Therapeutics targeting osteoclasts are commonly used treatments for bone metastasis; however, whether and how regulate premetastatic niche tropism largely unknown. In this study, we report that osteoclast precursors (OPs) can function as a component facilitates breast cancer (BCa) metastasis at early stages. At the molecular level, unbiased GPCR ligand/agonist screening in BCa cells suggested R-spondin 2 (RSPO2) RANKL, through interaction with their receptor LGR4, promoted osteoclastic...

Abstract Inflammatory diseases decrease the extracellular environmental pH. However, whether proton-activated G protein-coupled receptors (GPCRs) can regulate development of osteoarthritis (OA) is largely unknown. In this study, we report that GPR4 essential for OA development. We found a marked increase in expression human and mouse cartilage. Lentivirus-mediated overexpression joints accelerated OA, including promotion articular cartilage damage, synovial hyperplasia, osteophyte formation,...

Hypomanic personality traits (HPT) are susceptibility markers for psychiatric disorders, particularly bipolar disorder, and strongly associated with aggressive behaviors. However, the neuropsychological mechanisms underlying this association remain unclear. This study utilized psychometric network analysis Inter-Subject Representation Similarity Analysis (IS-RSA) to explore circuits that link HPT aggression in a large non-clinical population. Psychometric (n = 716) identified two key nodes:...

Ginsenoside Rb1, a major component of different ginseng species, can be bioconverted into compound K by gut microbiota, and the latter possess much stronger cancer chemopreventive potential. However, while initiation progression colorectal is closely associated with inflammation, to date, effects on inflammation-linked chemoprevention have not been reported. In present study, liquid chromatography quadrupole time-of-flight mass spectrometry analysis was applied evaluate biotransformation Rb1...

Andrographolide has a low aqueous solubility and oral bioavailability, which limits its clinical application. Reform the dosage forms of andrographolide to improve bioavailability. The formulation, characterisation, stability, anti-inflammatory effect, pharmacokinetics toxicity andrographolide-loaded microemulsion, were studied. An formulation O/W microemulsion consisting an oil phase isopropyl myristate, surfactant Tween 80, co-surfactant alcohol, water was found be ideal, with mean droplet...

Osteoarthritis (OA) is one of the most common chronic diseases in world. However, current treatment modalities mainly relieve pain and inhibit cartilage degradation, but do not promote regeneration. In this study, we show that G protein-coupled receptor class C group 5 member B (GPRC5B), an orphan G-protein-couple receptor, only inhibits also increases regeneration thereby protective against OA. We observed Gprc5b deficient chondrocytes had upregulation catabolic gene expression, along with...

In this experiment, we established an animal model of gestational diabetes mellitus rats using streptozotocin. Using the rat GDM, pregnant in 1-19d were divided into three groups: (1) Zuogui Wan group (group I, n = 12), (2) as control II, 11), and (3) normal pregnancy III, 11). Compared with group, can change indexes fasting blood glucose, body weight, total cholesterol, insulin, metabolism cage index significantly group. We conclude that has therapeutic effect on mellitus.
