We report the first fluorescent diamino-chalcone library and its application in discovery of a mouse embryonic stem cell (mESC) probe. CDg4, novel green mESC probe was discovered through high-content image based screening 160 members chalcone library. Interestingly, molecular binding target CDg4 identified as glycogen colony surface, rather than conventional protein from an intracellular source.
A better understanding of the cellular and molecular mechanisms involved in reprogramming somatic cells is essential for further improvement induced pluripotent stem (iPS) cell technology. In this study, we enriched actively undergoing at different time points by sorting stained with a cell-selective fluorescent chemical probe CDy1 their global gene expression analysis. Day-to-day comparison differentially expressed genes showed highly dynamic transient expressions during reprogramming,...
Two-photon (TP) microscopy has an advantage for live tissue imaging which allows a deeper penetration up to 1 mm comparing one-photon (OP) microscopy. While there are several OP fluorescence probes in use pancreatic islet imaging, TP of selective cells still remains challenge. Herein, we report the discovery first probe; TP-α (Two Photon-alpha) can selectively stain glucagon secreting alpha cells. Through fluorescent image based screening using three cell lines, discovered from dye library...
We report the first solid phase synthesis of a xanthone library CX and its application to embryonic stem cell probe development. The was further derivatised with an activated ester resin provide acetylated (CXAC) library. Screening these libraries led discovery novel fluorescent mESC probe, CDb8.
Lighting up the pancreas: A pancreatic beta cell selective probe PiY (red, see picture) was developed which stains Langerhans islets in live animals without toxicity or side effects. Through tail-vein injection, allowed for a comparison of between healthy and diabetic mice. also facilitated isolation by using fluorescence-guided surgical procedure.
A hallmark of Listeria (L . ) monocytogenes pathogenesis is bacterial escape from maturing entry vacuoles, which required for rapid replication in the host cell cytoplasm and cell-to-cell spread. The transcriptional activator PrfA controls expression key virulence factors that enable exploitation this intracellular niche. activity within infected cells controlled by allosteric coactivation. Inhibitory occupation coactivator site has been shown to impair functions, but consequences inhibition...
Stem cell research has gathered immense attention in the past decade due to remarkable ability of stem cells for self-renewal and tissue-specific differentiation. Despite having numerous advancements isolation manipulation techniques, there is a need highly reliable probes specific detection live cells. Herein we developed new fluorescence probe (CDy9) with high selectivity mouse embryonic CDy9 allows intact marginal impact on their function capabilities.
