Acinetobacter baumannii is a highly antibiotic-resistant bacterial pathogen for which novel therapeutic approaches are needed. Unfortunately, the drivers of virulence in A . remain uncertain. By comparing genomes among panel strains we identified specific gene variation capsule locus that correlated with altered virulence. While less virulent possessed intact gtr6 , hypervirulent clinical isolate contained spontaneous transposon insertion same gene, resulting loss branchpoint capsular...

Microbes synthesize and secrete siderophores, that bind solubilize precipitated or otherwise unavailable iron in their microenvironments. Gram (-) bacterial TonB-dependent outer membrane receptors capture the resulting ferric siderophores to begin uptake process. From similarity

Siderophores are iron-chelating molecules that solubilize Fe3+ for microbial utilization and facilitate colonization or infection of eukaryotes by liberating host iron bacterial uptake. By fluorescently labeling membrane receptors binding proteins, we created 20 sensors detect, discriminate, quantify apo- ferric siderophores. The sensor proteins originated from TonB-dependent ligand-gated porins (LGPs) Escherichia coli (Fiu, FepA, Cir, FhuA, IutA, BtuB), Klebsiella pneumoniae (IroN, FyuA),...

Gram-negative bacteria acquire ferric siderophores through TonB-dependent outer membrane transporters (TBDT). By fluorescence spectroscopic hgh-throughput screening (FLHTS), we identified inhibitors of enterobactin (FeEnt) uptake Escherichia coli FepA (EcoFepA). Among 165 found in a primary screen 17,441 compounds, evaluated 20 secondary tests: siderophore and colicin killing proton motive force-dependent lactose transport. Six hits inhibited activity all tests. Comparison their effects on...

A total of 230 cultures fungi and 43 bacteria, isolated from such sources as soil, butter, milk, were screened for their milk-clotting activity. The cultivated on semisolid media, the bacteria grown in milk media shake culture. Phytic acid, added calcium phytate, was found to stimulate production enzyme most bacterial isolates. Proteolytic activity invariably be associated with There considerable variation ratio two enzymes strain strain.

Acinetobacter baumannii is a problematic pathogen due to its common expression of extensive drug resistance (XDR) and ability survive in the healthcare environment. These characteristics are mediated, part, by signal transduction system BfmR/BfmS. We previously demonstrated, antimicrobial sensitive clinical isolates, that BfmR conferred increased meropenem polymyxin E. In this study, potential mechanisms were informed, prior transcriptome analysis isolate AB307-0294, which identified porins...

This study reports the identification of a chitin deacetylase gene in Cryptococcus laurentii strain RY1 over‐expressing under nitrogen limitation by differential display. The up‐regulation took place robustly growing cells rather than starving quiescent autophagic cells. Quantitative Real Time‐PCR, enzyme activity cell lysate and wall analysis corroborated limitation. These results suggest might play significant role limiting growth RY1.

The Acetobacteraceae family of the class Alpha Proteobacteria is comprised high sugar and acid tolerant bacteria. Thus identification organisms such environments greatly dependent on modern tools molecular biology which require a thorough knowledge...

Bacterial iron acquisition systems are potential targets for new antibiotics, because plays a central role in energy production, DNA synthesis, intermediate metabolism, nitrogen fixation, and oxygen detoxification. Bacteria actively acquire by secreting siderophores that chelate Fe 3+ , then transporting the ferric siderophore complexes. Pathogenic K. pneumoniae produces at least four siderophores: enterobactin, glycosylated yersiniabactin, aerobactin. Like all Gram‐negative species, obtains...
