SREBP2 is a master regulator of the mevalonate pathway (MVP), biosynthetic process that drives synthesis dolichol, heme A, ubiquinone and cholesterol also provides substrates for protein prenylation. Here, we identify as novel substrate USP28, deubiquitinating enzyme frequently upregulated in squamous cancers. Our results show silencing USP28 reduces expression MVP enzymes lowers metabolic flux into this pathway. We binds to mature SREBP2, leading its deubiquitination stabilisation....

Dedifferentiated liposarcoma (DDLPS) is a rare mesenchymal cancer originating from the adipose tissue, with poor survival rates for most patients, highlighting critical need novel treatment options. This report examines efficacy and safety of sequential pre-treatment marine-derived alkaloid trabectedin followed by checkpoint inhibition using anti-PD-1 antibody nivolumab in 63-year-old male patient unresectable retroperitoneal DDLPS. Treatment was initiated at time seventh relapse as part...

Abstract Postoperative disease recurrence in Crohn’s represents a relevant issue despite recent advancements surgical and medical therapies. Additional criteria are necessary to improve the identification of patients at risk enable selective therapeutic approaches. The role resection margins on remains unclear general recommendations lacking. A single-center retrospective analysis was performed including all who received ileocecal due disease. Resection were analyzed by two independent...

BACKGROUNDAntibiotic-Refractory Lyme Arthritis (ARLA) involves a complex interplay of T cell responses targeting Borrelia burgdorferi antigens progressing toward autoantigens by epitope spreading. However, the precise molecular mechanisms driving pathogenic response in ARLA remain unclear. Our aim was to elucidate program disease-specific Th cells.METHODSUsing flow cytometry, high-throughput receptor (TCR) sequencing, and scRNA-Seq CD4+ cells isolated from joints patients with living Europe,...

Abstract The two p53 homologues p63 and p73 regulate transcriptional programs in epithelial tissues several cell types these express both proteins. All members of the family form tetramers their active state through a dedicated oligomerization domain that structurally assembles as dimer dimers. share high sequence identity, but is more divergent it lacks functionally important C-terminal helix present other members. Based on structural differences, does not hetero-oligomerize with or p73. In...

Abstract The contribution of deubiquitylating enzymes (DUBs) to β-Catenin stabilization in intestinal stem cells and colorectal cancer (CRC) is poorly understood. Here, by using an unbiassed screen, we discovered that the DUB USP10 stabilizes specifically APC-truncated CRC vitro vivo. Mechanistic studies, including binding together with computational modelling, revealed mediated via unstructured N-terminus outcompeted intact APC, favouring β-catenin degradation. However, binds β-catenin,...

Abstract Molecular understanding of osteogenic differentiation (OD) human bone marrow-derived mesenchymal stem cells (hBMSCs) is important for regenerative medicine and has direct implications cancer. We report that the RNF4 ubiquitin ligase essential OD hBMSCs, RNF4-deficient hBMSCs remain as stalled progenitors. Remarkably, incubation in conditioned media differentiating restored OD. Transcriptional analysis RNF4-dependent gene signatures identified two secreted factors act downstream...
