Circular extrachromosomal DNA (ecDNA) in patient tumors is an important driver of oncogenic gene expression, evolution drug resistance and poor outcomes. Applying computational methods for the detection reconstruction ecDNA across a retrospective cohort 481 medulloblastoma from 465 patients, we identify circular 82 patients (18%). Patients with ecDNA-positive were more than twice as likely to relapse three times die within 5 years diagnosis. A subset harbored multiple lineages, each...

Abstract Cell-free DNA (cfDNA) is increasingly recognized as a promising biomarker candidate for disease monitoring. However, its utility in neurodegenerative diseases, like amyotrophic lateral sclerosis (ALS), remains underexplored. Existing discovery approaches are tailored to specific context or too expensive be clinically practical. Here, we address these challenges through new approach combining advances molecular and computational technologies. First, develop statistical tools select...

Ependymoma is a tumor of the brain or spinal cord. The two most common and aggressive molecular groups ependymoma are supratentorial ZFTA-fusion associated posterior fossa group A. In both groups, tumors occur mainly in young children frequently recur after treatment. Although mechanisms underlying these diseases have recently been uncovered, they remain difficult to target innovative therapeutic approaches urgently needed. Here, we use genome-wide chromosome conformation capture (Hi-C),...

It is widely believed that selective packaging of nucleic acids, especially microRNAs, into exosomes secreted by the cancer cells not only ensures their growth and survival but also helps in escape from immune surveillance. Keeping view fact human cellular miR‐2909 has emerged to regulate genes involved oncogenesis immunity, present study was addressed reveal nature expression within different tissue origin its incorporation these cells. Post‐transcriptional modification, 3′‐end adenylation...

SUMMARY Extrachromosomal circular DNA (ecDNA) is an important driver of aggressive tumor growth, promoting high oncogene copy number, intratumoral heterogeneity, accelerated evolution drug resistance, enhancer rewiring, and poor outcome. ecDNA has been reported in medulloblastoma (MB), the most common malignant pediatric brain tumor, but landscape its association with specific MB subgroups, impact on potential clinical implications, are not known. We assembled a retrospective cohort 468...

NO donor drugs showed a significant therapeutic effect in the treatment of many diseases, such as arteriopathies, various acute and chronic inflammatory conditions, several degenerative diseases. NO-releasing anti-inflammatory are prototypes novel class compounds, combining pharmacological activities anti-nociceptive with those NO, thus possessing potential applications great variety In this study, we designed predicted biological activity by targeting cyclooxygenase type 2 (COX-2) NF-κB...

Biochemical genetic and karyomorphological studies were evaluated for taxonomic importance. Five Schizothorax species namely: niger, curvifrons, esocinus, labiatus plagiostomus studied in this experiment as a model serum proteins chromosomal analysis. Serum protein band patterns of complex by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). Their electrophoretograms revealed similarities well differences the number molecular weight bands. S. esocinus showed five bands,...

Abstract Liver is the largest solid organ and performs hundreds of vital functions including drug metabolism. diseases, viral infections, cancer metabolic disorders account for about 2 million deaths annually, worldwide (1). Current models studying liver biology metabolism include 2D cultured primary cells/cell lines, which do not represent structural complexity human tissue. In vivo diseases are time consuming, cost prohibitive often ineffective. Therefore, better tissue mimetic needed....

Abstract Worldwide, breast cancer (BC), is the most frequently diagnosed among women. In US, 1 out of 8 women may expect to be with BC their lifetime. Current models for studying tumor biology and drug discovery include 2D cultured cell lines patient derived xenografts (PDX). As cells do not represent 3D complexity human tissue, PDX are time consuming cost prohibitive, better tissue mimetic needed. Organoids self-assembled cellular units that mimic corresponding structurally functionally...

Genomic regulation and functional significance of PVT‐1 gene locus, in the MYC‐driven cancers, has remained enigmatic ever since its discovery. With present study, an attempt is made to establish that cellular AATF genome encoded miR‐2909 RNomics pathway involving crucial genes coding for KLF4, Deptor, mTORC1, STAT3, p53 inherent capacity ensure sustained co‐amplification locus together with c‐Myc gene. Based upon these results, we propose may play a role tumorigenic locus.

Abstract Background & Aims MicroRNAs (miRNAs) are transported in body fluids within exosomes, and exosomal-miRNA sorting is highly selective. In colonic epithelial cells (CECs), Substance P-neurokinin-1-receptor (SP/NK1R) signaling regulates miR-21 into secreted exosomes. Here, we studied the molecular mechanisms driving exosomes (CEEs) under SP-stimulation. Methods We performed studies with human NCM460 overexpressing neurokinin-1-receptor (NCM460-NK1R) intestinal-epithelial-specific...
