Further insights on the secondary metabolites of a soft coral-derived fungus Aspergillus versicolor under guidance MS/MS-based molecular networking led to isolation seven known cycloheptapeptides, namely, asperversiamides A-C (1-3) and asperheptatides A-D (4-7) an unusual pyrroloindoline-containing new cycloheptapeptide, asperpyrroindotide A (8). The structure 8 was elucidated by comprehensive spectroscopic data analysis, its absolute configuration determined advanced Marfey's method....
Three new phenolic bisabolane-type sesquiterpenoids: (+)-methyl sydowate (1), 7-deoxy-7,14-didehydrosydonic acid (2), and 7-deoxy-7,8-didehydrosydonic (3), together with two known fungal metabolites were isolated from the fermentation broth of a marine-derived fungus Aspergillus sp., which was in turn gorgonian Dichotella gemmacea collected South China Sea. Their structures elucidated by combined spectroscopic methods, structure 1 further confirmed single-crystal X-ray data.
A pair of new enantiomeric alkaloid dimers, (+)- and (−)-pestaloxazine (1), with an unprecedented symmetric spiro[oxazinane-piperazinedione] skeleton, consisting 22 carbons 12 heteroatoms, were isolated from a Pestalotiopsis sp. fungus derived soft coral. Separation the dimers was achieved by chiral HPLC. Their structures including absolute configurations elucidated on basis comprehensive analysis their spectroscopic X-ray diffraction data CD calculations. (+)-Pestaloxazine exhibited potent...
Chemical investigation of a marine-derived fungus Nigrospora sp., isolated from an unidentified sea anemone, yielded two new hydroanthraquinone analogues, 4a-epi-9α-methoxydihydrodeoxybostrycin (1) and 10-deoxybostrycin (2), together with seven known anthraquinone derivatives (3–9). The structures the compounds were established through extensive NMR spectroscopy as well single-crystal X-ray diffraction analysis using Cu Kα radiation. antibacterial activities 1–9 10 acetyl (6a, 7a, 8a–8g, 9a)...
Non-alcoholic steatohepatitis (NASH) is a primary cause of cirrhosis and hepatocellular carcinoma. Unfortunately, there no approved drug treatment for NASH. AMP-activated kinase (AMPK) an important metabolic sensor whole-body regulator. It has been proposed that AMPK activators could be used treating diseases such as obesity, type 2 diabetes In this study, we screened marine natural compound library by monitoring activity found potent activator, candidusin A (CHNQD-0803). Further studies...
A new antibacterial chlorinated benzophenone derivative, (±)-pestalachloride D (1), along with a related analog, C (2), was recently isolated from the marine-derived fungus Pestalotiopsis sp. soft coral Sarcophyton collected Yongxing Island in South China Sea. Both chiral HPLC analysis and single-crystal X-ray data indicated that 1 is racemic mixture. Interestingly, did not exhibit any effect zebrafish embryo teratogenicity assay, while 2 led to abnormal growth. The potential impact on...
Marine natural products play critical roles in the chemical defense of many marine organisms and are essential, reputable sources successful drug leads. Sixty-seven 14-membered resorcylic acid lactone derivatives 3-27 30-71 product zeaenol (1) isolated from marine-derived fungus Cochliobolus lunatus were semisynthesized by chlorination, acylation, esterification, acetalization one to three steps. The structures these new established HRESIMS NMR techniques. All compounds (1-71) evaluated for...
Abstract Natural products are well established as an important resource and play role in drug discovery. Here, two pyrrolinone-fused benzoazepine alkaloids, (+)-asperazepanones A ( 1 ) B 2 with a 6/7/5 ring system, together the artifact (−)-asperazepanone ), were isolated from coral-derived Aspergillus candidus fungus. Their structures including absolute configurations elucidated by extensive spectroscopic methods, single crystal X-ray diffraction, ECD calculations. Furthermore, total...
Under the guidance of MS/MS-based molecular networking, four new cycloheptapeptides, namely, asperheptatides A–D (1–4), were isolated together with three known analogues, asperversiamide A–C (5–7), from coral-derived fungus Aspergillus versicolor. The planar structures two major compounds, A and B (1 2), determined by comprehensive spectroscopic data analysis. absolute configurations amino acid residues advanced Marfey's method. structurally related trace metabolites, C D (3 4),...
Abstract Two new xanthones, 8‐(methoxycarbonyl)‐1‐hydroxy‐9‐oxo‐9H‐xanthene‐3‐carboxylic acid (1) and dimethyl 8‐methoxy‐9‐oxo‐9H‐xanthene‐1, 6‐dicarboxylate (2) one known xanthone methyl 8‐hydroxy‐6‐methyl‐9‐oxo‐9H‐xanthene‐1‐carboxylate (3) were isolated from the culture broth of mangrove fungus Penicillium sp. (ZZF 32#) collected South China Sea. Their structures established by comprehensive analysis one‐dimensional (1D) two‐dimensional (2D) NMR data. The structure compound 3 was...
Fumiquinazoline L (1), an alkaloid with a heptacyclic skeleton formed via bridging hemiaminal linkage, was isolated from gorgonian-derived Scopulariopsis sp. fungus. The structure and absolute configuration of the new compound were identified by comprehensive spectroscopic data X-ray diffraction analysis. During acid hydrolysis 1, isomerization valine residue observed also studied in different conditions. (1) showed no cytotoxic or antibacterial activities.
Pestalachlorides E and F containing both point axial chirality were isolated from a marine-derived Pestalotiopsis (ZJ-2009-7-6) fungus.
With the emergence of drug-resistant strains, treatment tuberculosis (TB) is becoming more difficult and there an urgent need to find new anti-TB drugs. Mycobacterium marinum, as a model organism tuberculosis, can be used for rapid efficient screening bioactive compounds. The 14-membered resorcylic acid lactones (RALs) have wide range bioactivities such antibacterial, antifouling antimalarial activity. In order further study their bioactivities, we initially constructed RALs library, which...
Tuberculosis (TB), caused by the bacterium Mycobacterium tuberculosis (Mtb), is still a leading cause of mortality worldwide. Fifty-fungi from marine-derived fungal library were screened for anti-Mtb activity, and an Aspergillus candidus strain with strong activity was found.
