Background: Metabolic alterations provide substrates that influence chromatin structure to regulate gene expression determines cell function in health and disease. Heightened proliferation of smooth muscle cells (SMC) leading the formation a neointima is feature pulmonary arterial hypertension (PAH) systemic vascular Increased glycolysis linked proliferative phenotype these SMC. Methods: RNA sequencing was applied SMC (PASMC) from PAH patients with without BMPR2 (bone morphogenetic receptor...

The purpose of this study was to determine whether implementation public access defibrillation (PAD) improves the outcome after out-of-hospital cardiac arrest (OHCA) in school-age children at national level. We conducted a prospective, nationwide, population-based Japanese Utstein registry consecutive OHCA cases elementary and middle school (7–15 years age) who had bystander-witnessed presumed origin during 2005–09 received pre-hospital resuscitation by emergency responders. primary endpoint...

We previously reported heightened expression of the human endogenous retroviral protein HERV-K deoxyuridine triphosphate nucleotidohydrolase (dUTPase) in circulating monocytes and pulmonary arterial (PA) adventitial macrophages patients with PA hypertension (PAH). Furthermore, recombinant dUTPase increased IL-6 endothelial cells (PAECs) caused rats. Here we show that overexpressing dUTPase, as opposed to GFP, can release extracellular vesicles (EVs) cause mice association mesenchymal...

Rationale: The role of neutrophils and their extracellular vesicles (EVs) in the pathogenesis pulmonary arterial hypertension is unclear. Objectives: To relate functional abnormalities EVs to mechanisms uncovered by proteomic transcriptomic profiling. Methods: Production elastase, release traps, adhesion, migration were assessed from patients with control subjects. Proteomic analyses applied explain perturbations, data used find underlying mechanisms. CD66b-specific neutrophil isolated...

Abstract Background It is well-established that patients with pulmonary arterial hypertension (PAH) exhibit increased recruitment of circulating monocytes to their arteries. However, it remains unclear whether these have intrinsic abnormalities contribute and PAH pathogenesis. This study aimed characterize the gene expression profiles classical, intermediate, non-classical assess maturation trajectory in idiopathic (I) compared control subjects. Additionally, sought explore relationship...

We tested the hypothesis that phenotypically modulated smooth muscle cells (SMCs) and related inflammation are associated with progression of experimental occlusive pulmonary vascular disease (PVD). Occlusive PVD was induced by combined exposure to a endothelial growth factor receptor tyrosine kinase inhibitor Sugen 5416 hypobaric hypoxia for 3 weeks in rats, which were then returned ambient air. Hemodynamic, morphometric, immunohistochemical studies, as well gene expression analyses,...

Abstract Pulmonary arterial hypertension (PAH) is a progressive disease in which pulmonary (PA) endothelial cell (EC) dysfunction associated with unrepaired DNA damage. BMPR2 the most common genetic cause of PAH. We report that human PAEC reduced have persistent damage room air after hypoxia (reoxygenation), as do mice EC-specific deletion Bmpr2 (EC- -/- ) and hypertension. Similar findings are observed loss sensor ATM , Atm deleted EC ). Gene expression analysis EC- lung reveals Foxf1...

It remains unknown whether current disease-targeting therapy can histologically reverse obstructive pulmonary vasculopathy and how the timing of influences antiremodeling effects compound. We test hypothesis that a novel endothelin receptor antagonist macitentan reverses early and/or late stages occlusive vascular disease (PVD) in rats. Rats with arterial hypertension (PAH), which were produced by combined exposure to endothelial growth factor inhibitor Sugen 5416 hypobaric hypoxia for 3 wk,...

Abstract Background Patients with pulmonary arterial hypertension (PAH) carrying bone morphogenetic protein receptor type 2 ( Bmpr2 ) mutations present earlier severe hemodynamic compromise and have poorer survival outcomes than those without mutation. The mechanism underlying the worsening clinical phenotype of PAH has been largely unaddressed in rat models (PH) because difficulty reproducing progressive PH mice genetic modification rats. We tested whether a clinically-relevant mutation...

Factors initiating senile delirium were examined in 129 elderly inpatients (65 years or older). Sixty-eight patients males and 61 females, with a mean age of 76.3 years. Delirium developed most cases on the first two days admission hospital, appeared to be key factor precipitating about 30% patients. resolved improved 80% patients, but usually persisted dementia. Senile tended reappear repeatedly whose episode lasted for more than 2 weeks, was associated dementia, had prior history delirium.
