ABSTRACT Background and Aim Oxidative stress has been considered a factor in the development of post‐endoscopic retrograde cholangiopancreatography pancreatitis (PEP). The present clinical trial evaluated whether adding intravenous mesna to rectal indomethacin could prevent or alleviate PEP. Methods An open‐labeled was done on 698 participants undergoing endoscopic (ERCP). Eligible patients received 100 mg suppository 30 min before ERCP. Randomly, 400 nothing doing procedure. PEP incidence...

Leukemia is one of the most common cancers in prepubertal girls and adolescents, with advances improving survival rates. However, treatments like chemotherapy radiation are highly gonadotoxic, often causing ovarian insufficiency, early menopause, infertility, endocrine disorders. Fertility preservation for young female cancer patients, especially without mature germ cells, relies heavily on tissue cryopreservation. Yet, a major concern potential presence leukemic cells within preserved...

Background: Given the limited evidence on efficacy of α-pinene, a herbal terpenoid, in management ulcerative colitis (UC), this clinical trial aims to investigate effects α-pinene compared placebo. Methods: Eligible UC patients were randomized receive either or placebo, alongside their routine pharmacologic regimen, for two months. Quality life (QoL) and disease severity assessed using Simple Clinical Colitis Activity Index Questionnaire (SCCAIQ) Crohn’s Questionnaire-8 (CUCQ-8),...

Chemotherapeutic agents induce irreversible gonadotoxic side effects, resulting in endocrine dysfunction and infertility female cancer survivors. In the current study, we investigated strategies to protect ovarian function from chemotherapy-induced toxicity by evaluating effects of cisplatin, doxorubicin, or cyclophosphamide on vasculature primordial follicle survival. This investigation was conducted using adult CD-1, postnatal 5-7 oocyte-specific Trp63 conditional knockout (Trp63 cKO) mice...

Cyclophosphamide and doxorubicin lead to premature ovarian insufficiency as an off-target effect. However, their oocyte death pathway has been debated. Here, we clarified the precise mechanism of depletion induced by cyclophosphamide doxorubicin. Dormant oocytes instead activated with high PI3K activity were more sensitive cyclophosphamide. Checkpoint kinase 2 (CHK2) inhibitor rather than GNF2 protected from doxorubicin, up-regulated p-CHK2 depleted primordial follicles in Abl1 knockout...

Abstract Pancreatic ductal adenocarcinoma (PDAC) is one of the most lethal cancers with high incidence rates metastasis and cachexia. High circulating activin A, a homodimer inhibin βA subunits that are encoded by INHBA gene, predicts poor survival among PDAC patients. However, it still raises question whether A suppression renders favorable outcomes. Here, authors demonstrate abundantly detected in tumor stromal cells on tissue microarray mouse sections. In orthotopic male mice,...

Abstract Cisplatin, a platinum-containing alkylating agent, is used in the treatment of various tumors owing to its potent antitumor activity. However, it causes permanent and adverse effects, particularly hearing loss depletion ovarian reserve. Until recently, there were no clinically available protective agents mitigate side effects cisplatin-induced cytotoxicity. In 2022, sodium thiosulfate (STS) was approved by Food Drug Administration for mitigating children adolescents undergoing...

The TAp63α protein is highly expressed in primordial follicle oocytes, where it typically exists an inactive dimeric form. Upon DNA damage, undergoes hyperphosphorylation, transitioning from a to tetrameric structure, which initiates oocyte apoptosis by upregulating pro-apoptotic gene. Our results demonstrate that cisplatin, alkylating anti-cancer agent, predominantly produced the dimer rather than tetramer. We further observed accumulation occurred oocytes following cisplatin treatment, and...

This study evaluated the distribution and size of lipid droplets (LDs) in oocytes recovered from young adult ovine ovaries. Collected were categorised on basis their major diameter (small (SO), 70–90 µm; medium (MO), >90–110 large (LO), >110–130µm) stained with Nile red to detect LDs. In oocytes, a diffuse pattern LDs was dominant all classes except LO SO LO. Larger (i.e. >3µm) mostly present LO, whereas smaller (1–3µm) detected other oocyte categories.

Kim: Background: Chemotherapy demonstrates efficacy to destroying cancer cells; however, it can inadvertently impact normal healthy cells, leading irreversible side effects, particularly affecting the ovaries in female patients. Female survivors encounter endocrine dysfunction and infertility due ovarian damage caused by gonadotoxic therapies. The ovary, a vital organ responsible for producing oocytes hormones, faces...

Accumulated evidence indicates that cisplatin, a platinum-based alkylating agent, causes preferential DNA damage to oocytes of primordial follicles (PFs) in the ovary, suggesting oocyte-favored accumulation cisplatin. Copper transporter 1 (CTR1; Slc31a1 ) is implicated facilitating cisplatin uptake cells. Here we found PFs had constitutively higher expression CTR1 than other cell types mouse ovary. However, oocyte-specific knockout was not sufficient prevent cisplatin-induced depletion vitro...

Age-associated decline in female fertility is largely attributable to decrease oocyte quality. However, the molecular mechanisms that shape developmental competence, and may be involved reproductive aging, are yet elucidated. It appears functionally conserved across mammals; aberrant expression its members was observed several animal models differential mutations...
