ABSTRACT Multiple myeloma is a plasma cell malignancy characterized by an abnormal increase in monoclonal immunoglobulins. Despite significant advances treatment, some patients progress to more aggressive forms of multiple myeloma, including extramedullary disease or leukemia. Although the exact molecular mechanisms are not known, several studies have confirmed involvement small extracellular vesicle‐enriched microRNAs progression. Therefore, we performed expression profiling these molecules...

Rottlerin, a polyphenol isolated from Mallotus Philippinensis, has been recently used as selective inhibitor of PKC δ , although it can inhibit many kinases and several biological effects. Among them, we found that Rottlerin inhibits the Nuclear Factor κ B (NF B), activated by either phorbol esters or H 2 O . Because redox sensitivity NF on basis antioxidant property, hypothesized could prevent activation acting free radicals scavenger, other natural polyphenols. The current study confirms...

Polycyclic aromatic hydrocarbons (PAHs) are widespread environmental contaminants that interact in a complex manner with both the aryl hydrocarbon receptor (AhR) and estrogen receptors (ER). Their potential endocrine-disrupting activities may depend on inhibitory AhR-ER cross-talk AhR-dependent metabolic production of estrogenic PAH metabolites. Here, we analyzed impact AhR estrogen-like effects PAHs, such as benzo[a]pyrene (BaP), particular, control cell cycle progression/cell...

Abstract We verified the hypothesis suggesting modulation of effects sodium butyrate (NaBt) by ω–3 or ω–6 PUFAs. Comparing response human colon epithelial cell lines fetal (FHC) and adenocarcinoma (HT‐29, HCT‐116) origin, we detected significant differences in proliferation, differentiation apoptotic to treatment NaBt, arachidonic docosahexaenoic acids their combination. While FHC HT‐29 cells NaBt induced G0/G1 arrest, low level apoptosis, HCT‐116 G2/M no high degree apoptosis were detected....

The assay employing firefly luciferase as the end-point reporter is one of most popular gene systems. However, physiological conditions cells may affect expression, which makes an assessment cell viability desirable. Estimates be based on different principles. We tested for correlations between various assessments, including luminescent determination adenosine triphosphate in whole-cell lysate, and activity pluripotent embryonic colon adenocarcinoma cells. Luciferase lysate from both lines...

Sphingolipids (SLs), glycosphingolipids (GSLs), and eicosanoids are bioactive lipids, which play important roles in the etiology of various diseases, including cancer. However, their content cancer cells, particular exosomes derived from tumor remain insufficiently characterized. In this study, we evaluated alterations SL GSL levels transformed cells exosomes, using comparative HPLC-MS/MS analysis parental human bronchial epithelial HBEC-12KT derivative, benzo[a]pyrene-transformed...

Abstract Our recent study follows up an earlier one which demonstrated hypericin‐mediated photocytotoxic effects on HT‐29 adenocarcinoma cells by light fractionation with a longer dark pause between two unequal doses (Sackova, A. [2005] Photochem. Photobiol. 81 , 1411–1416). Here, we present closer events invoked sublethal dose (1 J cm −2 ) during the period of 6 h that is sufficient to invoke resistance second lethal (11 ). First, proved h, but not 1 resulted in better cell survival...
