GLUT4 in muscle and adipose tissue is important maintaining glucose homeostasis. However, the role of insulin-responsive central nervous system has not been well characterized. To assess its importance, a selective knockout brain (BG4KO) was generated by crossing Nestin-Cre mice with GLUT4-floxed mice. BG4KO had 99% reduction protein expression throughout brain. Despite normal feeding fasting glycemia, were intolerant, demonstrated hepatic insulin resistance, reduced uptake In response to...

Abstract Background BTRX-246040, a nociceptin/orphanin FQ peptide receptor antagonist, is being developed for the treatment of depressive patients[1,2]. However, underlying mechanism this potential antidepressant still largely unclear. Here, we studied antidepressant-related actions BTRX- 246040 in ventrolateral periaqueductal gray (vlPAG). Methods The tail suspension test, forced swim female urine sniffing sucrose preference and learned helplessness (LH) combined with pharmacological...

Objectives Assess direct versus indirect action(s) of ghrelin on hypothalamic NPY neurons. Materials and methods Electrophysiology was used to measure ion channel activity in NPY-GFP neurons slice preparations. Ca2+ imaging monitor activation isolated GFP-labeled Immunohistochemistry localize Trpm4, SUR1 Kir6.2 the hypothalamus. Results Acylated depolarized membrane potential (MP) brain slices. Depolarization resulted from a decreased input resistance (IR) ~70% (15/22) or an increased IR...

The interaction between phthalocyanine (ZnPc) and poly(ethylene oxide) (PEO) in water has been studied by dynamic laser light scattering transient absorption fluorescence spectroscopies. After the addition of PEO, intensity at ca. 490 nm, arising from triplet state nonaggregated ZnPc, increases, lifetime becomes longer. These results together with quenching experiments reveal that PEO behaves as a dispersion agent to disstack macrocycles into individual molecules. It found an equimolar...

The conantokins are a family of small, naturally occurring gamma-carboxyglutamate (Gla)-rich peptides that specifically antagonize the N-methyl-D-aspartate (NMDA) subtype ionotropic glutamate receptor. One member this family, conantokin-G (con-G), undergoes Ca(2+)-mediated self-assembly to form an antiparallel helical dimer. Subunit interactions in complex incumbent upon intermolecular Ca(2+) bridging Gla residues spaced at i, i + 4, 7, 11 intervals within monomer. Herein, we further probe...

AMPA-type ionotropic glutamate receptors mediate the majority of fast excitatory neurotransmission in mammalian central nervous system and are essential for brain functions, such as memory learning. Dysfunction these has been implicated a variety neurological diseases. Using laser-pulse photolysis technique, we investigated channel opening mechanism GluRDflip or GluR4flip (i.e., flip isoform GluRD), an AMPA receptor subunit. The minimal kinetic is consistent with binding two molecules per...

2,3-Benzodiazepine derivatives are AMPA receptor inhibitors, and they potential drugs for treating some neurological diseases caused by excessive activity of receptors. Using a laser-pulse photolysis rapid solution flow techniques, we characterized the mechanism action 2,3-benzodiazepine derivative, termed BDZ-f, measuring its inhibitory effect on channel-opening channel-closing rate constants as well whole-cell current amplitude homomeric GluA2Q channels. We also investigated whether BDZ-f...

The present study tests the hypothesis that changes in glucose sensitivity of lateral hypothalamus (LH) hypocretin/orexin glucose-inhibited (GI) neurons following weight loss leads to glutamate plasticity on ventral tegmental area (VTA) dopamine and drives food seeking behavior. C57BL/6J mice were calorie restricted a 15% body maintained at for 1 week. LH GI VTA was measured using whole cell patch clamp recordings brain slices. Food behavior assessed conditioned place preference (CPP)....

The most sufficient test methods are based on a case set which covers all combinations of parameters. However, the scale cases is always too large and their cost cannot be accepted. People will first consider implementation critical cases. Even if terminated suddenly, high importance have been executed.
