Abstract Interleukin‐6 (IL‐6) is a multifunctional cytokine produced by macrophages, T cells, B endothelial cells and tumour cells. able to promote growth upregulating anti‐apoptotic angiogenic proteins in In murine models it has been demonstrated that antibodies against IL‐6 diminish growth. Several reports have highlighted the prognostic importance of e.g. , prostate colon cancer. We addressed prospectively significance serum (sIL‐6), measured at diagnosis metastasis, 96 unselected...
Abstract Purpose: The prognostic significance of serum interleukin (IL)-8 was evaluated in patients with metastatic breast cancer. predictive value IL-8 for the presence occult tumor cells bone marrow aspirates operable and Experimental Design: Serum measured healthy controls, cancer, untreated, progressive In 69 either or advanced cytokeratin-positive were counted aspirates. Results: levels are increased 67% (52 77) Overall, these significantly higher cancer compared volunteers (P <...
Although angiogenesis is a prerequisite for the growth of most human solid tumours, alternative mechanisms vascularisation can be adopted. We have previously described non-angiogenic pattern in liver metastases colorectal adenocarcinomas (CRC) which tumour cells replace hepatocytes at tumour–liver interface, preserving architecture and co-opting sinusoidal blood vessels. The aim this study was to determine whether replacement occurs during metastasis breast (BC) lack an angiogenic switch...
Plasma levels of D-dimer are elevated in cancer patients. Activation the extrinsic coagulation system and fibrinolytic cascade within a tumour is thought to be related with growth, invasion metastasis. We have investigated relationship between these markers fibrin metabolism, standard clinicopathological variables serum angiogenic cytokines three cohorts: group A (n=30) consisted 30 healthy female volunteers, B (n=23) consecutive patients operable breast C (n=84) untreated or progressive...
We have compared the platelet number and serum concentration of vascular endothelial growth factor (VEGF), basic fibroblast (bFGF) interleukin-6 (IL-6) in 80 blood samples 50 patients with advanced cancer. also measured mitogenic effect patient sera on cells vitro order to estimate biological activity VEGF. Serum VEGF correlated (r = 0.61; P < 10–4). IL-6 levels count 0.36; 10–3), 0.55; 10–4) calculated load per 0.4; 3 × Patients thrombocytosis had a median which was 3.2 times higher (P...
1012 Background: Anti-PD(L)1 can result in durable responses patients with metastatic triple negative breast cancer (TNBC). However, only a subgroup of TNBC benefits from anti-PD(L)1 response rates 5-10% unselected cohorts. Strategies to render the tumor micro-environment (TME) more susceptible might include stimulation anti-cancer immune by induction treatment irradiation or low dose chemotherapy. Methods: In stage I (non-comparative Simon's two design) who received ≤ 3 lines palliative...
Abstract The prognostic significance of tumor-infiltrating lymphocytes (TILs) in breast cancer has been recognized for over a decade. Although histology-based scoring recommendations exist to standardize visual TILs assessment, interobserver agreement and reproducibility are hampered by heterogeneous infiltration patterns, highlighting the importance computational approaches. Despite advances automate quantification, adoption models hindered lack consensus on methods large-scale benchmarks....
The circulating angiogenic factors vascular endothelial growth factor-A, interleukin-6 and the fibrin D-dimer fragment were measured in mesenteric vein, uterine as well peripheral venous arterial samples 21 randomly selected patients with operable colorectal, ovarian cervical carcinoma. In addition, immunohistochemistry for factor-A was performed on colorectal tumours of such patients. Serum plasma not significantly elevated vein draining tumours, despite tumour cell expression factor-A. is...
About 50% of patients with breast cancer have no involvement axillary lymph nodes at diagnosis and can be considered cured after primary locoregional treatment. However, about 20–30% will experience distant relapse. The group risk is not well characterised: recurrence probably due to the establishment micrometastases before Given early steps metastasis in which tumour cells interact endothelial blood vessels, and, given independent prognostic value both quantification vascularisation...
Background: Higher stromal TILs (sTILs) are associated with higher pathologic complete response (pCR) rates CT in early-stage TNBC. We evaluated the association between sTILs and PD-L1 expression to pembro+CT as NAT for TNBC KEYNOTE-173 (NCT02622074). Methods: were quantified pretreatment (PT) on-treatment (OT; during first 3 weeks of pembro monotherapy). PT was reported combined positive score (CPS). Endpoints pCR rate (ypT0 ypN0; ypT0/Tis ypN0) objective (ORR) after 4 cycles...
574 Background: Higher quantity of pretreatment TIL (PT) is associated with improved pCR and EFS in HER2+ early breast cancer (BC). The value on-treatment unknown. Methods: NeoALTTO trial randomized 455 women BC to 12 weeks NAT trastuzumab, lapatinib or combination paclitaxel, followed by FEC after surgery. In the PAMELA 151 received 18 trastuzumab (±hormonal therapy). were quantified on PT (W2) biopsies using published method H&E slides, tested for associations (logistic regression), OS...
The liver is a highly vascularized organ which frequently hosts metastases in patients with colorectal adenocarcinomas. hypothesis of this study that the hypoxic drive angiogenesis might be minimal or absent those growing are capable preserving stromal structure, including numerous sinusoidal blood vessels. Representative paraffin sections from 26 adenocarcinoma were investigated. Three different growth patterns found. In desmoplastic and pushing (42% 46% all metastases, respectively),...
Abstract Background:Increasing quantities of stromal TILs (sTILs) are associated with higher pathologic complete response (pCR) rates conventional chemo in early-stage TNBC. We evaluated the association between sTILs and PD-L1 expression to pembro+chemo as NAT for TNBC KEYNOTE-173 trial (NCT02622074). Methods: were quantified using light microscopy H&E-stained slides from pretreatment on-treatment (during first 3 weeks pembro monotherapy) tumor biopsies by a pathologist blind data....
A series of 34 patients with pancreatic resections was evaluated respect to the occurrence local and general complications. Two groups were compared, depending on whether or not treatment somatostatin instituted. Postoperative morbidity mortality less frequent when given (resp. 50.0 5.5%) than it omitted 68.7 31.2%). Less complications observed after performed for chronic pancreatitis cancer in non somatostatin-treated group. It is concluded that may be beneficial preventing elective surgery.
