N-acylhomoserine lactones (AHLs) are signaling molecules in many quorum-sensing (QS) systems that regulate interactions between various pathogenic bacteria and their hosts. Quorum quenching by the enzymatic inactivation of AHLs holds great promise preventing treating infections, several such enzymes have been reported. In this study, we report characterization a novel AHL-degrading protein from soil bacterium Ochrobactrum sp. strain T63. This protein, termed AidH, shares no similarity with...
The Wnt/β-catenin signaling pathway plays essential roles in embryonic development and adult tissue homeostasis. Axin is a concentration-limiting factor responsible for the formation of β-catenin destruction complex. Wnt itself promotes degradation Axin. However, underlying molecular mechanism biological relevance this targeting have not been elucidated. Here, we identify SIAH1/2 (SIAH) as E3 ligase mediating Wnt-induced degradation. SIAH proteins promote ubiquitination proteasomal through...
Gene-editing technology has become a transformative tool for the precise manipulation of biological genomes and holds great significance in field animal disease-resistant breeding. Mastitis, prevalent disease husbandry, imposes substantial economic burden on global dairy industry. In this study, regulatory sequence gene editing breeding strategy successful creation gene-edited (GED) goats with enhanced mastitis resistance using ISDra2-TnpB system as model is proposed. This included targeted...
The Gabija system is a newly discovered bacterial immune that consists of GajA and GajB. Here we report the cryo-EM structure complex from Bacillus cereus VD045 at 3.6 Å, which provides direct evidence interactions between an octameric ring with four OLD nucleases family protein, while GajB belongs to SF1 helicases. has sequence-specific DNA nuclease activity prefers circular rather than linear as substrate, its more sensitive concentrations change nucleotides compared alone. Our data...
CDC73/Parafibromin is a critical component of the Paf1 complex (PAF1C), which involved in transcriptional elongation and histone modifications. Mutations human CDC73/HRPT2 gene are associated with hyperparathyroidism-jaw tumor (HPT-JT) syndrome, an autosomal dominant disorder. CDC73/parafibromin was initially recognized as suppressor by inhibiting cell proliferation via repression cyclin D1 c-myc genes. In recent years, it has also shown oncogenic features activating canonical Wnt/β-catenin...
Physic nut (Jatropha curcas L) seed oil is a natural resource for the alternative production of fossil fuel. Seed mainly depended on yield, which was restricted by low ratio staminate flowers to pistillate flowers. Further, mechanism physic flower sex differentiation has not been fully understood yet. Quantitative Real Time—Polymerase Chain Reaction reliable and widely used technique quantify gene expression pattern in biological samples. However, accuracy qRT-PCR, appropriate reference...
Significance Type I restriction-modification (R-M) enzymes are large molecular machines found in the majority of bacterial species. They can add methylation modifications to self-DNA and degrade invading unmodified DNA. The lack high-resolution structures type R-M complexes impairs our understanding mechanism subunit assembly conformational transition. Here we report first structure MTase complex its “open” conformation, including one DNA-recognition subunit, two DNA-modification subunits,...
The Lim domain binding proteins (LDB1 and LDB2 in human Chip Drosophila ) play critical roles cell fate decisions through partnership with multiple Lim-homeobox Lim-only diverse developmental systems including cardiogenesis, neurogenesis, hematopoiesis. In mammalian erythroid cells, LDB1 dimerization supports long-range connections between enhancers genes involved erythropoiesis, the β-globin genes. Single-stranded DNA (SSBPs) interact specifically LDB/Chip conserved (LCCD) of LDB stabilize...
Type I DNA methyltransferases contain one specificity subunit (HsdS) and two modification subunits (HsdM). The electron microscopy model of M.EcoKI-M2S1 methyltransferase shows a reasonable closed state this clamp-like enzyme, but the structure open is still unclear. 1.95 Å crystal from Thermoanaerobacter tengcongensis (TTE-HsdS) an unreported form inter-domain orientation subunit. Based on TTE-HsdS M.EcoKI-M2S1, we constructed potential type methyltransferase. Mutational studies indicated...
Although urea and guanidine hydrochloride are commonly used to denature proteins, the molecular underpinnings of this process have remained unclear for a century. To address question, crystal structures β-catenin were determined at various concentrations. These contained least 105 unique positions that occupied by molecules, each which interacted with protein primarily via hydrogen bonds. Hydrogen-bond competition experiments showed denaturing effects neutralized when polyethylene glycol was...
Abstract RecF is a principal member of the pathway. It interacts with RecO and RecR to initiate homologous recombination by loading RecA recombinases on single-stranded DNA displacing DNA-binding proteins. As an ATP-binding cassette ATPase, exhibits ATP-dependent dimerization structural homology Rad50 SMC However, mechanism action pattern remains unclear. Here, We determined three crystal structures TTERecF, TTERecF-ATP TTERecF-ATPɤS from Thermoanaerobacter tengcongensis that reveal novel...
A one-pot synthesis of 1,3-diyne-tethered trifluoromethylcyclopropanes starting from 2-CF3-3,5-diyne-1-enes and sulfur ylides via a ylide mediated cyclopropanation DBU-mediated epimerization sequence is described in this work. This process highly diastereoselective with broad substrate scope. Moreover, series synthetic transformations based on the diyne moieties were conducted smoothly, affording cyclopropanes featuring trifluoromethyl-substituted all-carbon quaternary centers.
Rock thermal physical properties play a crucial role in understanding deep conditions, modeling the structure of lithosphere, and discovering evolutionary history sedimentary basins. Recent advancements geothermal exploration, particularly identification high-temperature resources Datong Basin, Shanxi, China, have opened new possibilities. This study aims to characterize rocks explore factors influencing conductivity basins hosting resources. A total 70 groups rock samples were collected...
The mevalonate pathway of isoprenoid biosynthesis is the most chemically diverse in nature. This produces isopentenyl diphosphate,1 fundamental five-carbon building block for over 23,000 compounds,2 such as sterols,3 caroteniods,4 dolichols,5 ubiquinone,6 and some prominent classes prenylated proteins.7 These compounds are essential cell growth differentiation, gene expression, protein glycosylation, cytoskeletal assembly. In addition, posttranslational modification proteins involved...
RecR is an important recombination mediator protein in the RecFOR pathway. together with RecO and RecF facilitates RecA nucleoprotein filament formation homologous pairing. Structural biochemical studies of Thermoanaerobacter tengcongensis (TTERecR) its series mutants revealed that TTERecR uses N-N dimer as a basic functional unit to interact TTERecO monomer. Two dimers form ring-shaped tetramer via interaction between their C-terminal regions. The result crystallization only. Hydrophobic...
