This study evaluates the antiallodynic and antihyperalgesic effects of LMH-2, a new haloperidol (HAL) analog that acts as sigma-1 receptor (σ1 R) antagonist, in diabetic mice using model neuropathic pain induced by chronic hyperglycemia. Additionally, we compared its with those HAL. Hyperglycemia was nicotinamide-streptozotocin administration (NA-STZ, 50–130 mg/kg). Four weeks later, mechanical allodynia assessed up-down method, hyperalgesia evoked formalin 0.5%. We evaluated LMH-2 (5.6–56.2...

Heliopsis longipes is used as analgesic in Mexican traditional medicine. The present study assesses the possible antinociceptive effect of and describes pharmacological mechanism action affinin, identified one active principle acetone extract. Intraperitoneal administration H. extract affinin produced a dose-dependent when assessed mice submitted to acetic acid capsaicin tests. Affinin-induced antinociception (30 mg/kg, i. p.) was blocked by naltrexone (1 s. c.), p-chlorophenylalanine (80...

The antinociceptive pharmacological interaction between N-palmitoylethanolamide (PEA) and morphine (MOR), as well gabapentin (GBP), was investigated to obtain synergistic antinociception at doses where side effects were minimal. In addition, the possible mechanism of PEA + MOR or GBP combinations explored.Individual dose-response curves (DRCs) PEA, evaluated in female mice which intraplantar nociception induced with 2% formalin. Isobolographic method used detect combination GBP.The ED50...

Abstract Agave applanata is a Mexican agave whose fresh leaves are employed to prepare an ethanol tonic used relieve diabetes. It also applied skin varicose and diabetic foot ulcers, including wounds, inflammation, infections. In this study, the chemical composition of established its association with antihyperglycemic, anti-inflammatory, antimicrobial, wound healing activities discussed. The A. were extracted : H2O (85 15). A fraction extract was lyophilized, remainder partitioned into...

Heliopsis longipes (A. Gray) Blake (Asteraceae), commonly known in Mexico as "chilcuage" or "chilcuan", is widely used an analgesic and anesthetic agent. Affinin, the major metabolite of this plant, ethanol extract plant have shown antinociceptive properties mice. H. produces a complex mixture antioxidant chlorophylls polyamines well number possible antimutagens. The current study evaluated potential utilization natural product affinin isolated from antimutagenic possibly anticarcinogenic...

To establish the role of TRPV1 receptor in antinociceptive effect natural alkamides (i.e. affinin, longipinamide A, longipenamide A and B) isolated from Heliopsis longipes (A. Gray) S.F. Blake some related synthetic N-isobutyl-feruloylamide N-isobutyl-dihydroferuloylamide).The orofacial formalin test was used to assess activity (1-30 μg, region) (0.1-100 region). Preclinical Research The current work evaluates the interaction between two commonly used drugs, tramadol (Tra) and gabapentin (Gbp). Dose-response curves (DRC) isobolographic analysis were to confirm their synergistic antihyperalgesic anti-allodynic responses in a rat neuropathic pain model involving chronic constriction injury of sciatic nerve von Frey acetone tests. Tra Gbp produced dose-dependent effects. studies combinations combination showed DRC was leftward-shifted compared DRCs for...

Abstract Pain-induced functional impairment in the rat (PIFIR) is a model of inflammatory and arthritic pain similar to that clinical gout. Nociception induced by intra-articular injection uric acid into right hind limb, inducing its dysfunction. Animals then receive analgesic drugs recovery functionality over time assessed as an expression antinociception. We have examined role peripheral prostaglandins synthesized cyclooxygenase-1 (COX-1) cyclooxygenase-2 (COX-2) using PIFIR model....
