Abstract Secretion systems are protein export machines that enable bacteria to exploit their environment through the release of effectors. The Type 9 System (T9SS) is responsible for across outer membrane (OM) phylum Bacteroidota. Here we trap T9SS Flavobacterium johnsoniae in process substrate transport by disrupting motor complex. Cryo-EM analysis purified substrate-bound translocons reveals an extended translocon structure which previously described core augmented a periplasmic...

Rod-shape of most bacteria is maintained by the elongasome, which mediates synthesis and insertion peptidoglycan into cylindrical part cell wall. The elongasome contains several essential proteins, such as RodA, PBP2, MreBCD but how its activities are regulated remains poorly understood. Using E . coli a model system, we investigated interactions between core proteins in vivo Our results show that PBP2 RodA form complex mediated their transmembrane periplasmic parts independent catalytic...

In Gram negative bacteria the outer membrane (OM) is first line of defence against antimicrobial agents and immunological attacks. A key part OM biogenesis insertion proteins (OMPs) by beta barrel assembly machinery (BAM). Here we report cryo-electron microscopy (cryoEM) structure a BAM complex isolated from Flavobacterium johnsoniae, member phylum Bacteroidota that includes human commensals major anaerobic pathogens. This radically different canonical Escherichia coli system an extensive...

The gliding motility and Type 9 Secretion Systems (T9SSs) of Bacteroidota share a Power Chain that in current models uses Hub complex to distribute energy both the T9SS translocon moving track. In Flavobacterium johnsoniae cell surface adhesins are attached internal track by outer membrane-spanning protein SprF. We show SprF component PorG contain twin-cysteine motifs form disulfide link shared cysteine residue GldK. These linkages crucial for export, respectively. Since moves with track,...

Summary Peptidoglycan (PG) is the unique cell shape‐determining component of bacterial envelope, and a key target for antibiotics. PG synthesis requires transmembrane movement precursor lipid II, MurJ has been shown to provide this activity in Escherichia coli . However, how functions vivo not reported. Here we show that localizes both lateral membrane at midcell, recruited midcell simultaneously with late‐localizing divisome proteins MraY MurG. septal localization dependent on presence...

Insertion of new material into the Escherichia coli peptidoglycan (PG) sacculus between cytoplasmic membrane and outer requires a well-organized balance synthetic hydrolytic activities to maintain cell shape avoid lysis. Since most bacteria carry multiple enzymes carrying same type PG activity, we know little about specific function given enzymes. Here show that DD-carboxy/endopeptidase PBP4 localizes in PBP1A/LpoA FtsEX dependent fashion at midcell during septal synthesis. Midcell...

ABSTRACT Insertion of new material into the Escherichia coli peptidoglycan (PG) sacculus between cytoplasmic membrane and outer requires a well-organized balance synthetic hydrolytic activities to maintain cell shape avoid lysis. The enzymes outnumber that insert PG by far very little is known about their specific function. Here we show DD-carboxy/endopeptidase PBP4 localizes in PBP1A/LpoA FtsEX dependent fashion at midcell during septal synthesis. Midcell localization its non-catalytic...

Abstract Secretion Systems are protein export machines that enable bacteria to exploit their environment through the release of effectors. The Type 9 System (T9SS) is an essential virulence determinant in oral pathogens responsible for periodontitis and immune dysfunction. We have trapped structurally characterised translocon process substrate transport. active augmented by a large periplasmic structure incorporating homologue canonical chaperone Skp. Substrate proteins bind carrier within...

In the present paper, we are going to show that outside a slim set in sense of Malliavin (or quasi-surely), signature path (which consists iterated integrals every degree) Brownian motion is non-self-intersecting. This property relates closely non-degeneracy for rough arising naturally from uniqueness problem theory. As an important consequence conclude quasi-surely, does not have any tree-like pieces and sample uniquely determined by its up reparametrization.
