We report here that 4-dibenzo[a,d]cyclohepten-5-ylidene-1-[4-(2H-tetrazol-5-yl)-butyl]-piperidine (AT-56) is an orally active and selective inhibitor of lipocalin-type prostaglandin (PG) D synthase (L-PGDS). AT-56 inhibited human mouse L-PGDSs in a concentration (3-250 microm)-dependent manner but did not affect the activities hematopoietic PGD (H-PGDS), cyclooxygenase-1 -2, microsomal PGE synthase-1. L-PGDS activity competitive against substrate PGH(2) (K(m) = 14 microm) with K(i) value 75...

Abstract Several dog allergens cause allergic reactions in humans worldwide. Seven distinct allergens, designated Canis familiaris allergen 1 to 7 (Can f 1–Can 7), have been identified thus far. Can 6 shows high sequence similarity and cross-reactivity with Fel d 4 Equ c 1, major cat horse respectively. This study was conducted on the allergenic epitopes of based its structural characterization. We demonstrated that sera from 18 out 38 (47%) dog-sensitized patients reacted recombinant...

In higher eukaryotic cells, DNA molecules are present as chromatin fibers, complexes of with various types proteins; fibers highly condensed in metaphase chromosomes during mitosis. Although the formation chromosome structure is essential for equal segregation replicated chromosomal into daughter mechanism involved organization poorly understood. To identify proteins and/or maintenance chromosomes, we examined that dissociated from isolated human by 0.4 m NaCl treatment; this treatment led...

In vitro folding of disulfide‐containing proteins is generally regulated by redox molecules, such as glutathione. However, the role cross‐disulfide‐linked species formed between molecule and protein a intermediate in mechanism poorly understood. present study, we investigated effect charge on disulfide‐coupled folding. Several types aliphatic thiol compounds including glutathione were examined for disulfide‐containing‐proteins, lysozyme prouroguanylin. The results indicate that positive its...

Heat-stable enterotoxin (STa) produced by Enterotoxigenic E. coli is responsible for causing acute diarrhea in infants developing countries. However, the chemical synthesis of STa peptides with native conformation and correct intra-molecular disulfide bonds a major hurdle vaccine development. To address this issue, we herein report on design preparation analogs convenient method obtaining molecules conformation. develop an vaccine, focused structure type II β-turn molecule introduced D-Lys...

Heat-stable enterotoxin (STa) produced by enterotoxigenic E. coli causes acute diarrhea and also can be used as a specific probe for colorectal cancer cells. STa contains three intra-molecular disulfide bonds (C1–C4, C2–C5, C3–C6 connectivity). The chemical synthesis of provided not only the native type but topological isomer that had pairings. Interestingly, activity was approximately 1/10–1/2 STa. To further investigate bioactive conformation this molecule regulation disulfide-coupled...

Investigations of protein folding have largely involved the use disulfide-containing proteins, since disulfide-coupled proteins allows intermediates to be trapped and their conformations determined. However, studies mechanisms mid-size face several problems, one which is that detecting difficult. Therefore, solve this issue, a novel peptide reagent, maleimidohexanoyl-Arg5-Tyr-NH2, was designed applied detection model proteins. BPTI chosen as small estimate ability reagent detect...

Proopiomelanocortin (POMC) is a precursor protein of several peptide hormones, such as ACTH and β-endorphin. Almost all the hormones in POMC have been drastically investigated terms their biological activities. However, activity joining region (JP) unknown. Therefore, to explore JP, sequence analyses mammalian were performed. We found an -Arg-Gly-Asp- (RGD) motif species, porcine, suggesting that JP has cell adhesion activity. To validate this hypothesis, activities synthetic porcine...

Cocoonase is folded in the form of a zymogen precursor protein (prococoonase) with assistance propeptide region. To investigate role sequence on disulfide-coupled folding cocoonase and prococoonase, amino acid residues at degradation sites during refolding auto-processing reactions were determined by mass spectrometric analyses mutated to suppress numerous that occur reactions. In addition, Lys8 residue region was also estimate whether entire absolutely required for activation cocoonase....

Abstract Congenital contractual arachnodactyly (CCA) is a genetic connective tissue disorder that characterized by arachnodactyly, kyphoscoliosis, marfanoid habitus, and crumpled ears. We report case of boy with suspected Marfan syndrome. Genetic analysis revealed c.3207_3217+9del in heterozygote form the fibrillin-2 ( FBN2) gene. This patient was diagnosed CCA based on his phenotype, pathogenicity this variant classified according to cDNA protein modeling.

To investigate the substrate recognition mechanism of trypsin-like protease cocoonase (CCN), mutational analyses were conducted at key sites, Asp187 and Ser188, their effects on specificity enzymatic activity evaluated. Mutants with substitution exhibited a significant reduction in catalytic compared wild-type enzyme, whereas Ser188 mutants displayed comparatively minor effect activity. This indicates that plays crucial role function, serves complementary recognition. Interestingly, to Glu...
