Metazoan E(y)2/ENY2 is a multifunctional protein important for transcription activation and mRNA export, being component of SAGA/TFTC the export complex AMEX. Here, we show that ENY2 in Drosophila also stably associated with THO, involved mRNP biogenesis. The ENY2–THO required normal development, functioning independently on SAGA THO arrive transcribed region hsp70 gene after its activation, plays an role recruitment. no direct association elongating RNA polymerase II. Despite increasing data on the properties of replication origins, molecular mechanisms underlying origin recognition complex (ORC) positioning in genome are still poorly understood. The Su(Hw) protein accounts for activity best-studied Drosophila insulators. Here, we show that recruits histone acetyltransferase SAGA and chromatin remodeler Brahma to Su(Hw)-dependent insulators, which gives rise regions with low nucleosome density creates conditions ORC binding. We have isolated a novel Drosophila (d) gene coding for two distinct proteins via alternative splicing: homologue of the yeast adaptor protein ADA2, dADA2a, and subunit RNA polymerase II (Pol II), dRPB4. Moreover, we identified another in genome encoding second ADA2 (dADA2b). The dADA2 homologues, as well many putative homologues from different species, all contain, addition to ZZ SANT domains, several evolutionarily conserved domains. dada2a/rpb4 dada2b genes are differentially expressed at...

Transcription activation by RNA polymerase II is a complicated process driven combined, precisely coordinated action of wide array coactivator complexes, which carry out chromatin-directed activities and nucleate the assembly preinitiation complex on promoter. Using various techniques, we have shown existence stable supercomplex consisting chromatin-remodeling factor Brahma (SWI/SNF) transcription initiation TFIID, named BTFly (Brahma TFIID in one assembly). Drosophila SAYP, a homologue of human PHF10/BAF45a, is metazoan coactivator associated with Brahma and essential for its recruitment on the promoter. The role SAYP in DHR3 activator-driven transcription ftz-f1 gene, member ecdysone cascade was studied. In repressed state presence DHR3, Pol II complex pre-recruited promoter; starts but paused 1.5 kb downstream promoter, forming 'nucleosomal barrier' (a region high nucleosome density) ahead II. depletion leads to removal Brahma, thereby...

The PBAF subtype of the mammalian chromatin remodeling SWI/SNF complex has wide and diverse functions in transcription regulation development, being both activator repressor. However, a mechanism accounting for such functional diversity remains unclear. Human PHF10/BAF45a subunit plays an important role brain development but not been studied sufficiently. We have shown that PHF10 gene encodes 2 types evolutionarily conserved, ubiquitously expressed isoforms are incorporated into mutually...

SAGA/TFTC is a histone acetyltransferase complex that has second enzymatic activity because of the presence deubiquitination module (DUBm). Drosophila DUBm consists Sgf11, ENY2 and Nonstop proteins. We show Sgf11 other DUBm-independent functions. It associates with Cbp80 component cap-binding thereby recruited onto growing messenger ribonucleic acid (mRNA); it also interacts AMEX mRNA export essential for hsp70 export, as well general from nucleus. Thus, functions both SAGA biogenesis machinery.
