Hyaluronic acid (HA) ophthalmic solution is widely used in dry eye treatment worldwide. However, there are no reports comparing the effects of high molecular weight HA with low HA. Sixty eight-week-old C57BL/6 mice were assigned to following 6 groups and exposed environmental stress (EDES) that mimics office work environment: (1) 0.1% (LMWHA) drops, (2) 0.3% LMWHA (3) 3% diquafosol sodium (DQ) (4) 0.15% (HMWHA) (5) exposure EDES (EDES+/Treatment−), (6) without (EDES−/Treatment−). After EDES,...
Abstract Objective —To examine in vitro effects of various antiproteolytic compounds on activity matrix metalloproteinase (MMP)-2 and -9 the tear film horses with active corneal ulcers. Sample Population —Samples obtained from eyes 34 ulcerative keratitis. Procedure —Horses were sedated, samples collected lower fornix ulcerated by use capillary tubes. The protease inhibitors 0.2% EDTA, 0.1% doxycycline, 10% N-acetylcysteine (NAC), solution a modified dipeptide that contains hydroxamic acid...
Monoclonal antibodies (Mabs) were used for immunohistochemical location of integrin beta- (beta 1,3, and 4) alpha subunits (alpha 1-6 v) in the epithelium both normal tissue-cultured human cornea. Immunoreaction beta 1 subunit was most intense at membranes basal epithelial cells weaker superficial cell membranes. Anti-beta 4 immunofluorescence appeared part only, apposing basement membrane. Both anti-alpha 2 anti-beta 3 Mabs showed an immunoreaction with distribution similar to subunit. 6...
AbstractPlasmin can degrade fibronectin and laminin, two important components of the extracellular matrix facilitating cell sliding healing following a wound. In this study we investigated relationship between tear fluid level plasmin plasminogen activator corneal Anterior keratectomy (AKE) was performed for seven rabbits (11 eyes). Eight eyes were rewounded after re-epithelialization. Tear samples collected with capillaries before wounding during wound healing. Plasmin (PA) activities...
The aim of the HYLAN M study was to investigate if symptoms and/or signs patients suffering from severe dry eye disease (DED) can be improved by substituting individually optimized artificial tear therapy high molecular weight hyaluronan (HMWHA) drops. In this international, multicenter study, with at least ocular surface index (OSDI) 33 and corneal fluorescein staining (CFS) Oxford grade 3 were included. A total 84 per-protocol randomized in two arms. control group continued use their...
Autologous serum eye drops (ASED) are used in the treatment of most severe stages dry eye. Once introduced, it is currently considered impossible to return other lubricating or commercially available therapeutic regimen. In a randomized study, non-preserved high-molecular-weight hyaluronic acid were offered as an alternative 11 patients using autologous for at least 3 months. The control group (n = 5) continued their with ASED. verum 6) very-high-molecular-weight (Comfort Shield®) instead...

Abstract Tenascin, a novel extracellular matrix protein, was demonstrated immunohistochemically in rabbit corneas that had healed after anterior keratectomy. Tenascin‐like immunofluorescence (T‐LI) appeared the corneal stroma at wound area only. The edge showed most intense specific fluorescence. epithelium, general, displayed moderate T‐LI. Wounding did not induce any change epithelial T‐LI or located outside wound.
Abstract Purpose Lubricant eye drops are the main therapeutic resource for dry disease (DED), with each drop representing equivalent of ocular surface treatment. Thus, any reduction in frequency application reflects a degree success. Considering also socioeconomic burden DED, we investigated (DF) as parameter to potentially track success therapy severe DED. Hyaluronan (HA)‐containing have become first choice tear substitution many countries, and recent data indicate that average molecular...
The purpose of this study was to investigate the effect high molecular weight hyaluronan (HMWHA) eye drops on subbasal corneal nerves in patients suffering from severe dry disease (DED) and evaluate damage associated with DED. Designed as an international, multicenter study, 16 symptoms at least Ocular Surface Disease Index (OSDI) score 33, fluorescein staining (CFS) Oxford grade 3, were included randomized into two arms. control group continued use their individual optimum artificial tears...
A rapid (5- to 10-min), sensitive (detection limit 0.6 IU/L), and moderately specific fluorometric plasmin assay for small volume tear fluid samples was developed. Addition of albumin (up 0.1% final concentration) the buffer improved sensitivity test so that activity in healthy controls could be detected. pH reaction 8.0, Michaelis-Menten constant substrate, H-D-Val-Leu-Lys.7-amido-4-methyl-coumarin (AMC), 0.28 mM, substrate concentration 1 mM. Intra- interassay imprecisions were 1.6 4.4%,...
Aqueous humor (AH) collected from healthy eyes of man and rabbit was analysed for proteolytic activity using a casein-agar assay. Normal AH during surgery or aspirated before ocular irritation contained no caseinolytic activity. In rabbits, mechanical the iris subconjunctival as well intracameral injection capsaicin led to irritative response. samples after such manipulation considerable On other hand, anterior chamber puncture alone did not seem induce elevation in AH. The clinical...
