We analyzed survival and causes of death among 163 patients with primary hemochromatosis diagnosed between 1959 1983. The mean follow-up period was 10.5 +/- 5.6 years (+/- S.D.). Cumulative 92 per cent at 5 years, 76 10 59 15 49 20 years. Life expectancy reduced in cirrhosis the liver as compared those without (P less than or equal to 0.05), diabetes mellitus 0.002), who could not be depleted iron during first 18 months venesection therapy 0.001). Prognosis influenced by sex 0.5). Patients...

Chemotherapeutic drugs cause DNA damage and kill cancer cells mainly by apoptosis. p53 mediates apoptosis after damage. To explore the pathway of p53-dependent cell death, we investigated if is mediated CD95 (APO-1/Fas) receptor/ligand system. We hepatoma, gastric cancer, colon breast lines upon treatment with different anticancer agents known to act via accumulation. Cisplatin, mitomycin, methotrexate, mitoxantrone, doxorubicin, bleomycin at concentrations present in sera patients during...

Chemotherapeutic drugs are cytotoxic by induction of apoptosis in drug-sensitive cells. We investigated the mechanism bleomycin-induced cytotoxicity hepatoma At concentrations present sera patients during therapy, bleomycin induced transient accumulation nuclear wild-type (wt) p53 and upregulated expression cell surface CD95 (APO-1/Fas) receptor cells carrying wt (HepG2). Bleomycin did not increase with mutated (Huh7) or which were p53-/- (Hep3B). In addition, sensitivity towards...

Apoptosis occurs in the normal liver and various forms of disease. The CD95 (APO-1/Fas) (CD95) receptor mediates apoptosis, cells animal models are acutely sensitive to apoptosis initiated by this receptor. We have used primary human hepatocytes as a model system investigate CD95-mediated apoptotic damage. Treatment fresh with low concentrations agonistic antibodies against resulted > 95% cultured within 4 7.5 h. Immunohistology panel explanted tissues revealed that livers (n = 5)...

Wilson's disease is a rare inborn related to copper storage, leading liver cirrhosis and neuropsychological deterioration. Clinical data on larger cohorts are limited owing low frequency. We performed retrospective analysis of 163 patients with disease, examined at the University Heidelberg, Germany, determine clinical presentation, diagnostic course long-term outcome.Diagnostic criteria for non-caeruloplasmin-bound serum copper, caeruloplasmin, 24-h urinary excretion, content, presence...

Acute cholecystitis is a common disease, and laparoscopic surgery the standard of care.Optimal timing for acute remains controversial: either early shortly after hospital admission or delayed elective conservative treatment with antibiotics. The ACDC ("Acute Cholecystitis-early versus antibiotic therapy Delayed Cholecystectomy") study randomized, prospective, open-label, parallel group trial. Patients were randomly assigned to receive immediate within 24 hours (group ILC) initial treatment,...

When [14C]oleate-bovine serum albumin complexes were incubated in vitro with rat liver plasma membranes (LPM), specific, saturable binding of oleate to the was observed. Maximal heat-sensitive (i.e., specific) 3.2 nmol/mg membrane protein. Oleate-agarose affinity chromatography Triton X-100-solubilized LPM used isolate a single 40-kDa protein high for oleate. On gel filtration, comigrated various fatty acids but not [14C]bilirubin, [35S]sulfobromophthalein, [14C]taurocholate,...

We recently showed that mucus from patients with ulcerative colitis, a chronic inflammatory disorder of the colon, is characterized by low level phosphatidylcholine (PC) while clinical studies reveal therapeutic addition PC using slow release preparations beneficial. The positive role in this disease still elusive. Here we tested hypothesis exogenous application has anti-inflammatory properties three model systems. First, human Caco-2 cells were treated tumor necrosis factor-α (TNF-α) to...

Leptin is a pleiotropic hormone believed to regulate body weight. Its function in wasting during inflammatory disease humans unknown. We studied the effect of repeated tumor necrosis factor (TNF) infusion on serum leptin levels six patients with solid tumors. TNF day 1 resulted an increase from 3.1 (sem ± 0.28) ng/mL 5.2 0.6) after 12 h (P < 0.001). The returned baseline within 24 h. Similar results were obtained when was infused subsequent days. study shows that are under control TNF.

Helicobacter pylori infection is associated with chronic gastritis, peptic ulceration, and gastric carcinoma. The potential role of CD95-mediated apoptosis was investigated in a panel biopsies obtained from patients H. pylori-associated gastritis (n = 29) noninfected normal mucosa 10). Immunohistochemistry revealed increased CD95 receptor expression epithelial lamina propria cells gastritis. By situ hybridization, ligand mRNA absent or low but expressed at high levels lymphocytes and,...

Abstract Background: ATP-binding cassette (ABC) transporters are involved in many physiologic processes, such as lipid transport, sterol homeostasis, immune mechanisms, and drug cause various human inherited diseases. Thus, the analysis of ABC transporter mRNA expression profiles for basic research, especially field metabolism, clinical diagnosis, monitoring effects is great interest. Methods: We have developed a rapid, accurate, highly sensitive real-time reverse transcription-PCR (RT-PCR)...

Since iron-regulatory and antimicrobial peptide hormone hepcidin was originally isolated from human urine we investigated expression as well zonal cellular localization mammalian kidney developed an ELISA assay to analyze concentrations serum urine.
