In antigen (Ag) cross-presentation, dendritic cells (DCs) take up extracellular Ag and translocate them from the endosome to cytosol for proteasomal degradation. The processed peptides can enter conventional MHC I pathway. molecules responsible translocation of across endosomal membrane into are unknown. Here we demonstrate that heat shock protein 90 (HSP90) is critical this step. Cross-presentation -priming were decreased in both HSP90α-null DCs mice. CD8α(+) DC apoptosis mediated by...
Abstract In Legionella pneumophila infection, macrophages play a critical role in the host defense response. Metformin, an oral drug for type 2 diabetes, is attracting attention as new supportive therapy against variety of diseases, such cancer and infectious diseases. The novel mechanisms metformin actions include modulation effector functions other immune cells. this study, we have examined effects on L. infection vitro vivo. Metformin treatment suppressed growth time-...
Abstract Extracellular HSP90 associated with Ag peptides have been demonstrated to efficiently cross-prime T cells, following internalization by dendritic cells (DCs). In addition, the nature of cell-associated Ags required for cross-priming is implicated as and proteins chaperoned heat shock protein (HSP). However, role endogenous HSP in DCs during cross-presentation remains elusive. this paper, we show that essential both soluble DCs. Cross-presentation OVA OVA-loaded transporter...
Nontuberculous mycobacteria (NTM) are ubiquitous and can be inhaled from household water sources, posing a potential risk of pulmonary NTM disease. Exposure to 222-nm far ultraviolet-C (Far UV-C) shows comparable inactivation efficiency 254-nm (UV-C), while employing specialized optical filters significantly enhance safety for human skin eyes. Far UV-C radiation demonstrated an irradiance-dependent inactivating effect, highlighting its as safe effective disinfection.
In Japan, Staphylococcal cassette chromosome mec (SCC ) type IV methicillin-resistant Staphylococcus aureus (MRSA) is an increasingly prominent cause of bacteremia, but the virulence most these strains unclear. We aimed to investigate relationship between molecular characteristics and ability form biofilms in presence blood plasma (plasma-biofilms) MRSA isolated from bloodstream infections. this study, cultures 2015 2017 were analyzed by PCR-based assays, crystal violet staining, confocal...
Non-tuberculous mycobacteria infections, including Mycobacterium avium, are increasingly recognized as a growing public health concern, even among immunocompetent individuals. These infections significant cause of chronic pulmonary disease, and they characterized by the formation foamy macrophages (FMs) that facilitate bacterial persistence. Previously, we reported apoptosis inhibitor macrophage (AIM), protein secreted macrophages, promotes lipid droplet accumulation in M. avium-infected...
Vaccination with heat shock proteins (HSP) protects mice from challenge the tumor which HSP were isolated. The antigenicity of vaccination is thought to result HSP‐associated endogenous major histocompatibility complex class I peptides or their precursors. effect can be achieved in an adjuvant‐free manner and mediated by CD8 + T cells, indicating that act as a natural adjuvant cross‐prime cells vivo . We previously devised recombinant vaccine composed cell epitope fused carboxyl‐terminus...
Clostridium difficile infection (CDI) is a toxin-mediated intestinal disease. Toxin A, toxin B and binary are believed to be responsible for the pathogenesis of CDI, which characterized by massive infiltration neutrophils at infected mucosa. IL-17 one cytokines that play critical roles in several inflammatory immunological diseases through various actions, including promoting neutrophil recruitment. The aim this study was examine role cytokine CDI employing A F double knockout (IL-17 KO)...
Community-acquired pneumonia is a common disease with considerable morbidity and mortality, for which Streptococcus pneumoniae accepted as leading cause. Although β-lactam-plus-macrolide combination therapy this recommended in several guidelines, the clinical efficacy of strategy against pneumococcal remains controversial. In study, we examined effects on lethal mouse explored mechanisms action vitro vivo We investigated survival, lung bacterial burden, cellular host responses...
Antigens (Ag) from cancer or virus-infected cells must be internalized by dendritic (DCs) to presented CD8 + T cells, which eventually differentiate into Ag-specific cytotoxic lymphocytes (CTLs) that destroy and infected cells. This pathway is termed cross-presentation also implicated as an essential step in triggering autoimmune diseases such Type I diabetes. Internalized Ag locates within endosomes, followed translocation through a putative pore structure spanning endosomal membranes the...
It is controversial whether a functional androgen receptor (AR) on germ cells, including spermatogonia, essential for their development into sperm and, thus, initiation and maintenance of spermatogenesis. was recently shown that many spermatocytes underwent apoptosis in the testes Hsp90α KO mice. We had generated mice independently confirmed this phenotype. However, important question required to maintain spermatogenesis adult which testicular maturation already completed could not be...
Abstract In Mycobacterium avium infections, macrophages play a critical role in the host defense response. Apoptosis inhibitor of macrophage (AIM), also known as CD5L, may represent novel supportive therapy against various diseases, including metabolic syndrome and infectious diseases. The mechanisms AIM include modulating lipid metabolism other cells. We investigated M. infections vitro vivo. mouse model pneumonia, foamy were induced 6 wk after infection. bacteria localized these...
In this study, we analyzed interferon (IFN)-γ-producing cells and M1/M2 macrophage polarization in <i>Legionella pneumophila</i> pneumonia following anti-Gr-1 antibody treatment. Anti-Gr-1 treatment induced an M1-to-M2 shift of subtypes the lungs weakly peripheral blood, which was associated with increased mortality legionella-infected mice. CD8+ T lymphocytes natural killer were dominant sources IFN-γ acute phase, reduced number IFN-γ-producing...
Blood pressure (BP) increase as a reaction to major disasters has been well documented; however, the impact underdocumented for tsunamis. This study aimed confirm whether different levels of flooding/inundation and other damage caused by 2011 Tohoku (northeast Japan) tsunami were associated with BP among resident victims in Higashi-Matsushima, Miyagi. Cross-sectional household screening was conducted 7–19 weeks after disaster administrative areas totally or partially flooded tsunami....
Mycobacterium avium complex is a causative organism for refractory diseases. In this study, we examined the effects of N-acetyl-cysteine on M. infection in vitro and vivo. treatment suppressed growth A549 cells concentration-dependent manner. This effect was related to induction antibacterial peptide human β-defensin-2. mouse model, significantly reduced number bacteria lungs induced murine β-defensin-3. interleukin-17-deficient mice, disappeared, indicating that these mechanisms may be...
Priming of CD8(+) T cells requires two signals, one produced by T-cell receptor recognition antigen, and a second that is often provided the innate immune response. In this context, antigens non-covalently or covalently associated with heat shock proteins (HSP) are internalized processed in antigen-presenting (APC) to be presented MHC I molecules cells, thus, signal 1 has been well characterized pathway cross-presentation. Signal 2 not fully understood, although there reports Toll-like...
Abstract Heat shock protein 90 (HSP90) is a molecular chaperone required for efficient antigen presentation and cross-presentation. In addition, HSP90 was recently reported to interact with stabilize the activation-induced cytidine deaminase (AID) plays critical role in immunoglobulin gene hypermutation class switch recombination. mice humans, there are two isoforms, HSP90α HSP90β, but vivo of each isoform remains largely unknown. Here we have analyzed humoral immune responses...
