Long-acting (LA) HIV pre-exposure prophylaxis (PrEP) offers the potential to improve adherence by lowering burden of daily or on-demand regimens antiretroviral (ARV) drugs. This paper details fabrication and in vitro performance a subcutaneous trocar-compatible implant for LA delivery tenofovir alafenamide (TAF). The reservoir-style comprises an extruded tube biodegradable polymer, poly(ε-caprolactone) (PCL), filled with formulation TAF castor oil excipient. Parameters that affect release...
Small-sized (∼65 nm) doxorubicin (Dox)-loaded polymeric nanoparticles (PNPs) were modified with oligonucleotides to form colloidally stable Dox-loaded spherical nucleic acid (Dox-PSNA) nanostructures in biological media. The shell facilitates the cellular uptake of Dox-PSNA, which results vitro cytotoxicity against SKOV3 cancer cells.
A critical knowledge gap currently exists regarding the potential risks of exposure to nanoplastics (NPs), particularly early in life during key stages growth and development. Globally abundant plastics, polyamide (nylon) polystyrene (PS), exist various products have been detected food beverages as small-scale plastics. In this study, we evaluated how early-life NPs affects biological metrics rat pups. Male female animals received an oral dose (20 mg/kg/day) nylon-11 (114 ± 2 nm) or PS (85 1...
Fluorescent nanoparticles (NPs) comprising polyethylene terephthalate (PET) with a hydrodynamic diameter of 158 ± 2 nm were synthesized in bottom-up approach. Concentration-dependent uptake and cytotoxicity PET NPs macrophages are shown. The fabrication well-characterized NPs, derived from high-commodity polymers, will support future studies to assess effects on biological systems.
The versatility of copper-catalyzed alkyne–azide coupling (CuAAC) in functionalizing drug-loaded polymer nanoparticles is demonstrated via the modification surfaces acetylene-functionalized PNPs with folate, biotin, and gold nanoparticles.
A critical need exists to develop diverse biomedical strategies for the widespread use of HIV Pre-Exposure Prophylaxis (HIV PrEP). This manuscript describes a subcutaneous reservoir-style implant long-acting delivery tenofovir alafenamide (TAF) PrEP. We detail key parameters TAF formulation that affect performance, including ionization form, selection excipient and exposure aqueous conditions. Both in-vitro studies shelf stability tests demonstrate enhanced performance freebase (TAFFB) in...
A family of nanoparticles has been fabricated featuring cationic amino acid‐based side chains. This controlled surface modification provides a tool to investigate the effect various non‐covalent interactions at nanoparticle–DNA interface. The binding affinities these towards DNA were determined using fluorescence, exhibiting more than threefold modulation in 37‐mer strand. secondary structure strand was distorted upon nanoparticle binding, with extent distortion dependent on acid chain.
Macrophage-mediated inflammation drives autoimmune and chronic inflammatory diseases. Treatment with anti-inflammatory agents can be an effective strategy to reduce this inflammation; however, high concentrations of these have immune-dampening other serious side effects. Synergistic combination mitigate dosing by requiring less drug. Multiple were evaluated in for synergistic inhibition macrophage inflammation. The most potent synergy was observed between dexamethasone (DXM) fumaric acid...
Polymer nanoparticles (PNPs) possessing a high density of drug payload have been successfully stabilized against aggregation in biological buffers after amine modification, which renders these PNPs positively charged. The resulting charge-stabilized retain their original narrow particle size distributions and well-defined spherical morphologies. This stabilization allows to an improved anti-proliferative effect on MDA-MB-231-Br human breast cancer cells compared non-functionalized PNPs. As...
Abstract Objectives To advance the initiative of ending global epidemic, long-lasting HIV protection is needed through sustained release antiretroviral drugs for months to years. We investigated in macaques safety and efficacy biodegradable polycaprolactone implants releasing tenofovir alafenamide pre-exposure prophylaxis (PrEP). Methods Implants were administered subcutaneously arm using a contraceptive trocar. Efficacy against vaginal simian-HIV (SHIV) infection was six pigtailed that...
The administration of antiretrovirals (ARVs) for HIV pre-exposure prophylaxis (PrEP) is highly efficacious and may benefit from new long-acting (LA) drug delivery approaches. This paper describes a subcutaneous, reservoir-style implant the LA tenofovir alafenamide (TAF) documents preclinical assessment safety pharmacokinetics (PK) in New Zealand White (NZW) rabbits (3 groups
Microplastics (MPs) and nanoplastics (NPs) exist in certain environments, beverages, food products. However, the ultimate risk consequences of MPs NPs on human health remain largely unknown. Studies involving biological effects small-scale plastics have predominantly used commercially available polystyrene beads, which cannot represent breadth globally dominant plastics. Nylon is a commodity plastic that across various industry sectors with substantial global production. Here, series...
Controlled drug delivery systems can provide sustained release profiles, favorable pharmacokinetics, and improved patient adherence. Here, a reservoir-style implant comprising biodegradable polymer, poly(ε-caprolactone) (PCL), was developed to deliver drugs subcutaneously. This work addresses key challenge when designing these implantable systems, namely the accurate prediction of profiles using different formulations or form factors implant. The ability model predict behavior from an based...
Long-acting injectable cabotegravir is more effective than daily oral PrEP at preventing HIV transmission due to improved adherence, but requires bi-monthly large-volume intramuscular injections. Subcutaneous (SC) contraceptive implants can be formulated with antiretrovirals for extended-duration PrEP. Islatravir (ISL) a first-in-class, investigational antiretroviral pharmacologic properties well-suited implant delivery. We performed preclinical studies the development of reservoir-style,...
The prevention of HIV and unintended pregnancies is a public health priority. Multi-purpose technologies capable long-acting pregnancy are desirable for women. Here, we utilized preclinical macaque model to evaluate the pharmacokinetics biodegradable ε-polycaprolactone implants delivering antiretroviral islatravir (ISL) contraceptive etonogestrel (ENG). Three were tested: ISL-62 mg, ISL-98 ENG-33 mg. Animals received one or two ISL-eluting implants, with doses 42, 66, 108 µg ISL/day without...
