The 21-subunit Mediator complex transduces regulatory information from enhancers to promoters, and performs an essential role in the initiation of transcription all eukaryotes. Structural on two-thirds has been limited coarse subunit mapping onto 2-D images electron micrographs. We have performed chemical cross-linking mass spectrometry, combined results with X-ray crystallography, homology modeling, cryo-electron microscopy by integrative modeling approach determine a 3-D model entire...

Abstract Building models of a biological system that are consistent with the myriad data available is one key challenges in biology. Modeling structure and dynamics macromolecular assemblies, for example, can give insights into how systems work, evolved, might be controlled, even designed. Integrative modeling casts building structural as computational optimization problem, which information about assembly encoded scoring function evaluates candidate models. Here, we describe our open source...

The mechanism for hemorrhage enlargement in the brain, a key determinant of patient outcome following hemorrhagic stroke, is unknown. We performed computer-based stochastic simulation one proposed mechanism, which hemorrhages grow "domino" fashion via secondary shearing neighboring vessel segments. Hemorrhages were simulated by creating an initial site primary bleeding and associated risk rupture at adjacent sites that decayed over time. Under particular combinations parameters likelihood...

We have measured the neutron energy spectrum of an 241 Am-Be(α,n) source between 1.5 MeV and 9 using a scatter camera. The apparatus consists two segmented planes each with 16 liquid scintillator cells (Eljen EJ-309), for total 32 elements; is double elastic events. After unfolding resolution effects maximum likelihood technique, measurement compared to reference Am-Be spectra. Further, we...

Summary Cryo-electron microscopy (cryo-EM) has become a mainstream technique for determining the structures of complex biological systems. However, accurate integrative structural modeling been hampered by challenges in objectively weighing cryo-EM data against other sources information due to presence random and systematic errors, as well correlations, data. To address these challenges, we introduce Bayesian scoring function that efficiently accurately ranks alternative models...

A Monte Carlo model has been developed for the Sandia Neutron Scatter Camera. We have used this to optimize cell geometry and detector efficiency. The simulations improve understand response calibrations.

We have successfully ruggedized and deployed a field portable fast-neutron imager. Detection efficiency has been increased by > 5x more over the one fielded last year. Imaging demonstrated though 30 cm thick concrete at stand-off distance of 75 m in air. also imaged pulsed D-D neutron generator recorded its energy spectrum. These demonstrations show that our scatter camera could be useful tool not only lab but as detector for hidden SNM.
