The chemokine stromal cell-derived factor 1, SDF-1, is an important regulator of leukocyte and hematopoietic precursor migration pre-B cell proliferation. receptor for CXCR4, also functions as a coreceptor T-tropic HIV-1 entry. We find that mice deficient CXCR4 die perinatally display profound defects in the nervous systems. CXCR4-deficient have severely reduced B-lymphopoiesis, myelopoiesis fetal liver, virtual absence bone marrow. However, T-lymphopoiesis unaffected. Furthermore,...

Signal transducer and activator of transcription (STAT) proteins have been shown to mediate biological actions in response cytokines. Stat3, a member the STAT family, is activated by variety cytokines, including interleukin 6 family leptin, granulocyte colony-stimulating factor, epidermal growth factor. To address function we generated mice deficient Stat3 gene targeting. No viable Stat3-deficient could be obtained from heterozygote intercross. Analysis embryos at several gestation times...

Interleukin 1 (IL 1), IL 6, and tumor necrosis factor (TNF) are typical examples of multifunctional cytokines involved in the regulation immune response, hematopoiesis, inflammation. Their functions widely overlapping but each shows its own characteristic properties. 6 was originally identified as a B cell differentiation factor, thus one major is antibody induction. Transgenic mice have provided much needed information on pathophysiological role cytokines. With transgenic mice, deregulation...

Interleukin-6 (IL-6/BSF-2/IFNβ 2) is a multifunctional cytokine that regulates the growth and differentiation of various tissues, known particularly for its role in immune response acute phase reactions. A complementary DNA encoding human IL-6 receptor (IL-6-R) has now been isolated. The IL-6-R consists 468 amino acids, including signal peptide ∼19 acids domain ∼90 similar to immunoglobulin (Ig) superfamily. cytoplasmic ∼82 lacks tyrosine/kinase domain, unlike other factor receptors.

Interleukin-6 (IL-6), leukemia inhibitory factor, oncostatin M, interleukin-11, and ciliary neurotrophic factor bind to receptor complexes that share the signal transducer gp130. Upon binding, ligands rapidly activate DNA binding of acute-phase response (APRF), a protein antigenically related p91 subunit interferon-stimulated gene factor-3 alpha (ISGF-3 alpha). These cytokines caused tyrosine phosphorylation APRF ISGF-3 p91. Protein kinases Jak family were also phosphorylated, both Jak1...

Generation and proliferation of early B-cell progenitors have been known to require stromal cell-derived molecules. A cell line, PA6, was found produce a soluble mediator, which distinct from interleukin 7 (IL-7) stem factor supported the cell-dependent pre-B-cell clone, DW34. cDNA clone encoding this DW34 growth-stimulating isolated by expression cloning. The nucleotide sequence contained single substantial open reading frame 267 nucleotides an 89-amino acid polypeptide. amino cytokine,...

Abstract Objective Interleukin‐6 (IL‐6) is a pleiotropic cytokine that regulates the immune response, inflammation, and hematopoiesis. Overproduction of IL‐6 plays pathologic roles in rheumatoid arthritis (RA), blockade may be therapeutically effective for disease. This study was undertaken to evaluate safety efficacy humanized anti–IL‐6 receptor antibody, MRA, patients with RA. Methods In multicenter, double‐blind, placebo‐controlled trial, 164 refractory RA were randomized receive either...

Abstract High levels of interleukin 6 (IL 6/B cell stimulatory factor‐2) were detected in synovial fluids from the joints patients with active rheumatoid arthritis (RA). The cells found freshly isolated fluid constitutively expressed IL6 mRNA. tissues obtained by joint biopsy also to produce vitro. Immunohistochemical analysis demonstrated that CD2 + T as well CD20 blastoid B IL6. data indicate is generated RA and its overproduction may explain local generalized symptoms RA, since can...

The role of recombinant B cell stimulatory factor 2 (BSF-2/IL-6) in the regulation growth and differentiation cells was investigated. rBSF-2 at 200 pg/ml could induce 50% maximum Ig production lymphoblastoid lines, specific activity being estimated as 5 X 10(6) U/mg. augmented PWM-induced IgM, IgG, IgA mononuclear (MNC); effect exerted by directly acting on blast to production. However, did not any activated cells. In contrast, showed a potent murine hybridoma clone, MH60.BSF2. concentration...

Abstract Objective To establish the safety and efficacy of repeat infusions tocilizumab (previously known as MRA), a humanized anti–interleukin‐6 (IL‐6) receptor antibody, alone in combination with methotrexate (MTX), for treatment rheumatoid arthritis (RA). Methods The study group comprised 359 patients active RA whom response to MTX was inadequate. During stabilization period, these received their current dose at least 4 weeks. Following stabilization, they were randomized 1 7 arms,...
