Abstract Phototropic hypocotyl bending in response to blue light excitation is an important adaptive process that helps plants optimize their exposure light. In Arabidopsis thaliana, phototropic initiated by the receptors and protein kinases phototropin1 (phot1) phot2. responses also require auxin transport were shown be partially compromised mutants of PIN-FORMED (PIN) efflux facilitators. We previously described D6 PROTEIN KINASE (D6PK) subfamily AGCVIII kinases, which we proposed directly...
Light triggers chloroplast differentiation whereby the etioplast transforms into a photosynthesizing and thylakoid rapidly emerges. However, sequence of events during remains poorly understood. Using Serial Block Face Scanning Electron Microscopy (SBF-SEM), we generated series 3D reconstructions differentiation, revealing number volume extent envelope membrane surfaces. Furthermore, used quantitative lipid whole proteome data to complement (ultra)structural data, providing time-resolved,...
Malate dehydrogenases (MDHs) convert malate to oxaloacetate using NAD(H) or NADP(H) as a cofactor. Arabidopsis thaliana mutants lacking plastidial NAD-dependent MDH (pdnad-mdh) are embryo-lethal, and constitutive silencing (miR-mdh-1) causes pale, dwarfed phenotype. The reason for these severe phenotypes is unknown. Here, we rescued the embryo lethality of pdnad-mdh via embryo-specific expression pdNAD-MDH. Rescued seedlings developed white leaves with aberrant chloroplasts failed reproduce....
Abstract Plants must balance light capture for photosynthesis with protection from potentially harmful ultraviolet (UV) radiation. Photoprotection is mediated by concerted action of photoreceptors, but the underlying molecular mechanisms are not fully understood. In this study, we provide evidence that UV RESISTANCE LOCUS 8 (UVR8) UV-B, phytochrome red, and cryptochrome blue-light photoreceptors converge on induction FERULIC ACID 5-HYDROXYLASE 1 (FAH1) encodes a key enzyme in phenylpropanoid...
Abstract The plastid genome is transcribed by three different RNA polymerases, one called plastid-encoded polymerase (PEP) and two are nucleus-encoded polymerases (NEPs). PEP transcribes preferentially photosynthesis-related genes in mature chloroplasts while NEP housekeeping during early phases of plant development, it was generally thought that differentiation the building up transcription system precedes system. We have now analyzed detail establishment systems, PEP, germination seedling...
Abstract The plastid genome of higher plants is transcribed by two different types RNA polymerases named nucleus encoded polymerase (NEP) and polymerase. Plastid a multimeric enzyme comparable to eubacterial polymerases. NEP enzymes represent small family monomeric phage-type Dicotyledonous harbor three enzymes, RPOTm, RPOTp, RPOTmp. RPOTm exclusively targeted mitochondria, RPOTp plastids, RPOTmp plastids as well mitochondria. In this article, we have made use T-DNA insertion mutants answer...
Phototropism is a growth response allowing plants to align their photosynthetic organs toward incoming light and thereby optimize activity. Formation of lateral gradient the phytohormone auxin key step trigger asymmetric shoot leading phototropic reorientation. To identify important regulators formation, we developed an flux model that enabled us test in silico impact different morphological biophysical parameters on including contribution extracellular space (cell wall) or apoplast. Our...
Abstract Background Investigations of evolution knowledge and acceptance their relation are central to education research. Ambiguous results in this field study demonstrate a variety measuring issues, for instance differently theorized constructs, or lack standardized methods, especially cross-country comparisons. In particular, meaningful comparisons across European countries, with varying cultural backgrounds systems, rare, often include only few standardization. To address these deficits,...
Photosynthesis, the fundamental process using light energy to convert carbon dioxide organic matter, is vital for life on Earth. It relies capturing through light-harvesting complexes (LHC) in photosystem I (PSI) and PSII conversion of into chemical energy. Composition organization PSI core are well conserved across evolution. particularly sensitive photodamage but benefits from a large diversity photoprotective mechanisms, finely tuned handle dynamic ever-changing conditions. Upon imbibition, dry seeds rapidly gain metabolic activity and the switching on of a germination-specific transcriptional programme in nucleus goes ahead, with induction many nucleus-encoded transcripts coding for plastid-localized proteins. Dedifferentiated plastids present differentiate into chloroplasts cotyledons amyloplasts root hypocotyl, raising question whether beginning new plant's life cycle is also characterized by specific changes plastid programme. Here transcriptome during...
The plastid genome of dicotyledonous plants is transcribed by three different RNA polymerases; an eubacterial-type enzyme, PEP; and two phage-type enzymes, RPOTp RPOTmp. plays important role in chloroplast transcription, biogenesis, mesophyll cell proliferation. RPOTmp fulfills a specific function the transcription rrn operon proplasts/amyloplasts during seed imbibition/germination more general chloroplasts later developmental stages. In chloroplasts, tightly associated with thylakoid...
Abstract Phototropins are light-activated protein kinases, which contribute to photosynthesis optimization both through enhancement of photon absorption when light is limiting and avoidance responses in high light. This duality part endowed by the presence phototropins with different photosensitivity (phot1 phot2). Here we show that phot1, senses low promote positive phototropism (growth towards light), also limits response depends on phot1-mediated phosphorylation Phytochrome Kinase...

In the parasitic plant Striga hermonthica (Del. Benth), asparagine synthesis plays a prominent role in metabolism of host‐derived nitrogen and detoxification process steady‐state N‐excess. Here, we show that synthetase (EC 6.3.5.4), primary enzyme involved production plants, is encoded by small gene family, with at least two AS genes, including called ShAS related to class II Asparagine Synthetase genes. The functionality was demonstrated complementation an E. coli asn auxotroph mutant its...
Abstract Photoperiodic plants coordinate the timing of flowering with seasonal light cues, thereby optimizing their sexual reproductive success. The WD40-repeat protein REPRESSOR OF UV-B PHOTOMORPHOGENESIS 2 (RUP2) functions as a potent repressor UV RESISTANCE LOCUS 8 (UVR8) photoreceptor-mediated induction under noninductive, short-day conditions in Arabidopsis (Arabidopsis thaliana); however, contrast, closely related RUP1 seems to play no major role. Here, analysis chimeric ProRUP1:RUP2...
