In order to quantify autoantibodies in the sera of patients with autoimmune disease, we have created a microarray-based immunoassay that allows simultaneous analysis 18 known autoantigens. The microarrays contain serial dilutions various antigens, thereby allowing accurate determination autoantibody titer using minimal amounts serum. assay is very sensitive and highly specific: as little 40 fg protein standard can be detected or no cross-reactivity nonspecific proteins. signal intensities...
The pestivirus genome encodes a single polyprotein which is subject to co- and posttranslational processing by cellular viral proteases. map positions of all virus-encoded proteins are known with the exception hypothetical peptide (p?) interlinks glycoprotein E2 nonstructural protein NS2-3 approximately between amino acid 1060 1130. Expression studies recombinant vaccinia viruses bearing set C-terminally truncated E2-p?-NS2-encoding sequences derived from bovine diarrhea virus (BVDV) strain...
purpose. To determine efflux systems of the outer blood–retina barrier (oBRB) and compare oBRB with blood–brain (BBB). methods. Porcine structure transport characteristics freshly dissected intact tissue sheets were investigated scanning electron microscopy, immunocytochemistry, vital dye labeling, pharmacological agents, using HPLC/mass spectrometry. drug permeation across BBB, three different used: (1) in a two-chamber device vitro; (2) an vitro BBB model composed purified astrocytes brain...
The single-stranded genomic RNA of pestiviruses is positive polarity and encompasses one large open reading frame about 4,000 codons. resulting polyprotein processed co- posttranslationally by virus-encoded host cell proteases to give rise the mature viral proteins. A serine protease residing in nonstructural (NS) protein NS3 (p80) has been shown be essential for release NS proteins located downstream NS3. In this report protease-dependent cleavage sites bovine diarrhea virus (BVDV) strain...
Protein microarray technology facilitates the detection and quantification of hundreds binding reactions in one reaction from a minute amount sample. Proof-of-concept studies have shown that set-up sensitive assay systems based on protein arrays is possible, however, lack specific capture reagents limits their use. Therefore, generation characterisation molecules key topics for development array systems. Recombinant antibody technologies, such as HuCAL (human combinatorial library;...
Background: Liver impairment, ranging from steatosis to cirrhosis, is frequent in cystic fibrosis (CF) patients and becoming increasingly significant due their improved life expectancy. One aspect of hepatic alterations caused by increased fecal loss the essential nutrient choline, following enterohepatic bile phosphatidylcholine (PC) cycle impairment. Hepatic PC synthesis, both de novo via phosphatidylethanolamine-N-methyl-transferase (PEMT), for very low-density lipoprotein (VLDL)...
Major histocompatibility complex (MHC) class II molecules present peptide antigens to CD4 + ‐T cells. These heterogeneous peptides are derived from internalized exogenous proteins or endogenous membrane that processed by the antigen‐presenting cell. Peptides bound MHC in an extended conformation and extend out of binding groove. The aim this study was estimate influence every amino acid all possible undecapeptide amides (2.048×10 14 individuals) on human MHC‐DRB1 * 0101 (HLA‐DR1) identify...
Surfactant reduces surface tension at pulmonary air-liquid interfaces. Although its major component is dipalmitoyl-phosphatidylcholine (PC16:0/16:0), other PC species, principally palmitoylmyristoyl-PC, palmitoylpalmitoleoyl-PC, and palmitoyloleoyl-PC, are integral components of surfactant. The composition metabolism species depend on development, respiratory rate, pathologic alterations, which have largely been investigated in animals using radiolabeled precursors. Recent advances mass...
ABSTRACT Pestiviruses represent the first RNA viruses for which recombination with cellular protein-coding sequences has been reported. As a result of such recombinations cytopathogenic (cp) pestiviruses can develop from noncytopathogenic (noncp) viruses. In case bovine viral diarrhea virus (BVDV), generation cp mutants is linked to induction lethal syndrome mucosal disease (MD) in cattle. The BVDV JaCP was isolated an animal had come down MD. genome contains novel kind insertion (LC3*)...
ABSTRACT Bovine viral diarrhea virus (BVDV) isolates can either be cytopathogenic (cp) or noncytopathogenic (noncp). While both biotypes express the nonstructural protein NS2-3, generation of NS3 strictly correlates with cp phenotype. The production is usually caused by specific genome alterations, which were found to due RNA recombination. Molecular analyses BVDV strain Oregon revealed that it does not possess such alterations but nevertheless able generate via processing NS2-3. serine...
