Chitinase-3-like-1 (CHI3L1), also known as YKL-40, is a glycoprotein linked to inflammation, fibrosis, and cancer. This study explored CHI3L1's interactions with various oligosaccharides using microscale thermophoresis (MST) AlphaScreen (AS). These investigations guided the development of high-throughput screening assays assess interference small molecules in binding between CHI3L1 biotinylated or heparan sulfate-based probes. Small molecule binders YKL-40 were identified our chitotriosidase...
A series of new cyclopentaquinoline derivatives with 9-acridinecarboxylic acid and a different alkyl chain length were synthesized, their ability to inhibit cholinesterases was evaluated. All designed compounds, except derivative 3f, exhibited selectivity for butyrylcholinesterase (BuChE) IC50 values ranging from 103 539 nM. The 3b revealed the highest inhibitory activity towards BuChE (IC50 = 103.73 nM) suitable against AChE 272.33 nM). 3f most active compound 113.34 satisfactory 203.52...
Abstract Despite extensive research over the past three decades, there are still uncertainties regarding catalytic mechanism of human chitotriosidase-1. To fill gap, we reanalysed structural information available for this enzyme. Based on existing and new experimental data, complemented by multi-scale simulations, modelled full-length structure chitotriosidase-1 proposed general model its mechanism. We have elucidated role four highly conserved motifs present in glycoside hydrolases 18...
MOFOID is a new server developed mainly for automated modeling of protein structures by their homology to the deposited in PDB database. Selection template and calculation alignment performed with Smith-Waterman or Needleman-Wunsch algorithms implemented EMBOSS package. The final model built optimised programs from JACKAL wide spectrum options web-based interface possibility uploading user's own make suitable platform testing approaches building. available at https:// valis.ibb.waw.pl/mofoid/.
Protein–protein recognition, leading to the formation of specific functional complexes, involves complementary surfaces interacting subunits. Current docking protocols employ complex scoring functions, neglecting proper shape matching by use cubic grids. In present paper a algorithm based on tetrahedral grid model proteins is described, allowing more precise description complementarity. The software was tested benchmark, giving excellent results for rigid-body docking. Extension methodology...
Viral pneumonia caused by highly infectious SARS-CoV-2 poses a higher risk to older people and those who have underlying health conditions, including Alzheimer’s disease. In this work we present newly designed tacrine-based radioconjugates with physicochemical biological properties that are crucial for the potential application as diagnostic radiopharmaceuticals. A set of ten tacrine derivatives was synthesized, labelled gallium-68 fully characterized in context their properties. Based on...
The initial aim of this work was to elucidate the mutual influence different single-stranded segments (loops and caps) on thermodynamic stability RNA G-quadruplexes. To end, we used a new NAB-GQ-builder software program, construct dozens two-tetrad G-quadruplex topologies, based designed library sequences. Then, probe sequence-morphology-stability relationships performed molecular dynamics simulations. Their results provide guidance for design G-quadruplexes with balanced structures, in turn...
Azobenzene molecules exhibit reversible light-triggered changes in geometrical structure (cis/trans isomerism) and nanoscale mechanical properties. For this reason, they have been already widely used materials science to build photo-mechanical responsive systems, incorporated electronic switchable devices, for graphene slicing, optical switching data storage. We exploring further the photoresponsive nature of newly designed azobenzene derivatives exploiting potential these smart generation...
