Dysfunction of glucagon-secreting α-cells participates in the progression diabetes, and glucagon receptor (GCGR) antagonism is regarded as a novel strategy for diabetes therapy. GCGR upregulates glucagon-like peptide 1 (GLP-1) secretion and, notably, promotes β-cell regeneration diabetic mice. Here, we aimed to clarify role GLP-1 (GLP-1R) activated by and/or antagonism–induced regeneration. We showed that db/db mice type wild-type or Flox/cre mice, monoclonal antibody (mAb) improved glucose...

G protein–coupled receptor 39 (GPR39) senses the change of extracellular divalent zinc ion and signals through multiple proteins to a broad spectrum downstream effectors. Here, we found that GPR39 was prevalent at inhibitory synapses spinal cord somatostatin-positive (SOM + ) interneurons, mechanosensitive subpopulation is critical for conveyance mechanical pain. complexed specifically with glycine receptors (GlyRs) helped maintain glycinergic transmission in manner independent protein...

Abstract High myopia is one of the leading causes visual impairment worldwide with high heritability. We have previously identified genetic contribution SLC39A5 to nonsyndromic and demonstrated that disease‐related mutations dysregulate TGF‐β pathway. In this study, mechanisms underlying involvement in pathogenesis are determined. observed morphogenesis migration abnormalities knockout (KO) human embryonic kidney cells (HEK293) found a significant injury ECM constituents. To investigate the effects of long-term administration tacrolimus (also known as FK506) on pain-related behaviors in mice and to study underlying mechanism pain induced by FK506 via measuring effect synaptic expression phosphorylation alpha-amino-3-hyroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptor spinal cord dorsal horn mice.

Glucagon-secreting pancreatic α-cells can interact with β-cells. However, the long-term effect of glucagon on function and phenotype β-cells has remained elusive. Our new finding shows that induces β-cell dedifferentiation in cultured FoxO1 inhibitor mimicks whereas signaling blockage by GCGR mAb reverses glucagon. In type 2 diabetic mice, increases area, improves function, inhibits dedifferentiation, is partially mediated FoxO1.

Sodium-glucose co-transporter 2 inhibitors, including dapagliflozin, improve ß cell function in type diabetic individuals. Whether dapagliflozin can protect islet microvascular endothelial cells (IMECs) and thus contribute to the improvement of remains unknown. The db/db mice were treated with or vehicle for 6 weeks. function, capillaries levels inflammatory chemokines IMECs detected. Although large-scale neuroimaging studies have demonstrated similar patterns of structural brain abnormalities across major psychiatric disorders, the underlying genetic etiology behind these cross-disorder is not well understood. We quantified extent shared components between cortical structures and disorders (CS-MPD) by using genome-wide association study (GWAS) summary statistics 70 (surface area thickness whole cortex 34 regions) five consisting attention deficit hyperactivity disorder...

Background. Transumbilical single-incision laparoscopic surgery (TUSILS) has emerged as an advanced technique for minimally invasive surgery. Recently, the authors performed TUSIL resection of ileocecal junction benign lesions. Methods. The report surgical and clinical effect in 2 patients. Results. operations were completely successful with satisfactory scarless recovery. No postoperative complications occurred. Conclusions. was feasible safe patients might lead to expansion indications TUSILS.
