such as DNA, RNA and proteins containing high diversity has emerged an effective method to obtain molecules for various purposes. In the recent past, from non-immunoglobulins have attracted attention they mimic antibodies with respect binding potential provide further advantages. this regard, we attempted explore a three-finger neurotoxin protein (3F). 3F are small (~7 kDa), structurally well defined, thermally stable resistant proteolysis that presents...

Ustiloxin B, originally isolated from the fungus Ustilaginoidea virens, is a known inhibitor of microtubule assembly. B also produced by Aspergillus flavus and synthesized through ribosomal peptide synthesis pathway. In A. flavus, gene cluster associated with ustiloxin production contains 15 genes including those encoding fungal C6-type transcription factor precursor. Although koji mold oryzae, which genetically close to has corresponding cluster, it does not produce may be explained fact...

We constructed a protein expression vector with an improved enoA promoter that harbored 12 tandem repeats of the cis-acting element (region III) Aspergillus oryzae. The yielded reporter beta-glucuronidase (GUS) activity approximately 30-fold original promoter. Northern blot analysis confirmed GUS was increased at transcriptional level. transformant harboring seven copies novel showed more than 100,000 U/mg protein, which 30% all cell-free soluble proteins.
