Recently, two novel concepts have emerged in cancer biology: the role of so-called "cancer stem cells" tumor initiation, and involvement an epithelial-mesenchymal transition (EMT) metastatic dissemination epithelial cells. Using a mammary progression model, we show that cells possessing both tumorigenic characteristics can be derived from human following activation Ras-MAPK pathway. The acquisition these characters is driven by EMT induction.
The LAZ3/BCL6 (lymphoma-associated zinc finger 3/B cell lymphomas 6) gene frequently is altered in non-Hodgkin lymphomas. It encodes a sequence-specific DNA binding transcriptional repressor that contains conserved N-terminal domain, termed BTB/POZ (bric-à-brac tramtrack broad complex/pox viruses and fingers). Using yeast two-hybrid screen, we show here the domain interacts with SMRT (silencing mediator of retinoid thyroid receptor) protein. originally was identified as corepressor...
The import of many proteins into the nucleus is mediated by importin-alpha/beta heterodimer. While only one importin-beta gene has been found, several forms importin-alpha have described. In addition to three human importin-alphas already identified, we report here primary structure two new proteins. five known subunits can be classified subfamilies that appear conserved in higher eukaryotic organisms. We show immunoblotting different are expressed a variety tissues and mammalian cell lines.
Phosphorylation of transcription factors is regarded as a major mechanism to control their activity in regulation gene expression. C/EBP beta factor that becomes activated after phosphorylation induce genes involved inflammation, acute-phase response, cytokine expression, cell growth, and differentiation. The chicken homolog NF-M collaborates with Myb various kinase oncogenes normal myeloid differentiation well the leukemic transformation myelomonocytic cells. Here, we examined structure its...
The epithelial-mesenchymal transition (EMT) is an embryonic transdifferentiation process consisting of conversion polarized epithelial cells to motile mesenchymal ones. EMT–inducing transcription factors are aberrantly expressed in multiple tumor types and known favor the metastatic dissemination process. Supporting oncogenic activity within primary lesions, TWIST ZEB proteins can prevent from undergoing oncogene-induced senescence apoptosis by abolishing both p53- RB-dependent pathways....
Abstract A cDNA clone encoding the complete sequence of an active, rat choline acetyltransferase (ChoAcTase; acetyl‐CoA:choline O‐acetyltransferase, EC 2.3.1.6) has been isolated. Analysis deduced amino acid reveals 85% and 31% identity with porcine Drosophila melanogaster enzymes, respectively. To further elucidate molecular basis neurotransmitter‐related phenotypic plasticity, expression ChoAcTase mRNA was compared that tyrosine hydroxylase [TH; 3‐monooxygenase, L‐tyrosine,...

The transcription factor TWIST1 induces epithelial-mesenchymal transition and/or escape to the oncogenic-induced failsafe program, facilitating intravasation of breast cancer cells in systemic circulation and their dissemination lungs. Its involvement bone metastasis is unknown. To address this question, human osteotropic MDA-MB-231/B02 were stably transfected with a Tet-inducible vector encoding for TWIST1, whose expression was specifically repressed presence doxycycline (dox)....
Metastasis is a multistep process and the main cause of mortality in lung cancer patients. We previously showed that EGFR mutations were associated with copy number gain at locus encompassing TWIST1 gene on chromosome 7. highly conserved developmental involved embryogenesis may be reactivated cancers promoting both malignant conversion progression through an epithelial to mesenchymal transition (EMT). The aim this study was investigate possible implication reactivation acquisition phenotype...
BS69 is a transcriptional co-repressor protein and potential tumor suppressor that binds to the adenoviral oncoprotein E1A. We show C-terminal Mynd domain of (amino acids 516–561) or closely related domains theCaenorhabditis elegans proteins Bra-1 Bra-2 bind not only E1A but also Epstein-Barr virus EBNA2 Myc-related cellular MGA. Interaction depends on intact PXLXP motifs present in all three proteins. Moreover, viral compete for binding MGA PXLXP-dependent fashion. Because deletions cover...
By fostering cell commitment to the epithelial-to-mesenchymal transition (EMT), SNAIL proteins endow cells with motility, thereby favoring metastatic spread of tumor cells. Whether phenotypic change additionally facilitates initiation has never been addressed. Here we demonstrate that when a protein is ectopically produced in non-transformed mammary epithelial cells, are protected from anoikis and proliferate under low-adherence conditions: hallmark cancer The three show unequal oncogenic...
Tumor-initiating cells (TICs), aka "cancer stem cells", are believed to fuel tumors and sustain therapy resistance systemic metastasis. Breast cancer is the first human carcinoma in which a subpopulation of displaying specific CD44+/CD24-/low/ESA+ antigenic phenotype was found have TIC properties. However, not universal marker TICs all breast subtypes. The aim this study identify novel antigens with isolate population basal-A/basal-like cell lines. We used polychromatic flow-cytometry...
