Abstract N-linked glycans on immunoglobulin G (IgG) have been associated with pathogenesis of diseases and the therapeutic functions antibody-based drugs; however, low-abundance species are difficult to detect. Here we show a glycomic approach detect these human IgGs using specialized microfluidic chip. We discover 20 sulfated 4 acetylated N-glycans IgGs. Using multiple reaction monitoring method, precisely quantify previously undetected low-abundance, trace even ultra-trace N-glycans. From...
Danshen or Chinese red sage (Salvia miltiorrhiza, Bunge) is used by traditional medicine (TCM) practitioners to treat neurological, cardiovascular, and cerebrovascular disorders included in some TCM formulations control epileptic seizures. In this study, acetonic crude extracts of danshen inhibited pentylenetetrazol (PTZ)-induced seizure activity zebrafish larvae. Subsequent bioassay-guided fractionation the extract resulted isolation four major tanshinones, which suppressed PTZ-induced...
Abstract Introduction – Xuebijing injection (XBJ) is a traditional Chinese herbal prescription widely used in the treatment of sepsis. This first report concerning identification XBJ constituents. In addition, to evaluate XBJ's quality, partial least square discrimination analysis (PLS‐DA) was performed on chemical fingerprint data. Objective Establish an LC‐MS method identify components for purpose quality control. Methodology Compounds were separated by HPLC using C 18 column and gradient...
To investigate the effect of supersaturation induced by micelle formation during dissolution on bioavailability itraconazole (ITZ)/Soluplus® solid dispersion. Solid dispersions prepared hot melt extrusion (HME) were compressed into tablets directly with other excipients. Dissolution behavior ITZ was studied testing and morphology micelles in media using transmission electron microscopy (TEM). Drug transferring from stomach intestine simulated to obtain a supersaturated drug solution....
Carya cathayensis Sarg. green husk (CSGH), one kind of by-product in the nut industry, contains abundant triterpenic acids. This study explores a extraction approach acid from CSGH using NBs and ultrasound-assisted based on hydrophobic natural deep eutectic solvents (HBDES). Triterpenic acids were identified UPLC-Triple-TOF/MS. The HBDES was designed via Hansen solubility parameters, consisting thymol linalool (Thy/Lin), demonstrated highest yield generated Thy/Lin, with an average size...
Abstract Background: Ba/F3 engineered cells commonly cause tumor metastasis when implanted into immune-deficient mice, resulting in reduced survival time and narrower dosing windows. To overcome this limitation, we have developed a novel approach that modifies these without compromising target pathway activation while significantly reducing their metastatic potential. This modification extends both the duration therapeutic window available for treatment interventions. Methods: We utilized...
Phytochemical investigation of Kadsura angustifolia led to the isolation and identification 26 lignans two triterpenoids, including 11 new named kadangustins A−K (1–11). The structures stereochemistry 1–11 were elucidated by analysis spectroscopic data. Except for 20, all evaluated their inhibitory activity against HIV-1. Binankadsurin A (19) showed anti-HIV with an EC50 value 3.86 µM.
Abstract Drug resistance elicited by cancer cells continue to cause huge problems world-wide, for example, tens of thousands patients are suffering from taxol-resistant human ovarian cancer. However, its biochemical mechanisms remain unclear. Sphingolipid metabolic dysregulation has been increasingly regarded as one the drug-resistant various cancers, which in turn provides potential targets overcoming resistance. In current study, a well-established LC-MS based sphingolipidomic approach was...
Abstract Background Nanoparticle-based pulmonary drug delivery systems are commonly developed and applied for drug-targeted delivery. They exhibit significant advantages compared to traditional systems. However, developing the formulation of each is a time-consuming laborious task. Results In this study, universal lung-targeting nanoparticle was designed constructed. The self-assembled micelles were composed platycodon secondary saponin, 3- O -β- d -glucopyranosyl platycodigenin 682...
