Understanding the immune responses elicited by SARS-CoV-2 infection is critical in terms of protection against reinfection and, thus, for public health policy and vaccine development COVID-19. In this study, using either live particles or retroviruses pseudotyped with S viral surface protein (Spike), we studied neutralizing antibody (nAb) response serum samples from a cohort 140 qPCR-confirmed infections, including patients mild symptoms also more severe forms, those that required intensive...
The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), a β-coronavirus, is the causative agent of COVID-19 pandemic. Like for other coronaviruses, its particles are composed four structural proteins: spike (S), envelope (E), membrane (M), and nucleoprotein (N) proteins. involvement each these proteins their interactions critical assembly production β-coronavirus particles. Here, we sought to characterize interplay SARS-CoV-2 during viral process. By combining biochemical imaging...
Following severe adverse reactions to the AstraZeneca ChAdOx1-S-nCoV-19 vaccine1,2, European health authorities recommended that patients under age of 55 years who received one dose receive a second Pfizer BNT162b2 vaccine as booster. However, effectiveness and immunogenicity this vaccination regimen have not been formally tested. Here we show heterologous combination confers better protection against acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection than homologous in...
In a survey of household cats and dogs laboratory-confirmed COVID-19 patients, we found high seroprevalence SARS-CoV-2 antibodies, ranging from 21% to 53%, depending on the positivity criteria chosen. Seropositivity was significantly greater among pets COVID-19+ households compared those with owners unknown status. Our results highlight potential role in spread epidemic.
Abstract Dengue fever represents a significant medical and socio-economic burden in (sub)tropical regions, yet antivirals for treatment or prophylaxis are lacking. JNJ-A07 was described as highly active against the different genotypes within each serotype of disease-causing dengue virus (DENV). Based on clustering resistance mutations it has been assumed to target DENV non-structural protein 4B (NS4B). Using photoaffinity labeling compound with high structural similarity JNJ-A07, here we...
Crimean-Congo hemorrhagic fever virus (CCHFV) is a tick-borne orthonairovirus that has become serious threat to the public health. CCHFV single-stranded, tripartite RNA genome composed of L, M, and S segments. Cleavage M polyprotein precursor generates two envelope glycoproteins (GPs) as well three secreted nonstructural proteins GP38 GP85 or GP160, representing only linked mucin-like protein (MLD), double-membrane-spanning called NSm. Here, we examined relevance each M-segment...
Abstract Zika virus (ZIKV) has emerged as a global health issue, yet neither antiviral therapy nor vaccine are available. ZIKV is an enveloped RNA virus, replicating in the cytoplasm close association with ER membranes. Here, we isolate membranes from ZIKV-infected cells and determine their proteome. Forty-six host cell factors enriched remodeled membranes, several of these having role redox methylation pathways. Four proteins characterized detail: thioredoxin reductase 1 (TXNRD1)...
Abstract The Crimean-Congo hemorrhagic fever virus (CCHFV) is an emerging pathogen of the Orthonairovirus genus that can cause severe and often lethal diseases in humans. CCHFV has a broad tropism infect variety species tissues. Here, by using gene silencing, blocking antibodies or soluble receptor fragments, we identify low-density lipoprotein (LDL-R) as entry factor. LDL-R facilitates binding particles but does not allow Hazara (HAZV), another member genus. In addition, show apolipoprotein...
Dengue virus (DENV) is a major human pathogen that can cause hemorrhagic fever and shock syndrome. One important factor of DENV pathogenicity non-structural protein 1 (NS1), glycoprotein secreted from infected cells. Here we study the mode action widely used drug ivermectin, to treat parasitic infections recently shown lower NS1 blood levels in DENV-infected patients. We found ivermectin blocks nuclear transport transcription factors required for expression chaperones support folding...
For successful infection of host cells and virion production, enveloped viruses, including Zika virus (ZIKV), extensively rely on cellular lipids. However, how protein-lipid interactions contribute to the viral life cycle remains unclear. Here, we employ a chemo-proteomics approach with bifunctional cholesterol probe show that is closely associated ZIKV structural protein prM. Bioinformatic analyses, reverse genetics alongside photoaffinity labeling assays, atomistic molecular dynamics...

Despite the probable zoonotic origin of SARS-CoV-2, only limited research efforts have been made to understand role companion animals in SARS-CoV-2 epidemiology. According recent serological prevalence studies, human-to-companion animal transmission is quite frequent, which led us consider that risk from human, albeit negligible present context, may underestimated. In this study, we provide results a prospective survey was conducted evaluate isolation rate by qRT-PCR dogs and cats with...

Viroporins are small transmembrane proteins with ion channel activities modulating properties of intracellular membranes that have diverse proviral functions. Hepatitis C virus (HCV) encodes a viroporin, p7, acting during assembly, envelopment and secretion viral particles (VP). HCV p7 is released from the polyprotein through cleavage at E2-p7 p7-NS2 junctions by signal peptidase, but also exists as an E2p7 precursor, poorly defined properties. Here, we found ectopic expression in...
Abstract Following severe adverse reactions in patients vaccinated with the AstraZeneca ChadOx1 (Chad) vaccine, European health authorities have recommended that under age of 55 who received one dose Chad vaccine receive a second Pfizer BNT162b2 (BNT) as booster. However, effectiveness and immunogenicity this vaccination regimen not been formally tested. Here, we show heterologous Chad/BNT combination confers better protection against SARS-CoV-2 infection than homologous BNT/BNT population...
Abstract Although there are several reports in the literature of SARS‐CoV‐2 infection cats, few sequences from infected cats have been published. In this study, was evaluated two by clinical observation, molecular biology (qPCR and NGS), serology (microsphere immunoassay seroneutralization). Following observation symptomatic status confirmed RT‐qPCR and, one cat, serological analysis for antibodies against N‐protein S‐protein, as well neutralizing antibodies. Comparative five sequence...
Nup98, an essential component of the nuclear pore that also participates in annulate lamella structures localized cytosol, is involved hepatitis C virus (HCV) assembly. Here, we combined confocal microscopy and biochemical assays to study interplay between core (i.e., HCV capsid protein), viral genomes. Our results show HCV-infected cells, protein necessary sufficient induce relocalization Nup98 from lamellae lipid droplet-apposed areas which core/NS5A genomic RNA [(+)RNA] are clustered...
