Abstract The selection of DNA‐encoded libraries against biological targets has become an important discovery method in chemical biology and drug discovery, but the requirement modified immobilized remains a significant disadvantage. With terminal protection strategy ligand‐induced photo‐crosslinking, we show that iterated selections can be realized with unmodified non‐immobilized protein targets.
Mutations in leucine-rich repeat kinase 2 (LRRK2) are the most frequent known cause of late-onset Parkinson's disease (PD). To explore therapeutic potential small molecules targeting LRRK2 domain, we characterized two inhibitors, TTT-3002 and LRRK2-IN1, for their effects against activity vitro Caenorhabditis elegans models LRRK2-linked neurodegeneration. LRRK2-IN1 potently inhibited wild-type mutant proteins, attenuated phosphorylation cellular rescued neurotoxicity transfected cells....
Hydrogen, as an emerging energy source, is extensively utilized in industrial production. Owing to its high reactivity, hydrogen presents explosion risk during utilization, storage, and production, potentially resulting significant harm personnel equipment. To elucidate the dynamic characteristics of external explosions induced by different ignition positions hydrogen-related plants, this study developed a 45 m3 explosion-venting model using computational fluid dynamics technology. A...
Ubiquinone (UQ) is a lipid co-factor that involved in numerous enzymatic processes and present most cellular membranes. In particular, UQ crucial electron carrier the mitochondrial respiratory chain. Recently, it was shown clk-1 mutants of nematode worm Caenorhabditis elegans do not synthesize UQ(9) but instead accumulate demethoxyubiquinone (DMQ(9)), biosynthetic precursor (the subscript refers to length isoprenoid side chain). DMQ(9) capable carrying out function chain, as demonstrated by...
Detonation development inside spark ignition engines can result in the so called super-knock with extremely high pressure oscillation above 200 atm. In this study, numerical simulations of autoignitive reaction front propagation hydrogen/air mixtures are conducted and detonation regime is investigated. A hot spot linear temperature distribution used to induce propagation. With change gradient or size, three typical autoignition modes identified: supersonic front; subsonic front. The effects...
ABSTRACT Myocilin-associated glaucoma is a protein-conformational disorder associated with formation of toxic amyloid-like aggregate. Numerous destabilizing single point variants, distributed across the myocilin olfactomedin β-propeller (OLF, residues 245-504, 30 kDa) are accelerated disease progression. In vitro , wild type (WT) OLF can be promoted to form thioflavin T (ThT)-positive fibrils under mildly (37°C, pH 7.2) conditions. Consistent notion that only small number within protein...

Abstract Objective To determine the prevalence of mitochondrial diseases (MD) in Hong Kong (HK) and to evaluate clinical characteristics genetic landscape MD patients region. Methods This study retrospectively reviewed phenotypic molecular from participating public hospitals HK between January 1985 October 2020. Molecularly and/or enzymatically confirmed cases any age were recruited via Clinical Analysis Reporting System (CDARS) using relevant keywords International Classification Disease...
Amyloid beta (Aβ) aggregation is a slow process without seeding or assisted nucleation. Sodium dodecyl sulfate (SDS) micelles stabilize Aβ42 small oligomers (in the dimer to tetramer range); subsequent SDS removal leads 150-kD oligomer. Dodecylphosphorylcholine (DPC) also an tetramer. Here we investigate detergent-assisted oligomerization pathway by solid-state NMR spectroscopy and molecular dynamics simulations. SDS- DPC-induced have same structure, implying common pathway. An antiparallel...
Abstract Molecular knowledge of human gastric corpus epithelium remains incomplete. Here, by integrated analyses using single-cell RNA sequencing (scRNA-seq), spatial transcriptomics and assay for transposase accessible chromatin (scATAC-seq) techniques, we uncovered the spatially-resolved expression landscape gene-regulatory network epithelium. Specifically, identified a stem/progenitor cell population in isthmus corpus, where EGF WNT signaling pathways were activated. Meanwhile, LGR4, but...
Abstract A large-size potassium-doped tungsten (KW) plate with a thickness of 15 mm was fabricated via powder metallurgy technology and hot rolling. In order to appraise the irradiation resistance KW, surface deuterium (D) blistering D retention were studied on Fe 11+ pre-damaged (0, 0.05 0.5 dpa) KW pure (PW), which exposed ∼60 eV ∼5 × 10 21 m −2 s −1 plasmas at 500 K fluence ∼5.76 25 . The results indicate that alloy can better inhibit generation vacancy defects after ion damage compared...
