The importance of gut microbiota in human health and pathophysiology is undisputable. Despite the abundance metagenomics data, functional dynamics disease remain elusive. Urolithin A (UroA), a major microbial metabolite derived from polyphenolics berries pomegranate fruits displays anti-inflammatory, anti-oxidative, anti-ageing activities. Here, we show that UroA its potent synthetic analogue (UAS03) significantly enhance barrier function inhibit unwarranted inflammation. We demonstrate...
A hydrogel binds to inflamed tissues, delivering therapeutics locally and reducing systemic drug exposure in mouse models of inflammatory bowel disease.
Mopping up: Phase-selective gelators were synthesized from sugar alcohols using biocatalysis. The exhibited an unprecedented ability to phase-selectively gel only organic liquids including crude-oil fractions in the presence of water at room temperature. Oil can be recovered gelled phase by heating and distillation. Thus this class materials may useful recovering oil spills.
Abstract Local delivery of therapeutics for the treatment inflammatory arthritis (IA) is limited by short intra-articular half-lives. Since IA severity often fluctuates over time, a local drug method that titrates release to activity would represent an attractive paradigm in therapy. Here we report development hydrogel platform exhibits disassembly and controlled concentration enzymes expressed during flares. In vitro, loaded with triamcinolone acetonide (TA) releases on-demand upon exposure...
We report a novel approach for the controlled delivery of an antiinflammatory, chemopreventive drug by enzyme-triggered release mechanism via degradation encapsulated hydrogels. The hydro- and organogelators are synthesized in high yields from renewable resources using regioselective enzyme catalysis, known antiinflammatory drug, i.e., curcumin, is used model study. occurred at physiological temperature, control rate achieved manipulating concentration and/or temperature. byproducts formed...
To date, a wide range of industrial materials such as solvents, fuels, synthetic fibers, and chemical products are being manufactured from petroleum resources. However, rapid depletion fossil resources is encouraging current future chemists to orient their research toward designing safer chemicals, products, processes renewable feedstock with an increased awareness environmental impact. Advances in genetics, biotechnology, process chemistry, engineering leading new manufacturing concept for...
Various organic self-assembled systems such as low-molecular-weight gels and liquid crystals have been utilized efficient hosts for in situ synthesis stabilization of gold nanoparticles using vitamin-C-based amphiphiles.
Tacrolimus-encapsulated hydrogels improve long-term survival of a vascularized composite allograft.
Temporal control over self-assembly process is a desirable trait in the quest towards adaptable and controllable materials. The ability to devise synthetic ways growth, as well decay of materials has long been property which only biological systems could perform seamlessly. A common strategy works on principles such chemical fuel-driven temporal profile not completely realized synthetically. Here we show, filled this dearth by showing that fuel driven self-assembling system can be grown...
Monitoring the location, distribution and long-term engraftment of administered cells is critical for demonstrating success a cell therapy. Among available imaging-based tracking tools, magnetic resonance imaging (MRI) advantageous due to its noninvasiveness, deep penetration, high spatial resolution. While in preclinical models via internalized MRI contrast agents (iron oxide nanoparticles, IO-NPs) widely used method, IO-NPs suffer from low iron content per particle, uptake nonphagocytotic...
Numerous studies signify that diets rich in phytochemicals offer many beneficial functions specifically during pathologic conditions, yet their effects are often not uniform due to inter-individual variation. The host indigenous gut microbiota and modifications of dietary have emerged as factors greatly influence the efficacy phytoceutical-based intervention. Here, we investigated biological activities one such active microbial metabolite, Urolithin A (UA or...
Abstract Pancreatic ductal adenocarcinoma (PDAC) is an aggressive malignancy and highly resistant to standard treatment regimens. Targeted therapies against KRAS, a mutation present in overwhelming majority of PDAC cases, have been largely ineffective. However, inhibition downstream components the KRAS signaling cascade provides promising therapeutic targets management warrants further exploration. Here, we investigated Urolithin A (Uro A), novel natural compound derived from pomegranates,...
Delivering drugs directly to the inflamed intestinal sites treat inflammatory bowel disease (IBD), particularly Crohn's and ulcerative colitis, is highly challenging. Recent advances in colitis therapy medications are expanding opportunities for improving local on-site drug availability by minimising associated systemic side-effects. Drug delivery with targeted carrier systems has shown potential increase site-specificity, stability, therapeutic efficacy. Herein, we report development of a...
The survival rate of colorectal cancer patients is adversely affected by the selection tumors resistant to conventional anti-cancer drugs such as 5-fluorouracil (5FU).Although there mounting evidence that commensal gut microbiota essential for effective colon treatment, detailed molecular mechanisms and role microbial metabolites remain elusive. The goal this study decipher impact metabolite, urolithin A (UroA) its structural analogue, UAS03 on reversal 5FU-resistant (5FUR) cancers.Methods:...
New urea containing gels have been used to prepare and stabilise gold nanoparticles by in situ reduction.
Covalently conjugated sialyl Lewis X (SLeX) on the mesenchymal stem cell (MSC) surface through a biotin−streptavidin bridge imparts leukocyte-like rolling characteristics without altering phenotype and multilineage differentiation potential. We demonstrate that conjugation of SLeX MSC is stable, versatile, induces robust response P-selectin coated substrates. These results indicate potential to increase targeting efficiency any type specific tissue.
