Structures formed by self-assembly of α- and β-1-n-dodecyl d-maltosides in water depend on the configuration at anomeric center. The α-anomer forms quasi-spherical aggregates, while β-maltoside larger oblate ellipsoidal micelles. This difference behavior suggests that head group influences orientation polar residue hence packing monomers during self-assembly.
ABSTRACT Phenylketonuria (PKU) is an inborn error of metabolism responsible for accumulation phenylalanine, which leads to cognitive and developmental disorders if left untreated. Most studies adult PKU focus on neuropsychiatric complications, but new questions have been raised about systemic manifestations in adulthood. Fifteen adults with classic poor metabolic control 15 matched healthy controls were recruited compare their blood metabolomes by untargeted multimodal approach (polar,...
Abstract Attention-Deficit Hyperactivity Disorder (ADHD) is one of the most common neurodevelopmental disorder characterized by inattention, impulsivity, and hyperactivity. The neurobiological mechanisms underlying ADHD are still poorly understood, its diagnosis remains difficult due to heterogeneity. Metabolomics a recent strategy for holistic exploration metabolism well suited investigating pathophysiology diseases finding molecular biomarkers. A few clinical metabolomic studies have been...
The transcription factor Krüppel-like 10 (Klf10), also known as Tieg1 for TGFβ (Inducible Early Gene-1) is to control numerous genes in many cell types that are involved various key biological processes (differentiation, proliferation, apoptosis, inflammation), including metabolism and human disease. In skeletal muscle, particularly the soleus, deletion of Klf10 gene (Klf10 KO) resulted ultrastructure fiber disorganization mitochondrial deficiencies, characterized by muscular hypertrophy. To...
We investigated here the micellization in water of three surfactants: (N-dodecylamino)lactitol, (N-dodecyl)lactobionamide, and (N-acetyl N-dodecyl)lactosylamine, having n-alkyl chain lengths 12 carbon atoms different lactose-based polar heads. The is studied by small-angle neutron X-ray scattering. characteristic dimensions micelles are derived from I(q) curve fittings. demonstrated existence spherical or oblate ellipsoidal micellar structures. differences were attributed to steric hindrance...
In this study, we investigated the micellization in water of four surfactants from series (N-alkylamino)lactitols by small angle neutron and X-ray scattering. These differed one another number carbon atoms their alkyl chain: 8, 9, 10, or 12. The comparison I(q) curves intensity scattered micellar solutions as a function concentration indicated that micelle size was weakly dispersed at concentrations. C8 derivative presented singular behavior which may be attributed to formation premicellar...
Selection of the most promising radiotracer candidates for radiolabeling is a difficult step in development pharmaceuticals, especially brain. Mass spectrometry (MS) an alternative to study ex vivo characteristics candidates, but MS studies are complicated by pharmacologic doses injected and dissection regions candidate biodistribution. In this study, we tested ability triple quadrupole analyzer (TQ LC–MS/MS) quantify low concentrations validated precursor targeting DAT (LBT-999) dissected...
Microbubble (MB)-assisted ultrasound (US) is an innovative modality for the non-invasive, targeted, and efficient delivery of therapeutic molecules into brain. Previously, we reported first metabolomic signature blood-brain barrier opening (BBBO) induced by MB-assisted US. In present study, neurometabolic consequences acoustically-mediated BBBO on cerebral tissue were investigated using multimodal metabolomics approaches. Sinusoid US waves (1 MHz, peak negative pressure 0.6 MPa, burst length...
Microbubble (MB)-assisted ultrasound (US) is a promising physical method to increase non-invasively, transiently, and precisely the permeability of blood-brain barrier (BBB) therapeutic molecules. Previous preclinical studies established innocuity this procedure using complementary analytical strategies including transcriptomics, histology, brain imaging, behavioral tests. This cross-sectional study rats aimed investigate metabolic processes following acoustically-mediated BBB opening in...
Abstract Phospholipids and cholesterol combine with a protein fraction (IgA an acid polypeptide) in bile to form the lipoprotein complex. We wished determine whether lysosomes participated only IgA secretion or if their secretory role also involved lipid components of This aspect was studied single acute injection chloroquine, lysosomotropic drug. The results show that nonnegligible quantity travels through lysosomes. In addition, phospholipid levels undergo significant (P<0.05) decrease...
Studying the metabolome of specific gestational compartments is growing interest in context fetus developmental disorders. However, metabolomes placenta and amniotic fluid (AF) are poorly characterized. Therefore, we present validation a fingerprinting methodology. Using pregnant rats, performed exhaustive robust extractions metabolites AF lipids more polar placenta. For AF, compared extraction capabilities methanol (MeOH), acetonitrile (ACN), mixture both. placenta, (i) dichloromethane,...
Objective Over the past years, interest in role of gut microbiota neurodegenerative diseases has emerged. Despite numerous publications over decade, both human and pre-clinical studies, there is no clear consensus on microbiota's or involvement ALS. Few studies mouse models ALS highlighted a correlation between specific bacteria species prognostic severity disease. Still these results lack reproducibility remain controverted. In this article we present study fecal SOD1G93A model associated...
