This study reports a facile method for the synthesis of multi-enzyme co-embedded organic-inorganic hybrid nanoflowers, using glucose oxidase (GOx) and horseradish peroxidase (HRP) as organic components, Cu3(PO4)2 · 3H2O inorganic component. The synthesized nanoflowers enable combination two-enzyme cascade reaction in one step, which GOx component oxidizes to generate H2O2, then reacts with adjacent HRP on oxidize chromogenic substrates, resulting an apparent color change. Given close...
3-Phenylthiophene-based water-soluble copolythiophenes (CPT1) were designed for colorimetric and fluorometric detection of lipopolysaccharide (LPS). The sensor (CPT1-C) shows a high selectivity to LPS in the presence other negatively charged bioanalytes as well an extreme sensitivity with limit at picomolar level, which is lowest ever achieved among all synthetic sensors available thus far. Significantly, sensing interaction can be apparently observed by naked eyes, presents great advantage...
The aim of our study was to investigate the relationship among gut microbiota community, metabolite profiles and thyroid carcinoma (TC). First, 30 TC patients 35 healthy controls (HCs) fecal samples were applied characterize microbial community using 16S rRNA gene sequencing. Differential compositions observed, with significant enrichment 19 depletion 8 genera in compared those HCs ( Q value <0.05), some correlated various clinical parameters, such as lipoprotein A apolipoprotein B....
Cholangiocarcinoma (CCA) is a malignant tumor that resistant to chemotherapy, so new therapeutic agents are needed. Allicin which rapidly converted from allin by allinase, one of the most biologically active compounds in freshly crushed garlic and has been shown have strong anti-tumor effects. Our aim was explore molecular mechanism allicin affects cell proliferation invasion CCA.Cell viability apoptosis were measured using CCK-8 assay, colony formation flow cytometry. Cell migration...
Proximity labeling has emerged as a powerful strategy for interrogating cell-cell interactions. However, the nanometer-scale radius impedes use of current methods indirect cell communications and makes recording spatial organization in tissue samples difficult. Here, we develop quinone methide–assisted identification (QMID), chemical with matching dimension. The activating enzyme is installed on surface bait cells, which produces QM electrophiles that can diffuse across micrometers label...
Affinity labeling of biomacromolecules is vital for bioimaging and functional studies. However, affinity probes recognizing glycans with high specificity remain scarce. Here we report the development glycan recombinant binders (GRABs) based on mutant bacterial sialidases, which are enzymatically inactive but preserve stringent sialoglycan substrates. By mutating a key catalytic residue Streptococcus pneumoniae neuraminidase A (SpNanA) Ruminococcus gnavus H (RgNanH), develop GRAB-Sia...
Electrostatic interaction between streptomycin and mercaptoacetic acid modified gold nanoparticles can be used for a facile reliable probe with high sensitivity selectivity.
Summary Colorectal cancer (CRC) is a common disease worldwide that strongly associated with the gut microbiota. However, little known regarding microbiota after surgical treatment. 16S rRNA gene sequencing was used to evaluate differences in among colorectal adenoma patients, CRC postoperative patients and healthy controls by comparing diversity, overall composition taxonomic signature abundance. The of developed accordance adenoma‐carcinoma sequence, impressive shifts before or during...
Background: Drugs can alter the gut microbiota structure, and dysbiosis in turn is correlated with drug side effects through intestinal endotoxemia hypothesis. Whether antithyroid drugs (including methimazole propylthiouracil) cause whether induced liver injury unknown. Methods: Initial Graves’ disease patients were randomly divided into group ( n = 20) propylthiouracil followed up every 2 weeks; 50 healthy controls also included. The structure function of compared from cross sectional...
The β3-adrenoceptor (β3-AR) is implicated in cardiac remodeling. Since metabolic dysfunction due to loss of mitochondria plays an important role heart diseases, we examined the effects β3-AR on mitochondrial biogenesis and energy metabolism atrial fibrillation (AF).Atrial was created by rapid pacing adult rabbits. Rabbits were randomly divided into 4 groups: control, (P7), antagonist (L748337), agonist (BRL37344) groups. Atrial effective refractory period (AERP) AF induction rate measured....
This study aimed to assess the effectiveness of inhibiting cholesterol acyltransferase 1 (ACAT-1) in chimeric antigen receptor T (CAR-T) cells on potentiating antitumor response against mesothelin (MSLN)-expressing pancreatic carcinoma (PC) cells. We engineered ACAT-1-inhibited CAR-T (CAR-T-1847 and CAR-T-1848) using targeting MSLN CAR lentiviral vector small interfering RNA (siRNA) conserved region ACAT-1 gene, characterized efficacy these modified terms cytotoxicity cytokine release both...
It is known that an imbalance in the intestinal f lora plays a crucial role colorectal cancer (CRC), but effect of food consumption patterns on types flora remains to be clarified. We aimed analyze associations between intake and healthy CRC individuals.Food data were recorded using Food Frequency Questionnaire (FFQ). The composition diversity detected by 16S rRNA gene sequencing, analyzed R version 3.1.1 software.Higher red meat or pickled foods, lower white meat, fruits, vegetables, beans,...
OBJECTIVE: Intestinal microbiota plays a vital role in the pathogenesis of colorectal cancer (CRC), which is crucial for assessing risk and prognosis CRC. Most studies regarding human gut mainly based on feces, but exact composition vary significantly due to fecal easily affected by many factors. We aim evaluate whether intestinal lavage fluid (IVF) better substitution mirroring microbiota. METHODS: performed 16S rRNA gene analysis IVF samples from 30 CRC patients 25 healthy individuals,...
Background and Aims Oral xerostomia remains one of the most common complications differentiated thyroid carcinoma patients (DTC) after radioiodine therapy (RAI). Environmental factors in etiology are largely unknown. We aimed to characterize oral microbiota signatures related biological functions associated with identify environmental affecting them. Methods Saliva was collected from 30 DTC (XAs), 32 without (indicated as non-XAs) following RAI total thyroidectomy, 40 healthy people (HCs)...
