N-acetyl-aspartyl-glutamate (NAAG) is a peptide-based neurotransmitter that has been extensively studied in many neurological diseases. In this study, we show specific role of NAAG cancer. We found more abundant higher grade cancers and source glutamate expressing carboxypeptidase II (GCPII), the enzyme hydrolyzes to N-acetyl-aspartate (NAA). Knocking down GCPII expression through genetic alteration or pharmacological inhibition results reduction both concentrations cancer growth. Moreover,...
Abstract ADP-ribosylation is a protein modification responsible for biological processes such as DNA repair, RNA regulation, cell cycle and biomolecular condensate formation. Dysregulation of implicated in cancer, neurodegeneration viral infection. We developed ADPriboDB (adpribodb.leunglab.org) to facilitate studies uncovering insights into the mechanisms significance ADP-ribosylation. 2.0 serves one-stop repository comprising 48 346 entries 9097 ADP-ribosylated proteins, which 6708 were...
Cancer-associated fibroblasts (CAFs), a major component of the tumor microenvironment (TME), play an important role in cancer initiation, progression, and metastasis. Recent findings have demonstrated that TME not only provides physical support for cells but also directs cell-to-cell interactions (in this case, interaction between CAFs). As progresses, CAFs coevolve, transitioning from inactivated state to activated state. The elucidation understanding will pave way new therapies [1-3].
Summary Type I interferons (IFN-I) are cytokines with potent antiviral and inflammatory capacities. IFN-I signaling drives the expression of hundreds stimulated genes (ISGs), whose aggregate function results in control viral infection. A few these ISGs tasked negatively regulating response to prevent overt inflammation. ISG15 is a negative regulator absence leads persistent, low-grade elevation ISG concurrent, self-resolving mild autoinflammation. The limited breadth persistence expressed...
Dysregulation of the immune system in individuals with Down syndrome is thought to play a major role pathophysiology many clinical presentations. This natural history disease study took comprehensive evaluation prevalence different related diagnoses cohort 1299 patients compared 2605 control without at Mount Sinai Health System NY, NY over past 18 years. We conducted stepwise analysis odds receiving diagnosis Chapter, Sub-chapter and Diagnosis level ICD-CM-10 code system. Individuals our had...
Abstract Rationale: In previous studies, it has been found that cancer-associated fibroblasts (CAFs) interaction with carcinoma cells and helped cancer cell grow which contributed to progression aggressiveness. However, how CAFs grow, specifically what nutrition provides tumors not yet fully understood. Objectives: We are studying metabolism identify pathways target for therapy. Therefore, in this study, we sought determine key metabolites glucose metabolism, glycolysis the tricarboxylic...
