The rice species Oryza sativa is considered to be a model plant because of its small genome size, extensive genetic map, relative ease transformation and synteny with other cereal crops1,2,3,4. Here we report the essentially complete sequence chromosome 1, longest in genome. We summarize characteristics structure biological insight gained from sequence. analysis 43.3 megabases (Mb) non-overlapping reveals 6,756 protein coding genes, which 3,161 show homology proteins Arabidopsis thaliana,...

To determine the chromosomal positions of expressed rice genes, we have performed an sequence tag (EST) mapping project by polymerase chain reaction–based yeast artificial chromosome (YAC) screening. Specific primers designed from 6713 unique EST sequences derived 19 cDNA libraries were screened on 4387 YAC clones and used for map construction in combination with genetic analysis. Here, describe establishment a comprehensive YAC-based transcript that contains 6591 sites covers 80.8% genome....

Endoxylanases are important enzymes in bioenergy research because they specifically hydrolyze xylan, the predominant polysaccharide hemicellulose fraction of lignocellulosic biomass. For effective biomass utilization, it is to understand mechanism substrate recognition by these enzymes. Recent studies have shown that specificities bacterial and fungal endoxylanases classified into glycoside hydrolase family 30 (GH30) were quite different. While functional differences been described, still...

Exo-1,5-α-L-arabinofuranosidases belonging to glycoside hydrolase family 43 have strict substrate specificity. These enzymes hydrolyze only the α-1,5-linkages of linear arabinan and arabino-oligosaccharides in an exo-acting manner. The enzyme from Streptomyces avermitilis contains a core catalytic domain C-terminal binding module carbohydrate 42. We determined crystal structure intact exo-1,5-α-L-arabinofuranosidase. is composed 5-bladed β-propeller topologically identical other enzymes. had...

The possibility of using two kinds sorghum as raw materials in consolidated bioprocessing bioethanol production Flammulina velutipes was investigated. Enzymatic saccharification sweet not high brown mid-rib (bmr) mutated sorghum, but the amount ethanol higher. Ethanol from bmr significantly increased when enzymes were added to culture.

Basidiomycetes have the ability to degrade lignocellulosic biomass, and some basidiomycetes produce alcohol dehydrogenase. These characteristics may be useful in direct production of ethanol from lignocellulose. Ethanol fermentation by was investigated examine possibility consolidated bioprocessing (CBP) using Flammulina velutipes. F. velutipes converted d-glucose with a high efficiency (a theoretical recovery rate 88%), but pentose not observed. properties are similar those Saccharomyces...

Plasmid pL32 from the Natto strain of Bacillus subtilis belongs to a group low-copy-number plasmids in gram-positive bacteria that replicate via theta mechanism replication. We studied DNA region encoding replication protein, RepN, pLS32, and obtained following results. Transcription repN gene starts 167 nucleotides upstream translational start site repN. The copy number repN-coding plasmid pHDCS2, which was placed downstream IPTG (isopropyl-1-thio-beta-D-galactopyranoside)-inducible Pspac...

To develop a gene transformation method for Flammulina velutipes, we constructed vector with hph under control of the trp1 promoter. The was integrated into protoplast derived from mycelia by calcium-polyethylene glycol method, as it has not been reported F. velutipes. Bacterial extracellular vesicles (BEVs), produced by a broad spectrum of bacteria, play crucial role in enhancing intercellular communication through DNA transfer. A vital determinant their gene transfer efficiency is the content diversity within BEVs, an aspect that conventional metagenomics fails to capture. Our study bridges this gap with novel microdroplet-based sequencing technique precisely details hundreds individual BEVs. This revealed unique profile BEVs from oral pathogen...
