To understand roles of transcriptional factors and miRNAs in regulating gene expression the epididymis from postnatal development through aging, systematic profiling genes expressed newborn, young adult, aged human epididymides was performed by cDNA array miRNA analysis, respectively. The newborn fewest mRNAs but largest number miRNAs, whereas adult most a negative correlation between during aging. By integrative set targets were predicted based on arrays. In epididymis, 127 exclusively or...
DNA-encoded chemical libraries (DELs) are a cost-effective technology for the discovery of novel probes and drug candidates. A major limiting factor in assembling productive DELs is availability DNA-compatible reactions aqueous media. In an effort to increase accessibility structural diversity small molecules displayed by DELs, we developed robust Suzuki-Miyaura reaction protocol that compatible with DNA structures. By employing water-soluble Pd-precatalyst, conditions allow efficient...
Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has killed more than 4 million humans globally, but there is no bona fide Food and Drug Administration-approved drug-like molecule to impede the COVID-19 pandemic. The sluggish pace of traditional therapeutic discovery poorly suited producing targeted treatments against rapidly evolving viruses. Here, we used an affinity-based screen billion DNA-encoded molecules en masse identify a potent class virus-specific inhibitors SARS-CoV-2...
The discovery of monokinase-selective inhibitors for patients is challenging because the 500+ kinases encoded by human genome share highly conserved catalytic domains. Until now, no selective unique a single transforming growth factor β (TGFβ) family transmembrane receptor kinase, including bone morphogenetic protein type 2 (BMPR2), have been reported. This dearth receptor-specific kinase hinders therapeutic options skeletal defects and cancer as result an overactivated BMP signaling...
Fishway monitoring can verify the effectiveness of fishway, optimise operation mode, and achieve scientific management fishway operations. Traditional approaches, hindered by their inefficiency substantial disruption fish, are ill-suited for long-term surveillance; thus, employing video coupled with object detection technology presents an alternative or complementary solution. However, challenges such as constrained computational capacity onsite equipment in fishways, complexities involved...
In this paper, based on the improved DeepSORT algorithm, four target species of passing fish ( Schizothorax o’connori , waltoni Oxygymnocypris stewartii and Schizopygopsis younghusbandi ) from a fishway project in middle reaches Y River were used to achieve dynamic identification automatic counting using fishways monitoring video. This method YOLOv5 model as detection model. view large deformation by body twisting, network structure re-identification (ReID) was deepened strengthen feature...
A strategy for DNA-compatible, palladium-catalyzed hydroxycarbonylation of (hetero)aryl halides on DNA–chemical conjugates has been developed. This method generally provided the corresponding carboxylic acids in moderate to very good conversions iodides and bromides, poor chlorides. These conditions were further validated by application within a DNA-encoded chemical library synthesis subsequent discovery enriched features from selection experiments against two protein targets.
To explore the functions of human ribonuclease 9 (RNase 9), we constructed a mammalian fusion expression vector pcDNA-hRNase9, prepared recombinant RNase 9-His protein from HEK293T cells and determined its N-terminal amino acid sequences. According to mature protein, was in E. coli. Ribonucleolytic activity antibacterial were detected, distribution on tissues ejaculated spermatozoa vitro capacitated analyzed via indirect immunofluorescence assay. The results showed that did not exhibit...
Chemiluminogenic bioimaging has emerged as a promising paradigm due to its independence from light excitation, thereby circumventing challenges related penetration depth and background autofluorescence. However, the availability of effective chemiluminophores remains limited, which substantially impedes their bio‐applications. Herein, we discovered for first time that cyclometalated iridium(III) Schiff base complexes can unexpectedly generate chemiluminescence. Notably, chemiluminescence...
Somatic cell nuclear transfer (SCNT) was used to generate patient-specific embryonic stem cells (ESCs) from blastocysts cloned by (ntESCs). In this study, a total of 135 oocytes were obtained 12 healthy donors (30-35 years). Human oocytes, within 2 h following transvaginal aspiration, enucleated using Spindle Imaging System position the spindle and chromosomes that lay on metaphase plate, Zona Infrared Laser Optical open single hole in zona pellucida at ~ o'clock position. fibroblasts...
Abstract The development of SARS-CoV-2 main protease (M pro ) inhibitors for the treatment COVID-19 has mostly benefitted from X-ray structures and preexisting knowledge inhibitors; however, an efficient method to generate M inhibitors, which circumvents such information would be advantageous. As alternative approach, we show here that DNA-encoded chemistry technology (DEC-Tec) can used discover . An affinity selection a 4-billion-membered chemical library (DECL) using as bait produces novel...
MicroRNAs (miRNA) are known to play a key role in the etiology and treatment of epilepsy through controlling expression gene. However, miR-125a-5p is little known. Epilepsy rat models was induced by Pentylenetetrazol (PTZ) miR- 125a-5p profiles hippocampus were investigated our experiment. Also, relationship between calmodulin-dependent protein kinase IV (CAMK4) identified related mechanism also illustrated. The mRNA levels evaluated quantitative real time polymerase chain reaction (qRT-PCR)....
C-type lysozyme genes (Lyzls) belong to the class of lysozymes and are highly expressed in testis epididymis. The members Lyzl4 Spaca3 have been reported play a role sperm-egg binding fertilisation mice. However, function remaining two mouse c-type genes, Lyzl1 Lyzl6, is still not clear. In present study, we analysed tissue expression androgen-dependent possible contribution human recombinant LYZL6 (rLYZL6) immunity. The Lyzls was detected by RT-PCR, Western blots, immunohistochemistry...
Heat stress is demonstrated to have an effect on the function of male testis, however, limited information has been reported its effects sperm maturation. In present study, a comparative proteomic analysis was performed rat caput epididymal fluids responsible for maturation, identify key heat‑stress‑associated maturation proteins. The results 21 proteins corresponding 29 differential protein spots, including 10 downregulated and 11 upregulated in heat treatment group. Functional that these...
Peroxiredoxin 6 (Prdx6) is widely expressed in mammalian tissues. Our previous study demonstrated that Prdx6 was human epididymis, present seminal fluid, and spermatozoa. The protective role of maintaining the viability DNA integrity spermatozoa also detected. Here, we demonstrate potential mechanism epididymis epithelial cells (HEECs).Western blotting used to measure expression levels key proteins Janus kinase/signal transducer activator transcription (JAK/STAT) signaling pathway....
