Defective insulin signaling in hepatocytes is a key factor type 2 diabetes. In obesity, activation of calcium/calmodulin-dependent protein kinase II (CaMKII) suppresses ATF6, which triggers PERK-ATF4-TRB3 pathway that disrupts signaling. Elucidating how CaMKII ATF6 therefore essential to understanding this resistance pathway. We show phosphorylates and blocks nuclear translocation histone deacetylase 4 (HDAC4). As result, HDAC4-mediated SUMOylation the corepressor DACH1 decreased, protects...

Purpose: Dyslipidemia is characterized by high lipid blood levels that are risk factors for cardiovascular diseases, which leading causes of death. However, it unclear whether dyslipidemia a cause the dry eye syndrome (DES). Therefore we determined in transgenic mice models dyslipidemia, there an association with DES development. Methods: Dyslipidemic included male and female adult overexpressing apolipoprotein CIII (Apo CIII), LDL receptor knockout (LDLR-KO) ApoE (ApoE-KO). They were...

Abstract New onset of diabetes is associated with the use statins. We have recently demonstrated that pravastatin‐treated hypercholesterolemic LDL receptor knockout (LDLr −/− ) mice exhibit reductions in insulin secretion and increased islet cell death oxidative stress. Here, we hypothesized these diabetogenic effects pravastatin could be counteracted by treatment antioxidant coenzyme Q 10 (CoQ ), an intermediate generated cholesterol synthesis pathway. LDLr were treated and/or CoQ for 2...

Statins are successful drugs used to treat hypercholesterolemia, a primary cause of atherosclerosis. In this work, we investigated how hypercholesterolemia and pravastatin treatment impact macrophage mitochondria functions, the key cell involved in atherogenesis. By comparing bone marrow-derived macrophages (BMDM) wild-type (WT) LDL receptor knockout (LDLr −/− ) mice, observed increased number contact sites at mitochondria-associated endoplasmic reticulum (ER) membranes (MAMs), enhanced...

Summary Commercial concentrated Concord (CCJ) and Isabel (CIJ) grapes juices were stored at 4–5 °C while pasteurised ready‐to‐drink of the same grape cultivars (PCJ PIJ) kept 20–25 under indirect light for 10 months, simulating industrial storage conditions. (+)‐catechin preservation during ranged between 63% (PCJ) 52% (PIJ); (−)‐epicatechin retention was 32% 15% (CIJ). Total phenols from 93% to 84% radical scavenging activity (RSA) 87% (PIJ) 85% (CCJ PCJ). Concentrated showed higher...

ABSTRACT The study evaluated the sensory characteristics and phenolic contents of concentrated red grape juice two cultivars (Concord Isabel), over 8 months storage. To approach aspects, modified descriptive analysis was used. Total phenols were quantified by Folin–Ciocalteu assay catechins ([ + ]‐catechin [ − ]‐epicatechin) high‐performance liquid chromatography with fluorescent detection. results showed fairly stable behavior both juices a higher stability Concord. An early event in atherogenesis is the recruitment and infiltration of circulating monocytes macrophage activation subendothelial space. Atherosclerosis subsequently progresses as a unresolved inflammatory disease, particularly hypercholesterolemic conditions. Although physical exercise training has been widely accepted strategy to inhibit atherosclerosis, its impact on arterial wall inflammation phenotype function not yet directly evaluated. Thus, aim this study was investigate effects...

ABSTRACT Chronic exercise protects pancreatic beta cells from diabetogenic stress, reducing insulin secretion through unknown mechanisms. We tested the hypothesis that IL-6/mitochondrial STAT3 (pS-STAT3) axis plays a role in this protective effect. C57BL/6N mice were subjected to endurance training ahead of islet isolation and functional analysis. Similar vitro experiments performed using insulin-producing INS-1E islets untrained mice, cultured with serum trained animals treated or without...
