Abstract Excretory/Secretory (ES) proteins play an important role in the host-parasite interactions. Experimental identification of ES is time-consuming and expensive. Alternative bioinformatics approaches are cost-effective can be used to prioritize experimental analysis therapeutic targets for parasitic diseases. Here we predicted functionally annotated T. solium genome using integration tools. Additionally, developed a novel measurement evaluate potential antigenicity secretome sequence...
Light chain-associated amyloidosis is a fatal disease characterized by the aggregation and pathologic deposition of monoclonal light chain-related fragments as amyloid fibrils in organs or tissues throughout body. Notably, it has been observed that proteins encoded lambda variable chain (V(L)) gene segment 6a are invariably associated with deposition; however, contribution to this phenomenon not established. In regard, we have determined thermodynamic stability kinetics vitro fibrillogenesis...
The prognosis of myeloma is based on controlling the plasma cell burden and thus, management production monoclonal light chains has improved considerably, expanding survival quality life. However, effects in various organs result alterations that may lead to renal failure. There a crucial need ameliorate or abolish damage. Organ-based therapies must be developed. Glomerulopathic interact with mesangial cells using SORL1 receptor downstream divergent alterations. While multi-step process...
The high-quality draft genomes of two Vibrio parahaemolyticus strains, one that causes the acute hepatopancreatic necrosis disease (AHPND) in cultured shrimps (FIM-S1708(+)), and another does not (FIM-S1392(-)) are reported. A chromosome-scale assembly for FIM-S1392(-) genome is reported here. analysis gives some clues regarding genomic differences between strains.
Hepatocellular carcinoma (HCC) is a highly lethal liver cancer with late diagnosis; therefore, the identification of new early biomarkers could help reduce mortality. We determine tissue and plasma status five annexins during hepatocarcinogenesis by diethylnitrosamine-induced cirrhosis-HCC. found that Anxa5 was earliest upregulated gene at week 12 after HCC initiation, while Anxa1 Anxa2 were in advanced stages (weeks 18 22). Furthermore, protein level Annexin A1, A2, A5 A10 increased from...
Approximately 25% of the λ6 light chains have glycine rather than arginine at position 25, which is an allelic variant IGLV6‐57 ( 6a ) locus. The Gly25 has been shown to decrease folding stability germline V L protein 6aJL2 by 1.7 kcal·mol −1 . In this work, we compared thermodynamic and fibrillogenic properties amyloidosis (AL) derived recombinant (r) AR, contains Gly25, with those rV 6aJL2‐R25G disease‐associated proteins Wil Jto (myeloma). Our experiments show that four AR least stable;...
The Excreted/Secreted (ES) proteins play important roles during Mycobacterium tuberculosis invasion, virulence, and survival inside the host they are a major source of immunogenic proteins. However, molecular complexity bacillus cell wall has made difficult experimental isolation total bacterial ES Here, we reported genomes two Beijing genotype M. clinical isolates obtained from patients Vietnam (isolate 46) South Africa 48). We developed bioinformatics pipeline to predict their secretomes...
Abstract Amyloid light‐chain (AL) amyloidosis is a rare disease in which plasma‐cell‐produced monoclonal immunoglobulin light chains misfold and become deposited as fibrils the extracellular matrix. λ6 subgroup are particularly fibrillogenic, around 25 % of amyloid‐associated exist allotypic G24R variant that renders protein less stable. The molecular details this process, well structures fibrils, unknown. We have used solid‐state NMR to investigate different fibril polymorphs. secondary...
Abstract Immunoglobulin light chain-derived (AL) amyloidosis is a debilitating disease without known cure. Almost nothing about the structural factors driving amyloidogenesis of chains. This study aimed to identify fibrillogenic hotspots model protein 6aJL2 and in pursuing this goal, two complementary approaches were applied. One them was based on several web-based computational tools optimized predict fibrillogenic/aggregation-prone sequences different biophysical properties polypeptide...
Hepatocellular carcinoma (HCC) is one of the deadliest cancers worldwide, often preceded by cirrhosis and usually diagnosed at advanced stages; therefore, identifying molecular changes early stages an attractive strategy for detection timely treatment. Here, we investigated progressive transcriptomic during experimental hepatocarcinogenesis to identify novel tumor markers in HCC model induced chronic administration sublethal doses diethylnitrosamine. An analysis differentially expressed...
Abstract Variable domain (V L ) gene segments exhibit variable tendencies to be associated with light chain amyloidosis (AL). While few of them are very frequent in AL and give rise most the amyloidogenic chains compiled at sequence databases, other rarely found among cases. To analyze which extent these depend on folding stability aggregation propensity germline V protein, we characterized proteins encoded by four AL-associated one not AL. We that rV differ widely conformational vitro...
Abstract The potential role of hepatocytes versus hepatic progenitor cells (HPC) on the onset and pathogenesis hepatocellular carcinoma (HCC) has not been fully clarified. Because administration 2‐acetylaminofluorene (2AAF) followed by a partial hepatectomy, selectively induces HPC proliferation, we investigated effects chronic 2AAF HCC development caused carcinogen diethylnitrosamine (DEN) for 16 weeks in rat. DEN + protocol impeded weight gain animals but promoted prominent hepatomegaly...
